Mesoscale convective systems (MCSs) are important components of tropical weather systems and the climate system. Longterm data of MCS are of great significance in weather and climate research. Using long-term (1985-2008) global satellite infrared (IR) data, we developed a novel objective automatic tracking algorithm, which combines a Kalman filter (KF) with the conventional area-overlapping method, to generate a comprehensive MCS dataset. The new algorithm can effectively track small and fast-moving MCSs and thus obtain more realistic and complete tracking results than previous studies. A few examples are provided to illustrate the potential application of the dataset with a focus on the diurnal variations of MCSs over land and ocean regions. We find that the MCSs occurring over land tend to initiate in the afternoon with greater intensity, but the oceanic MCSs are more likely to initiate in the early morning with weaker intensity. A double peak in the maximum spatial coverage is noted over the western Pacific, especially over the southwestern Pacific during the austral summer. Oceanic MCSs also persist for approximately 1 h longer than their continental counterparts.
Lead (Pb) and cadmium (Cd) are highly toxic and are widespread in agricultural soils, representing risks to plant and human health. In this study, Davidia involucrata was cultivated in soil with different concentrations of Pb and Cd and sampled after 90 days. We used ANOVA to analyse the photosynthesis of D. involucrata and the ability of Pb and Cd to enrich and migrate in roots, stems and leaves. Various results are described here. 1) Under individual and combined Pb and Cd stress, the accumulation factors in the roots were greater than 1, which was significantly greater than those in the stems and leaves (P < 0.05), and the translocation factors both were less than 1. The Pb and Cd enrichment ability of D. involucrata roots was significantly higher than that of stems and leaves, and the migration ability of the two heavy metals in D. involucrata was weak. 2) The Mg-dechelatase activities of chlorophyll degradation products increased under stress due to high concentrations of Pb and Cd. However, chlorophyllase activity was higher at relatively low concentrations of the two heavy metals (P < 0.05). δ-Aminolevulinic acid and porphobilinogen of chlorophyll synthesis products are easily converted to uroporphyrinogen III under low concentrations of Cd, which promotes the synthesis of chlorophyll.3) The effect of Cd stress alone on the chlorophyll concentration was not significant. Under combined stress, concentrations of Pb and Cd in the range of 400~800 mg�kg -1 and 5~20 mg�kg -1 significantly promoted an increase in photosynthetic pigments (P < 0.05). 4) Inhibition of the net photosynthetic rate increased with increasing Pb and Cd concentrations under both individual and combined stress. In addition, the root of D. involucrata had a strong absorption and fixation effect on heavy metals, thereby reducing metal toxicity and improving the tolerance of D. involucrata to heavy metals. , et al. Cell number is a critical factor that determines kernel size in maize (Zea mays). Rapid mitotic divisions in early endosperm development produce most of the cells that make up the starchy endosperm; however, the mechanisms underlying early endosperm development remain largely unknown. We isolated a maize mutant that shows a varied-kernel-size phenotype (vks1). Vks1 encodes ZmKIN11, which belongs to the kinesin-14 subfamily and is predominantly expressed in early endosperm development. VKS1 dynamically localizes to the nucleus and microtubules and plays key roles in the migration of free nuclei in the coenocyte as well as in mitosis and cytokinesis in early mitotic divisions. Absence of VKS1 has relatively minor effects on plants but causes deformities in spindle assembly, sister chromatid separation, and phragmoplast formation in early endosperm development, thereby resulting in reduced cell proliferation. Severities of aberrant mitosis and cytokinesis within individual vks1 endosperms differ, thereby resulting in varied kernel sizes. Our discovery highlights VKS1 as a central regulator of mitosis in early maize endosperm development and provides a potential approach for future yield improvement.
High sensitivity of seeds to water loss is a widespread phenomenon in the world's plant species. The molecular basis of this trait is poorly understood but thought to be associated with critical changes in membrane function. We profiled membrane lipids of seeds in eight species with varying levels of desiccation tolerance and found a close association between reducing seed viability and increasing phosphatidic acid (PA). We applied hydration-dehydration cycles to Arabidopsis seeds, which are normally desiccation tolerant, to mimic the onset of desiccation sensitivity with progression towards germination and examined the role of phospholipase D (PLD) in desiccation stressinduced production of PA. We found that PLDα1 became more abundant and migrated from the cytosol to the membrane during desiccation, whereas PLDδ did not change, and that all desiccation-induced PA was derived from PLDα1 hydrolysis. When PLDα1 was suppressed, the germination level after each hydration-dehydration cycle improved significantly. We further demonstrated that PLDα1-mediated PA formation modulates desiccation sensitivity as applying its inhibitor improved seed desiccation tolerance and its suppression in protoplasts enhanced survival under dehydration. The insights provided by comparative lipidomics enable us to propose a new membrane-based model for seed desiccation stress and survival.
Identification, protection and enhancement of essential habitats are priority issues for management and restoration of exploited species. The shores utilized by Asian horseshoe crabs as nurseries were surveyed and the coastal habitat characteristics were described in the northern Beibu Gulf of China. Regression models were applied to explore species‐habitat relationships. Fourteen and ten nursery sites were identified for Tachypleus tridentatus and Carcinoscorpius rotundicauda populations, respectively. Xiacun and Jinhaiwan in the eastern region of the northern Beibu Gulf were the essential nurseries for T. tridentatus, whereas Shanxin and Jiaodong in the western part were the primary nursery shores for C. rotundicauda. These shores supported high densities (4–6 individuals/100 m2) of juvenile horseshoe crab populations. Mangrove and seagrass coverage area, coupled with sediment physico‐chemical parameters, particularly grain size, and the environmental heterogeneity of nursery habitats explained the distribution pattern of juvenile populations. Most juvenile populations were found along the outer fringe of mangroves in the small shallow estuary, particularly near outflows of tidal creeks with generally higher chlorophyll a and organic carbon contents. The distribution of high‐density juvenile populations of both species also overlapped with areas of seagrass patches. These findings highlight the importance of mangroves and seagrasses in the nursery habitat use of Asian horseshoe crabs. Preserving the estuarine habitats with these vegetation types and identifying the high‐use nursery sites should be prioritised in China and other Asian places to conserve the declining Asian horseshoe crab populations.
BackgroundMany Poaceae species show a gametophytic self-incompatibility (GSI) system, which is controlled by at least two independent and multiallelic loci, S and Z. Until currently, the gene products for S and Z were unknown. Grass SI plant stigmas discriminate between pollen grains that land on its surface and support compatible pollen tube growth and penetration into the stigma, whereas recognizing incompatible pollen and thus inhibiting pollination behaviors. Leymus chinensis (Trin.) Tzvel. (sheepgrass) is a Poaceae SI species. A comprehensive analysis of sheepgrass stigma transcriptome may provide valuable information for understanding the mechanism of pollen-stigma interactions and grass SI.ResultsThe transcript abundance profiles of mature stigmas, mature ovaries and leaves were examined using high-throughput next generation sequencing technology. A comparative transcriptomic analysis of these tissues identified 1,025 specifically or preferentially expressed genes in sheepgrass stigmas. These genes contained a significant proportion of genes predicted to function in cell-cell communication and signal transduction. We identified 111 putative transcription factors (TFs) genes and the most abundant groups were MYB, C2H2, C3H, FAR1, MADS. Comparative analysis of the sheepgrass, rice and Arabidopsis stigma-specific or preferential datasets showed broad similarities and some differences in the proportion of genes in the Gene Ontology (GO) functional categories. Potential SI candidate genes identified in other grasses were also detected in the sheepgrass stigma-specific or preferential dataset. Quantitative real-time PCR experiments validated the expression pattern of stigma preferential genes including homologous grass SI candidate genes.ConclusionsThis study represents the first large-scale investigation of gene expression in the stigmas of an SI grass species. We uncovered many notable genes that are potentially involved in pollen-stigma interactions and SI mechanisms, including genes encoding receptor-like protein kinases (RLK), CBL (calcineurin B-like proteins) interacting protein kinases, calcium-dependent protein kinase, expansins, pectinesterase, peroxidases and various transcription factors.
