Detection of Fungi in Acca Sellowiana (Berg) Burret Seeds ABSTRACTAcca sllowiana (Berg) Burret is a native fruit species in southern Brazil, which is attracting great interest due to the high economic potential of its fruit organoleptic potential; the main form of propagation of the species is sexual way. Aiming to determine the sanitary quality of seeds, three forms of detection of fungi were compared with and without surface sterilization. The seeds were derived from four counties in the state of Rio Grande do Sul and three counties in the state of Santa Catarina. The culture media of Potato Dextrose Agar (PDA), V8 (tomato juice) and the method of "Blotter Test" were tested. The following fungi were identified: Aspergillus niger, Aspergillus flavus, Penicillium sp., Colletotrichum gloeosporioides, Fusarium sp., Curvularia sp., Alternaria sp. Trichoderma sp. Epicoccum spp. and Phomopsis sp. The agarized media are more sensitive in the detection of fungi in Acca sellowiana seeds. The sterilization of seeds reduces the incidence of weed fungi.
Goiaba-serrana, a fruit species native to the southern region of Brazil, is promising in ecological and commercial terms. The species propagates naturally through sexual (seeds); however, it may be produced by cutting, grafting, and micropropagation. Several fungal genera can be associated with seeds and be transmitted to seedlings. Thus, this study aimed to identify fungal genera and to analyze different substrates for the germination of Acca sellowiana, associated with seeds from a different origin. To evaluate the microorganisms associated with the seeds, the method of incubation on a paper substrate (“blotter test”) was used. The germination test was carried out in a completely randomized design, in a factorial scheme, (2 x 4), with 4 repetitions of 25 seeds per substrate and lot. The factors corresponded to two origin (Serafina Corrêa and Vacaria, state of Rio Grande do Sul) and four substrates (sand, paper roll, on paper and vermiculite). The fungal genera associated with goiaba-serrana seeds were: Penicillium spp., Aspergillus spp. and Trichoderma spp. The sanity test showed the origin influences the incidence of fungi in the seeds and physiological quality. Moreover, the origin of Serafina Corrêa presented better physiological quality in relation to the Vacaria lot. Any of the substrates used can be used for the germination of goiabaserrana seeds.
Brown leaf spot disease, caused by the fungus Ragnhildiana diffusa, is responsible for losses in pecan orchards and quantifi cation is essential in order to conduct epidemiological studies to establish levels of productivity losses, and assess control strategies. Therefore, the aim of this study was to develop and validate a diagrammatic scale for assessing the severity of brown leaf spot in pecan. 240 symptomatic leafl ets were collected from adult trees in Anta Gorda, in the Brazilian state of Rio Grande do Sul, and used to assess the current percentage severity of the disease. The scale was based on the minimum and maximum observed severity of the disease, with six graduations defi ned according to the Weber-Fechner Visual Stimulus Law. The scale therefore consisted of eight levels (1.5, 4.6, 10.6, 17.3, 39.5, 65.1, 77.5, and 98%). The estimates of eight evaluators provided a basis for validating the scale. They made estimates of disease severity on 40 symptomatic digitalized pecan leafl ets fi rst, without using the scale and then referring to it. The scale provided greater precision, accuracy, and reproducibility of estimates and should result in more reliable epidemiological studies.
