Avian mycoplasmas occur in a variety of bird species. The most important mycoplasmas for chickens and turkeys are Mycoplasma gallisepticum (MG), M. synoviae (MS), and M. meleagridis. Besides, M. iowe (MI) is an emerging pathogen in turkeys, but of little concern for chickens. Mycoplasmas are bacteria that lack cell wall and belong to the class Mollicutes. Although they have been considered extracellular agents, scientists admit nowadays that some of them are obligatory intracellular microorganisms, whereas all other mycoplasmas are considered facultative intracellular organisms. Their pathogenic mechanism for disease include adherence to host target cells, mediation of apoptosis, innocent bystander damage to host cell due to intimate membrane contact, molecular (antigen) mimicry that may lead to tolerance, and mitotic effect for B and/or T lymphocytes, which could lead to suppressed T-cell function and/or production of cytotoxic T cell, besides mycoplasma by-products, such as hydrogen peroxide and superoxide radicals. Moreover, mycoplasma ability to stimulate macrophages, monocytes, T-helper cells and NK cells, results in the production of substances, such as tumor necrosing factor (TNF-α), interleukin (IL-1, 2, 6) and interferon (α, β, γ). The major clinical signs seen in avian mycoplasmosis are coughing, sneezing, snicks, respiratory rales, ocular and nasal discharge, decreased feed intake and egg production, increased mortality, poor hatchability, and, primarily in turkeys, swelling of the infraorbital sinus(es). Nevertheless, chronic and unapparent infections are most common and more threatening. Mycoplasmas are transmitted horizontally, from bird to bird, and vertically, from dam to offspring through the eggs. Losses attributed to mycoplasmosis, mainly MG and MS infections, result from decreased egg production and egg quality, poor hatchability (high rate of embryonic mortality and culling of day-old birds), poor feed efficiency, increase in mortality and carcass condemnations, besides medication costs. Mycoplasmas are diagnosed by serologic tests, culture and PCR and are sensitive to antimicrobials whose action sites are other than the bacterial cell wall, such as tetracyclines, macrolides, quinolones and tiamulin. However, mycoplasma control is more efficiently achieved by acquisition of birds free of MG, MS, MM and/or MI, vaccination of layers, and monitoring of breeder flocks, followed by elimination of the infected flocks that are detected.
RESUMO: A presença de Salmonella spp. em produtos de origem avícola e seus subprodutos se mostra um grande desafio para a produção comercial. Dados de prevalência, dos sorotipos circulantes e do perfil de susceptibilidade antimicrobiana de cepas de Salmonella spp. no Estado do Rio de Janeiro são escassos. Portanto, objetivou-se detectar a presença Salmonella spp. em frangos vivos e carcaças em matadouros do Estados do Rio de Janeiro, identificar os sorotipos e avaliar a susceptibilidade antimicrobiana dessas cepas para fluoroquinolonas e betalactâmicos. Foram coletadas 60 amostras cloacais de frangos vivos e 60 amostras de carcaça de seis matadouros sob Inspeção Estadual (SIE). Os isolados foram sorotipificados e testados frente a oito antimicrobianos: enrofloxacina, ciprofloxacina, norfloxacina, cefalotina, ceftiofur, cefotaxima, amoxicilina/ácido clavulânico e ampicilina pelo método de difusão em disco. Os resultados mostraram uma prevalência de Salmonella spp. de 1,66% (1/60) em amostras de suabe de cloaca e de 26,66% (16/60) em carcaças. Em amostras de suabe de cloaca, somente o sorotipo Senftenberg (1,66%) foi isolado. No total, foram isolados sete sorotipos diferentes nas carcaças: Senftenberg (15%) o mais frequente, seguido por Mbandaka (8,3%), Schwarzengrund (3,3%), Cerro (3,3%), Ohio (3,3%), Minnesota (1,66%) e Tennessee (1,66%). Em relação à susceptibilidade antimicrobiana, 29 (87,87%) isolados foram sensíveis a todos os antimicrobianos testados e 4 (12,12%) isolados foram resistentes a pelo menos três antimicrobianos betalactâmicos ou mais. Não foi observada resistência às fluoroquinolonas. Os resultados encontrados demonstram uma prevalência de Salmonella spp. acima da esperada em matadouros do Estado do Rio de Janeiro, além da presença de vários sorotipos de Salmonella spp. A resistência encontrada para betalactâmicos alerta para a disseminação dessas cepas pela cadeia alimentar.
This study investigated the efficiency of an enrofloxacin ELISA test kit to detect the presence of enrofloxacin residues in broiler tissues compared with LC-MS/MS. Broiler tissues from 72 samples consisting of 60 breast muscle, six pools of livers (500 g each) and six pools of kidneys (500 g each) were obtained from six different slaughterhouses. Breast muscle from 10 carcasses and pools of livers and kidneys from approximately 200 carcasses of the same flock were collected from each slaughterhouse. ELISA and HPLC were used to identify and quantify the contamination of the samples with enrofloxacin. A total of 72% of the analysed samples contained enrofloxacin residues detected by the ELISA and 22.2% were detected by LC-MS/MS. The mean values of enrofloxacin contamination found in chicken breast by ELISA and HPLC were 8.63 and 12.25 μg kg(-1), respectively. None of the samples exceeded the maximum limit of 100 μg kg(-1) by both methods set by the European Union as well as the Brazilian Agriculture Ministry. All positive samples for enrofloxacin residues detected by LC-MS/MS were also positive by ELISA. These data confirm the efficiency of the ELISA test, and suggest its use as a screening method for enrofloxacin residues in poultry tissues due to its quick results, low price and ease of applicability.

Os cuidados com a sanida- The Brazilian poultry industry improves annually and is more representative on production and exportation of their products. Care on poultry health cooperates to these developments, however, respiratory agents that affect weight and carcass quality, continue to threaten poultry production. Airsacculitis is considered the main cause of total and/ or partial condemnation of broilers carcasses, being Mycoplasma gallisepticum (MG) the most important agent of it. This study aimed to detect MG by "Polymerase Chain Reaction" (PCR) and to correlate its positivity with airsacculitis, weight losses and condemnated broilers by Federal Sanitary Inspection. A total of 40 ϐlocks of slaughter broilers under Federal Inspection in Rio Grande do Sul, Brazil, were randomly selected. In each ϐlock three broilers, regardless of sex, were randomly chosen for necropsy where tracheas were collected and polled in one sample for analysis. For PCR, DNA was extracted by the method of phenol-chloroform and ampliϐied with pairs of speciϐic primers for MG. From the 40 ϐlocks PCR analyzed, 20% (8/40) were positive for MG. MG detection was found to be correlected with airsacculitis increase and weight decrease by multiple logistic regression equation (p<0,05), LogitPi= 7.9409 + (0,5601 x X1) -(3.3080 x X2). Airsacculitis rate increase also correleted with decrease in body weight by simple linear regression equation (p<0.05), Y= 2.1050 -0.6397X. In conclusion, MG positivity is related to airsacculitis which causes weight loss in broilers. In addition, the PCR was an effective technique for the detection of MG in ϐlocks of broilers, but was not affected by the kind of biological specimens collected, as tracheal scraping or swab. The present report aims at describing the occurrence of Marek's Disease (MD) in a free range poultry farm based on macroscopic and microscopic lesions. For this purpose, seven free-range chickens were evaluated by gross and microscopic examination. Tumor lesions were observed in the liver and ovary, in addition of enlarged kidney and spleen, skin thickening and whitish lesions in streak form in the breast muscle. Fragments of the affected organs were collected and placed in 10% formalin for processing by the usual histopathology techniques, and stained with hematoxylin-eosin. Under microscopic evaluation, we observed pleomorphic populations of neoplastic lymphoreticular cells with pyknotic nuclei in the same organs, all compatible with MD. The macroscopic and microscopic lesions presented in this study were consistent with MD, meaning that the MD virus is present in the rearing environment of the studied chicken farm.
