Background: COVID-19 is an extremely severe infectious disease. However, few studies have focused on the epidemiological and clinical characteristics of pediatric COVID-19. This study conducted a retrospective review of the epidemiological and clinical features of COVID-19 in children. Methods: A retrospective study was conducted on children with a definite diagnosis of COVID-19 in mainland China using the web crawler technique to collect anonymous COVID-19 updates published by local health authorities. Results: Three hundred forty-one children aged 4 days to 14 years with a median age of 7 years were included. Sixty-six percent of pediatric patients were infected via family members with COVID-19. The median incubation period was 9 days (interquartile range, 6 to 13). Asymptomatic cases accounted for 5.9%, of which 30% had abnormal chest radiologic findings. A majority of pediatric COVID-19 cases showed mild to moderate clinical features, and only a few developed severe or critical diseases (0.6% and 0.3%, respectively). Fever (77.9%) and cough (32.4%) were the predominant presenting symptoms of pediatric COVID-19. The pediatric patients had fewer underlying diseases and complications than adults. The treatment modalities for pediatric COVID-19 patients were not as complex as those of adult COVID-19 patients. The overall prognosis of pediatric COVID-19 was benign with a decent recovery. The median time from onset to cure was 16 days (interquartile range, 13 to 21). Conclusions: Compared to adults, COVID-19 in children has distinct features of epidemiology and clinical manifestations. The findings from this study might help to guide the development of measures to prevent and treat this ongoing global pandemic. Cervical cancer is still one of the leading causes of cancer deaths in women worldwide, especially in the developing countries. It is a major metabolic character of cancer cells to consume large quantities of glucose and derive more energy by glycolysis even in the presence of adequate oxygen, which is called Warburg effect that can be exaggerated by hypoxia. The high risk subtype HPV16 early oncoprotein E6 contributes host cell immortalization and transformation through interacting with a number of cellular factors. Hypoxia-inducible factor 1α (HIF-1α), a ubiquitously expressed transcriptional regulator involved in induction of numerous genes associated with angiogenesis and tumor growth, is highly increased by HPV E6. HIF-1α is a best-known target of the von Hippel-Lindau tumor suppressor (VHL) as an E3 ligase for degradation. In the present work, we found that HPV16 E6 promotes hypoxia induced Warburg effect through hindering the association of HIF-1α and VHL. This disassociation attenuates VHL-mediated HIF-1α ubiquitination and causes HIF-1α accumulation. These results suggest that oncoprotein E6 plays a major role in the regulation of Warburg effect and can be a valuable therapeutic target for HPV-related cancer.
Insecticide resistance can arise from a variety of mechanisms, including changes to the target site, but is often associated with substantial fitness costs to insects. Here we describe two resistance-associated target-site mutations that have synergistic and compensatory effects that combine to produce high and persistent levels of resistance to fipronil, an insecticide targeting on γ-aminobytyric acid (GABA) receptors. In Nilaparvata lugens, a major pest of rice crops in many parts of Asia, we have identified a single point mutation (A302S) in the GABA receptor RDL that has been identified previously in other species and which confers low levels of resistance to fipronil (23-fold) in N. lugans. In addition, we have identified a second resistance-associated RDL mutation (R300Q) that, in combination with A302S, is associated with much higher levels of resistance (237-fold). The R300Q mutation has not been detected in the absence of A302S in either laboratory-selected or field populations, presumably due to the high fitness cost associated with this mutation. Significantly, it appears that the A302S mutation is able to compensate for deleterious effects of R300Q mutation on fitness cost. These findings identify a novel resistance mechanism and may have important implications for the spread of insecticide resistance.
The NE striking Longmen Shan Thrust Belt (LSTB) forms the northeastern margin of Tibetan Plateau and merges northward with the southwestern Qinling orogenic belt. The LSTB is characterized by the Late Cenozoic SE‐ward overthrusting and a current 60‐ to 70‐km crustal thickness. Structural constraints link the Late Cenozoic deformation and crustal thickening of the LSTB to the formation of the Tibetan plateau. However, although older Mesozoic deformations are widely identified, including Triassic shortening, they remain underappreciated in the development of the LSTB as the Cenozoic history has been the recent research focus. Thus, it is important to document the early Mesozoic tectonics of the LSTB to better understand the Cenozoic orogeny along the northeastern margin of the Tibetan plateau. Here we synthesize existing and new mapping, structural analyses and geochronology of three regions of the northern LSTB: the Bikou, Tangwangzhai, and Longwangmiao regions. Our observations suggest that the Bikou, Tangwangzhai, and Longwangmiao thrust complexes have similar kinematic styles of southward foreland fold‐and‐thrust propagation. New 40Ar/39Ar dating results show that these thrust complexes have deformational ages at circa 237 Ma to circa 180 Ma, respectively, and diachronously emplaced before 201–174 Ma. Triassic shortening structures involving metamorphic rocks are unconformably overlain by Lower to Middle Jurassic strata. We propose a new kinematic model in which Middle to Late Triassic, top‐to‐the south, in‐sequence imbricate thrusts developed in the northern and central LSTB, suggesting that the LSTB was initially built in the early Mesozoic by the southward propagation of the foreland belt of the SW Qinling orogenic belt.
BACKGROUND: As the largest transporter gene family in metazoans, ATP-binding cassette (ABC) transporters regulate the efflux of a broad spectrum of substrates from the cytoplasm to the outside of the cell. In arthropods, ABC transporters are involved in phase III of the detoxification process, and play important roles in the metabolism and transport of insecticides. RESULTS: We identified 54 ABC transporters from the genome and transcriptome of Chilo suppressalis, one of the most damaging pests of rice in China. The identified ABC transporters were classified into eight subfamilies (ABCA to ABCH) based on NCBI BLAST and phylogenetic analysis. Synergism studies showed that treatment with verapamil, a potent inhibitor of ABC transporters, resulted in significantly increased toxicity of chlorantraniliprole against C. suppressalis larvae. Among the 21 tested ABC genes, three ABC transporter genes including CsABCC8, CsABCG1C and CsABCH1 were significantly upregulated after chlorantraniliprole treatment. CONCLUSION: ABC transporters play important roles in the detoxification and transport of chlorantraniliprole in C. suppressalis. The results from our study provide valuable information on C. suppressalis ABC transporters, and are helpful in understanding the roles of ABC transporters in chlorantraniliprole resistance mechanisms in C. suppressalis and other insect pests.
To elucidate current domestic factors influencing pharmacogenomics (PGx) implementation and its future in China, we conducted a questionnaire survey on PGx applications and testing. A questionnaire-based survey was created on the popular online professional survey platform “Wenjuanxing” (www.wjx.cn) and performed via the social media platform WeChat. Among 422 participants, there were physicians (27.7%), pharmacists (31.3%), and researchers (41.0%). We found that less than 50% of physicians were aware of the importance of PGx in drug therapy, while over 50% of pharmacists and researchers recognized the importance. Only 38.5% of physicians, 40.9% of pharmacists, and 55.5% of researchers concurred that PGx analysis could lower the economic burdens for patients. However, most of the responders affirmed that PGx should be effectively implemented in clinical practices. A lack of sector standards, a lack of clinical research, and a lack of guidelines were found to be the major factors for hindering PGx clinical application. Among drugs associated with PGx assays, the most common were warfarin and clopidogrel. Although PGx research has advanced rapidly in recent years in mainland China, the clinical implementation of PGx has a long way to go.
