Abstract:The Mandinga Lagoon in the Mexican State of Veracruz is an important ecological zone that produces 32% of the oyster output in the state of Veracruz, the main oyster producer in Mexico. Samples of water, sediment, and oysters were collected in 2003 and 2004 to study heavy metal pollution. Metal concentrations were determined in water, soil, and oyster tissues from fresh and detoxified Crassostrea virginica, and histology samples were analyzed. Metal (Cr, Cd, and Pb) concentrations in water were within the Mexican legal limits. The recorded values in sediments corresponded to those not producing biological effects (ERL). In the tissues, the highest concentrations corresponded to Pb, above 5.84 µgg -1 dry weight (d.w.); Cd was of 2.23 µgg -1 d.w., and Cr above 6 µgg -1 d.w. The metal levels detected in oysters exceeded the maximum permissible limits (MPL) for Cd and Pb, and oysters were unable to eliminate the concentrations of the bioaccumulated metals during the detoxification stage. The histopathological analysis revealed lesions in the digestive gland, edema, atrophy of epithelia in the digestive tubules, the presence of brown vesicles, hemocytic reaction, and necrosis. During detoxification, a higher number of epithelia were observed in the tubules, as well as an increase in brown vesicles and hemocytic reaction. Forty seven percent of oysters presented histopathological lesions related to metal concentrations. It is important to monitor metal concentrations, to detect the source of pollution, andto evaluate the effects on organisms to establish culture areas and adequate criteria for the exploitation of such an important fishery resource. The treatment of bacterial diseases in aquaculture is done using antibiotics, their applications has resulted in contamination and bacterial resistance. Natural extracts are a potential alternative as an antimicrobial, they have demonstrated effectiveness in their use aimed at treating conditions. The purpose of this study was to evaluate the antimicrobial activity of Lemna minor extracts against Pseudomonas fluorescens with different solvent for extraction. Methanol, chloroform and hexane were used. Subsequently, the safety assessment of the extracts in Danio rerio embryos and larvae was performed to validate as ecologically harmless. Antibacterial activity was detected in three extracts with significant differences (p = 0.001). Hexane extract had the highest antibacterial activity, followed by chloroform and methanol extracts. The three extracts have differences with respect to the control, between times and concentrations tested (p = 0.001). Minimum inhibitory concentration values (MIC) at 24 h methanolic extract ME 0.05 µg mL −1 . In embryos and larvae increased safety of the LC 50 methanolic extract was evidenced followed by the hexane and chloroform extract. No morphological or tissue changes were observed in embryos and larvae. The hexane extracts of L. minor had a greater bactericidal effect against P. fluorescens and are functional because of their antibacterial activity, but methanolic extract is more safety in embryos and larvae of D. rerio , making it a potential alternative for use in the treatment and control of septicemia in fish.

Common carp (Cyprinus carpio) is an aquatic organism of commercial value able to survive in polluted environments; carps contain proteolytic enzymes of physiological importance and potential industrial application. The objective of this work was partially purify and study the proteolytic activity at different pH of carp proteases living in a polluted environment. Three carps were captured in different zones of Zumpango polluted lagoon (Mexico) at 1 m of maximum deep. Protease crude extracts were obtained from dorsal muscle by aqueous extraction and fractionated by 20 %, 50 %, 80 %-saturated (NH4)2SO4. Fractions extracted with 50 % and 80 %-saturated (NH4)2SO4 were selected for their high proteolytic activity and concentrated by ultrafiltration through 100 kDa molecular weight cutoff membranes and analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The crude proteolytic extract had significantly higher activity (19.7 - 20.3 U / mg) at pH 2, 5, and 7 (P < 0.001). Fractions obtained with 20 %, 50 % and 80 % - saturated (NH4)2SO4 showed peak activity at pH 5 (2.8 U / mg) and pH 6 (2.2 U / mg); pH 6 (4.3 U / mg) and pH 3 - 4 (3.6 - 3.7 U / mg); pH 3 (10.8 U / mg) and pH 10 (10.6 U / mg); respectively. Subfractions of < 100 kDa, obtained with 50 % and 80 %-saturated (NH4)2SO4, had peak proteolytic activity at alkaline pH. A < 100 kDa fraction, obtained with 80 %-saturated (NH4)2SO4, had the highest proteolytic activity (37.3 - 43.7 U / mg) at pH 8 - 10, purification factor of 3 and 19.1 % recovery. Thirteen proteins between 9.8 to 104.8 kDa were identified in the crude extract. Peak protein concentration was observed for 31 - 33 and 39 - 41 kDa, suggesting the possibility predominance of serine- and aspartyl- proteases, respectively. We suggest this protease with maximum activity at alkaline pH is related to the adaptation of C. carpio to polluted waters with high pH. Although unsuitable for human consumption, these organisms can be a source of protease production aimed to several uses as in the industry and waste water treatment among others.
