ResumoEste trabalho teve como objetivo elucidar, por meio da pirólise analítica acoplada à cromatografia gasosa e espectrometria de massa, alterações na composição química da lignina nas bainhas de plantas de arroz das cultivares BR-Irga 409 e Labelle supridas ou não com silício (Si) e infectadas por Rhizoctonia solani. A concentração de Si nas bainhas das plantas supridas com esse elemento foi significativamente maior (2,7 dag kg -1 ) em comparação com as plantas não supridas (0,45 dag kg -1 ). Na presença de Si, a área abaixo da curva do progresso da queima das bainhas foi significativamente reduzida em 19 e 25%, respectivamente, para as plantas das cultivares BR-Irga-409 e Labelle em relação à ausência desse elemento na solução nutritiva. Com base nos espectros de massas obtidos, foram identificados 33 compostos, dos quais 10 foram produtos da degradação de carboidratos e 23 derivados da lignina. Dentre os derivados da lignina, oito compostos eram do tipo p-hidroxifenila, 11 compostos do tipo guaiacila e quatro compostos do tipo siringila. Nas bainhas das plantas das duas cultivares de arroz, supridas ou não com Si, a concentração de lignina (p-hidroxifenila, siringila (S) e guaiacila (G)) foi de, aproximadamente, 15%. Houve aumento na relação S/G apenas nas bainhas das plantas da cultivar BR-Irga 409 supridas com Si e infectadas por R. solani. A maior concentração de Si nas bainhas das plantas de arroz das duas cultivares, que por sua vez resultou em aumento na relação S/G, contribuiu para reduzir os sintomas da queima das bainhas.Palavras-chave: queima das bainhas, lignina, metabólitos secundários, resistência. Silicon altering compounds derived from the pyrolyses of leaf sheaths of rice plants infected with Rhizoctonia solani AbstractThis study elucidated, through analytical pyrolysis coupled to gas chromatography and mass spectrometry, changes in the chemical composition of lignin on leaf sheaths of rice plants of cultivars BR-Irga 409 and Labelle supplied or non-supplied with silicon (Si) and infected with Rhizoctonia solani. The Si concentration on leaf sheaths of plants supplied with this element was significatively higher (2.7 dag kg ). A ferrugem da soja, causada por Phakopsora pachyrhizi, tem sido controlada pela aplicação de fungicidas devido a indisponibilidade de cultivares resistentes. O silício (Si) tem aumentado a resistência de várias espécies de plantas a patógenos. Assim, este trabalho avaliou o efeito da aplicação foliar desse elemento na severidade da ferrugem e na potencialização da atividade de quitinases, β-1,3glucanases, peroxidases, polifenoloxidases, lipoxigenases e fenilalanina amônia-liases. Plantas de soja (cultivar MG/BR-46 Conquista) foram pulverizadas com água (controle), silicato de potássio (KSi) (pH 10,5), KSi (pH 5,5) e acibenzolar-S-metílico (ASM) 24 horas antes da inoculação. Não houve diferença significativa entre os tratamentos para os teores foliares de Si e potássio. A severidade foi significativamente menor em plantas pulverizadas com KSi, independente do pH, e ASM em relação ao controle. O ASM reduziu significativamente a severidade em 65,5% em relação ao controle sem diferença significativa do ASM para a aplicação de KSi pH 5,5. Aplicação de KSi, independente do pH, também reduziu a severidade. Não houve potencialização da atividade das enzimas estudadas pela aplicação de KSi, independente do pH, e do ASM, embora houve redução da severidade em relação ao controle, o que poderia ser explicado pelo possível efeito desses produtos sobre o fungo. Este trabalho avaliou o efeito do silício (Si) em alguns componentes de resistência do cafeeiro à ferrugem e o possível aumento na atividade de quitinases (QUI), β-1,3-glucanases (GLU) e peroxidases (POX). Mudas de cafeeiro (cv. Catuaí vermelho 44) foram crescidas em solução nutritiva de Clarck modificada, aerada, contendo 0 (-Si) ou 2 mmol L -1 Si (+Si) por 45 dias e inoculadas com H. vastatrix. Não houve diferença significativa entre os tratamentos -Si e +Si para o teor foliar de Si, mas o teor desse elemento nas raízes foi significativamente maior para o tratamento +Si. Os tratamentos -Si e +Si não foram estatisticamente diferentes para o período de incubação, período latente, número de pústulas por folha, área pustular, expansão de pústulas e área abaixo da curva do progresso da ferrugem. A atividade de QUI, GLU e POX aumentou após a inoculação com H. vastatrix, embora com aumentos e decréscimos durante as épocas de coletas. O Si não teve efeito pronunciado em maximizar a atividade dessas enzimas, salvo incrementos em épocas de coletas específicas. ABSTRACT Inefficiency of silicon in leaf rust control on coffee grown in nutrient solutionCoffee leaf rust, caused by Hemileia vastatrix, is the most important disease on coffee. This study aimed to evaluate the effect of silicon (Si) on some components of coffee resistance to rust and to determine if it could increase the activity of chitinases (CHI), β-1,3-glucanases (GLU), and peroxidases (POX). Coffee plants (cultivar "Catuaí vermelho 44") were grown on aerated Clarck's nutrient solution containing 0 (-Si) or 2 mmol L -1 Si (+Si) for 45 days and inoculated with H. vastatrix. There was no significant difference between the -Si and +Si treatments for Si content in leaf tissue, but Si content in root tissue was significantly higher for the +Si treatment. The -Si and +Si treatments were not significantly different for incubation period, latent period, number of pustules per leaf, pustule size, pustule expansion, and area under rust progress curve. The activity of CHI, GLU, and POX increased after inoculation with H. vastatrix, even with increase or decrease in activities during the sampling times. There was not a pronounced effect of Si in increasing the activity of these enzymes, except for rare increases in specific sampling times. Results from this study indicated that coffee plants were inefficient in uptaking Si from the nutrient solution and translocating it to the shoot. Mudas de café (cultivar Catuaí Vermelho 44) foram pulverizadas com água destilada, solução de silicato de potássio (KSi) (35 g/L, pH 10,5), KSi (35 g/L, pH 5,5) e solução de acibenzolar-S-metil (ASM) (200 µg/L) 24 horas antes da inoculação. Não houve diferença significativa entre os tratamentos para os teores foliares de Si e potássio. ABSTRACT Effect of foliar silicon application on resistance against coffee leaf rust and on the potentiation of defense enzymes in coffeeThe major method for control of coffee leaf rust, caused by Hemileia vastatrix, is the use of fungicides. The supply of silicon (Si) to plants is an interesting control strategy because this element can increase the resistance of many plant species to pathogens. This study aimed to evaluate the effect of foliar application of Si on reducing rust severity as well as to determine if it can potentiate the activity of enzymes related to host resistance to pathogens. Coffee plants (cultivar Catuaí Vermelho 44) were sprayed with distilled water, potassium silicate (KSi) solution (35 g/L, pH 10.5), KSi (35 g/L, pH 5.5), and acibenzolar-S-methyl (ASM) solution (200 µg/L) 24 hours before inoculation. There was no significant difference among the treatments regarding the content of Si and potassium in the leaf tissue. Coffee leaf rust, caused by Hemileia vastatrix, is the most devastating disease of coffee. Since limited information is available in the literature on silicon (Si) affecting plant diseases in coffee, this study was designed to investigate foliar application of potassium silicate (PS), a source of soluble (Si), on infection process of coffee leaf rust at the microscopic level. The foliar Si concentration for plants sprayed with water and PS has no significant difference (0.24 and 0.30 dag kg −1 , respectively). X-ray microanalysis indicated that the deposition of Si on the leaves of the plants that were sprayed with PS was greater in comparison to the leaf samples from the plants sprayed with water. Rust severity on leaves of plants sprayed with water or sprayed with PS reached 44% and 32%, respectively, at 36 days after inoculation (dai). Plates of polymerised PS were observed on the leaf surfaces of the plants sprayed with the product, in contrast to its absence on the leaf surfaces of plants sprayed with water. At 36 dai, a greater number of uredia were observed on the leaf surfaces of plants sprayed with water in comparison to the leaf surfaces of plants sprayed with PS. On fractured leaf tissues that were sprayed with PS, less fungal colonisation was observed in comparison to the leaves of plants sprayed with water. In conclusion, the results of this study suggest that the effect of foliar-applied Si on the control of the coffee leaf rust development may be attributed to the physical role of the polymerised PS, its osmotic effect against urediniospores germination, or both.
RESUMONo primeiro experimento, plantas de cafeeiro com três pares de folhas foram pulverizadas com silicato de potássio (K 2 SiO 3 ), epoxiconazole, acibenzolar-S-metil (ASM) e água destilada de duas maneiras: pulverização do 3º par de folhas, a partir do ápice, protegendo o 2º par de folhas ou pulverização de um par de folhas da lateral esquerda da planta e protegendo o par de folhas da lateral direita. Após 24 h, a face abaxial dos pares de folhas protegidos foi inoculada com Hemileia vastatrix. No segundo experimento, o 3º par de folhas de plantas, a partir do ápice, foi pulverizado com K 2 SiO 3 , ASM e água destilada protegendo o 2º par de folhas. Aos 1, 5, 15, 25 e 35 dias após aplicação dos produtos, a face abaxial do 2º (proteção sistêmica) e do 3º par de folhas (proteção local) de oito plantas de cada tratamento foram inoculadas com H. vastatrix. O K 2 SiO 3 pulverizado no 3º par de folhas ou em um par de folhas da lateral esquerda foi ineficiente em aumentar a concentração de silício e reduzir a intensidade da esporulação (IE), o número total de pústulas (NTP) por folha e a severidade da ferrugem no 2º par de folhas ou no par de folhas da lateral direita, ao contrário do epoxiconazole e do ASM, os quais apresentaram sistemicidade na planta. A pulverização do K 2 SiO 3 no 3º par de folhas também não garantiu, devido a sua natureza não sistêmica, redução da IE, do NTP e da severidade da ferrugem comparado com a proteção local. Os resultados desse trabalho apontam para a possibilidade de se utilizar a pulverização com silicato de potássio para reduzir a intensidade da ferrugem do cafeeiro preventivamente. ABSTRACT Local protection, not systemic, of potassium silicate to decrease coffee leaf rust symptomsIn the first experiment, coffee plants with three pairs of leaves were sprayed with potassium silicate (K 2 SiO 3 ), epoxiconazole, acibenzolar-S-methyl (ASM), and distilled water in two different ways: spraying the 3 rd pair of leaves from the apex and protecting the 2 nd pair of leaves or spraying the two pair of leaves on the left side of the plant and protecting the pair of leaves on the right side. Pestalotia leaf spot, caused by the fungus Pestalotiopsis longisetula Guba, has become the major disease affecting strawberry production in Brazil. Strawberry seedlings with 4-5 leaves were inoculated with a conidial suspension of P. longisetula (2 9 10 5 conidia/ml), and leaf samples were collected at 48, 72, 96 and 144 h after inoculation (hai) for observation in the scanning electron microscope. Conidia germinated within 48 hai. At 72 hai, conidia had formed very long germ tubes over the epidermal cells without any evidence of appressorial formation nor direct penetration. At 96 hai, fungal hyphae grew inter-and intracellularly in the lacunous parenchyma and also through tracheary elements. Pycnidia were first observed on the leaf surface at 96 hai. At 144 hai, conidia of P. longisetula were first liberated from the pycnidia. This study adds new information to better understand of the infection process of P. longisetula that may help in developing more effective disease control strategies.
