Acetyleholin ist ein Ester, der bekanntlich sehr leicht hydrolysiert und dessen Verseifbarkeit durch katalytisehe Vorg/~nge besehleunigt werden kann. Die vorliegende Arbeit besch~ftigt sieh mit der Spaltung des Acetylcholins durch Blur und Blutbestandteile, die sehon yon Plattner 1 und Mitarbeitern und Loewi 2 und Mitarbeitern auf katalytische Prozesse zuriiekgeffihrt wurde. Es wurden Serum, ungerinnbar gemachtes Gesamtblur und lysiertes Blut von Mensch, Pferd, Rind, Hund und Kaninehen miteinander in bezug auf ihre zerstSrende Eigenschaft ffir Acetyleholin verglichen. Besonders interessierte uns in dieser Arbeit das Verh~ltnis der spaltenden Wirkung des Serums zu der des Gesamtblutes und des H~molysates bei Mensch und Tieren und das Verhalten des acetylcholin-zerst6renden Fermentes bei demselben Individuum im Verlauf von Wochen und innerhalb eines Tages. W/~hrend Galehr und Plattner a sich den Mechanismus der Acetyleholinspaltung durch Blut mit Hilfe oberfl/ichenka~alytischer Prozesse erkl/iren, sueht Loewi a zu beweisen, dab ein Ferment im Blur und Herzextrakt 2 vorhanden sein mu$, welches die Spaltung yon Aeetyleholin beschleunigt. Aueh Dale 5 spraeh friiher die Vermutung aus, dal] die Spaltung yon Acetylcholin dureh Blur auf fermentativem Wege vor sieh gehen mfisse. Bei der Versuehsanordnung dieser Arbeit sind die Blutkonzentrationen so klein gew/s dab die Oberfl/iehenwirkung woM kaum eine Rolle spie]en konnte, wie aueh noch bewiesen werden wird, denn im Gegensatz zu Plattner, der eine 25 % ige Blutkonzentra?Gion gebrauchte, wandten wir 0,5--1,125 % ige Blutans/itze an, so dal~ wit in erster Linie Aussagen fiber die fermentative Acetylcholinspaltung gemaeht haben. Ffir den Nachweis der Acetyleholinspaltung wurde die gasanalytische, manometrische Methode yon Ammon 6 angewandt.Es wurde auSerdem die schon yon Loewi 7 und Mitarbeitel~ festgestellte hemmende Eigensehaft des Physostigmins auf die acetylcholin-

Die proteolytische Aktivität des Papains, des eingetrockneten Saftes der unreifen Frucht des tropischen Melonenbaums Carica papaya, gegenüber Eiweißstoffen und Peptiden ist schon lange bekannt und eingehend untersucht worden 1 . Es gelang, zwei kristalline proteolytische Enzyme aus frischem Latex und getrocknetem Papain zu isolieren, das Papain (im engeren Sinn) 2 und das Chymopapain 3 . Die Untersuchungen auf andere Fermentaktivitäten im Papain ergaben folgendes: Die blutgerinnungsbeschleunigende Wirkung des Papains beruht nach Dyckerhoff und G ig ante auf einer Beimengung von pflanzlichem Thrombin, sog. Phytothrombin 4 . Weiterhin wiesen Meyer und Mitarbeiter 5 im rohen Papainpulver das Vorkommen eines Lysozyms mit mucolytischer und bakteriolytischer Aktivität nach. Das Lysozym konnte von Smith und Mitarbeitern 6 aus getrocknetem Papain als Hg-Derivat kristallisiert erhalten werden. Eine deutlich feststellbare Amylase-Aktivität wurde nur in frischem PapayaLatex beobachtet. Über die esterspaltenden Fermente im Papainpulver weiß man sehr wenig. Kilmer 7 berichtete zum erstenmal von einer schwachen Lipasewirkung des Papaya-Saftes. Sandberg und Brand 8 stellten ein wasserunlösliches Lipasepräparat mit erhöhter Aktivität aus dem Rohpapain her. Eine hydrolytische Wirkung des Wasserextraktes aus handelsüblichem Papain auf Triacetin bemerkte Falk im Rahmen einer Untersuchung über die Einwirkung von Proteasen auf die Lipase-1 "
