L~il]t man n~m-]ich den racemisehen Ester unvo]lsti~ndig, d, h. nicht bis zu 100% yon dem Ferment in Mandels~ure und den zugehSrigen Alkohol spa]ten, isoliert man die vom Ferment in Freiheit gesetzte S~ure und untersucht nun diese im Polarisationsaploarat, so ist sie rechtsdrehend. Die Schweineleberesterase hat also bei ihrer Einwirknng auf den dl-Ester einer Modi-fik~tion und zwar der d-Form den Vorzug gegeben.Sp/iter wurde gefunden 2, dab eine groi~e Reihe yon Esterasen anderer Herkunft ebenfMls eine solehe Konfigurationsspezifits bei Anwendung yon Substraten mit asymmetrisehem C-Atom zeigen.Fiir die Menschenleberesterase wurden speziell folgende Ergebnisse erhalten ~. Wird racemischer Mandels~ure-Methyl-oder ~thylester in niedriger I~onzentration durch dieses Ferment gespMten, so ist die Menschenleberesterase ,,linksorientiert", d.h. die vom Ferment in Freiheir gesetzte Mandels~ure ist linksdrehend, wird die Anfangskonzen-tr~tion des racemischen Substrats erhSht, so nimmt die Drehung der in Freiheit gesetzten Si~ure ab. Bei noeh hSherer Konzentration wird schliel~lieh die Menschenleberesterase ,,rechtsorientiert", d. h. der d-Ester wird im Racemat bevorzugt hydrolysiert.
