Introdução: A anemia falciforme (AF) é uma doença genética de grande prevalência mundial; causada pela presença de uma forma anômala de hemoglobina, a Hemoglobina S (HbS). Objetivo: caracterizar o perfil clínico dos pacientes portadores de anemia falciforme acompanhados em uma unidade de assistência farmacêutica na cidade de Teresina – Piauí, que fazem uso da hidroxiuréia (HU). Resultados: Notou-se uma queda significativa dos valores nominais e percentuais comparados ao controle, nos valores de reticulócitos, leucócitos totais e plaquetas desde o 1º trimestre analisado, e queda no 5º trimestre de Hemácias e do hematócrito. Conclusão: o uso de HU em pacientes portadores de AF mostrou toxicidade, com variações em função do tempo.
Background:
The main protease of SARS-CoV-2 (Mpro) is one of the targets identified in SARS-CoV-2, the causative agent of COVID-19. The application of X-ray diffraction crystallography made available the three-dimensional structure of this protein target in complex with ligands, which paved the way for docking studies.
Objective:
Our goal here is to review recent efforts in the application of docking simulations to identify inhibitors of the Mpro using the program AutoDock4.
Method:
We searched PubMed to identify studies that applied AutoDock4 for docking against this protein target. We used the structures available for Mpro to analyze intermolecular interactions and reviewed the methods used to search for inhibitors.
Results:
The application of docking against the structures available for the Mpro found ligands with an estimated inhibition in the nanomolar range. Such computational approaches focused on the crystal structures revealed potential inhibitors of Mpro that might exhibit pharmacological activity against SARS-CoV-2. Nevertheless, most of these studies lack the proper validation of the docking protocol. Also, they all ignored the potential use of machine learning to predict affinity.
Conclusion:
The combination of structural data with computational approaches opened the possibility to accelerate the search for drugs to treat COVID-19. Several studies used AutoDock4 to search for inhibitors of Mpro. Most of them did not employ a validated docking protocol, which lends support to critics of their computational methodology. Furthermore, one of these studies reported the binding of chloroquine and hydroxychloroquine to Mpro. This study ignores the scientific evidence against the use of these antimalarial drugs to treat COVID-19.
Os portadores de HIV iniciam seus tratamentos no do Sistema Único de Saúde (SUS) através da Assistência da Estratégia da Saúde da Família (ESF) e esta, por sua vez, coordena todos os cuidados prestados e o encaminhamento do paciente a uma unidade de referência para aconselhamento, acompanhamento e controle da infecção de forma integral e resolutiva. Contudo esperamos que este trabalho contribua para compreendermos os tipos de reações adversas ocorridas pelos usuários dos medicamentos para tratamento do HIV. Foram utilizadas todas as publicações que possuírem dados de tratamentos farmacológicos para HIV, dentre a determinada data (2019-2020). Os dados foram coletados utilizando as bases cientificas: Literatura Latino-Americana e do Caribe em Ciências da Saúde (LILACS), National Library of medicine (PUBMED), PERIODICOS, SCIENCE DIRECT, Biblioteca Virtual em saúde – (BVS), Scientific Eletronic Library Online (SCIELO) e Cochrane Library; e as bases tecnológicas: World Intellectual Property Organization (WIPO) e Instituto Nacional de Propriedade Industrial (INPI). Utilizamos como descritores: REAÇÕES ADVERSAS AOS ANTIRRETROVIRAIS, REAÇÕES ADVERSAS AOS ANTIRRETROVIRAIS EM GESTANTES e ANTIRRETROVIRAIS. O idioma de publicação estava em português, inglês e espanhol, atendendo a uma série histórica de pelo menos cinco anos. Foi observado 208.425 artigos nas bases cientificas. Esse tratamento do vírus não cura por conta do acumulo persistente de células infectadas latentemente. Mesmo com sintomas leves, podem levar a uma diminuição da qualidade de vida do portador, assim necessário mais pesquisas sobre o tema. Devemos lembrar que a qualidade de vida deles, impacta diretamente na nossa sociedade e, portanto, importa a todos.
This study reports the action of essential oils (EO) from five plants on the activity of native and recombinant acetylcholinesterases (AChE) from Rhipicephalus microplus. Enzyme activity of native susceptible AChE extract (S.AChE), native resistant AChE extract (R.AChE), and recombinant enzyme (rBmAChE1) was determined. An acetylcholinesterase inhibition test was used to verify the effect of the EO on enzyme activity. EO from Eucalyptus globulus, Citrus aurantifolia, Citrus aurantium var.dulcis inhibited the activity of S.AChE and R.AChE. Oils from the two Citrus species inhibited S.AChE and R.AChE in a similar way while showing greater inhibition on R.AChE. The oil from E. globulus inhibited native AChE, but no difference was observed between the S.AChE and R.AChE; however, 71% inhibition for the rBmAChE1 was recorded. Mentha piperita oil also inhibited S.AChE and R.AChE, but there was significant inhibition at the highest concentration tested. Cymbopogon winterianus oil did not inhibit AChE. Further studies are warranted with the oils from the two Citrus species that inhibited R.AChE because of the problem with R. microplus resistant to organophosphates, which target AChE. C. winterianus oil can be used against R. microplus populations that are resistant to organophosphates because its acaricidal properties act by mechanism(s) other than AChE inhibition.
Os Coronavírus são vírus que causam infecções respiratórias podendo afetar animais e humanos. A epidemia de SARS-CoV-2, causador da COVID-19 teve início em Wuhan (China) em dezembro de 2019, e se espalhou por diversos países tornando-se uma pandemia perigosa e sem tratamento especifico. Esse trabalho pretende mostrar casos tratados com os principais fármacos para o SARS-CoV-2 avaliando sua eficácia e desvantagens. Foram utilizadas publicações com dados de tratamentos farmacológicos para o novo SARS-CoV-2, publicações cientificas, oficiais da Organização Mundial da Saúde (World Health Organization - WHO) e governamentais, dentre a data (2019-2020). Os dados foram coletados nas bases cientificas: LILACS, PUBMED, PERIODICOS, GOOGLE ACADEMICOS, SCIENCE DIRECT, BIRAME, Biblioteca Virtual em saúde - BVS, SCIELO, Cochrane Library, HighWire Press, Scopus e Elsevier; e as bases tecnológicas: WIPO e INPI. A utilização desses fármacos pode ter ocasionado melhora e cura de pacientes globalmente. Acredita-se que são eficazes contra a doença. Porém, temos a necessidade de comprovações cientificas. Nos resultados observamos os fármacos utilizados mundialmente e as patentes aprovadas até (15/09/2020). Não existe tratamento para a Covid19, utilizou-se medicamentos amenizadores dos sintomas gripais da doença. Nos casos emergenciais foram utilizados a combinação do antibiótico azitromicina e hidroxicloroquina, usados para malária e outros medicamentos. Este trabalho avaliou grande parte dos medicamentos que estão em uso para o tratamento do SARS-CoV-2, com o intuito de apresentar um fármaco ou mais que seja eficaz para o tratamento da doença, mostrando ainda se foram realizadas pesquisas in vitro e in vivo na busca de uma melhor tratativa.
