Artemisinin is an important new antimalarial agent containing a bridged endoperoxide. The in vitro antimalarial activity of an artemisinin derivative, arteether, is antagonized by two iron chelators, pyridoxal benzoylhydrazone and 1,2-dimethyl-3-hydroxypyrid-4-one. Similarly, the acute toxicity of artemisinin in mice is antagonized by another chelator, deferoxamine-hydroxyethylstarch. A combination of artemisinin and hemin oxidizes erythrocyte membrane thiols in vitro, and this oxidation is also inhibited by an iron chelator. Thus, iron plays a role in the mechanisms of action and toxicity of artemisinin. The combination of artemisinin and hemin also decreases erythrocyte deformability. Iron probably catalyzes the generation of free radicals from artemisinin since ao-tocopherol antagonizes the thiol-oxidizing activity of artemisinin and since a spin-trapped free radical signal can be seen by electron paramagnetic resonance only when artemisinin is incubated in the presence of iron.Malaria remains an important global health problem, affecting an estimated 270 million people per year and killing 1 to 2 million people per year (47). Resistance to currently used antimalarial drugs such as chloroquine, quinine, mefloquine, and Fansidar is spreading rapidly (47). Thus, there is a great need for new antimalarial agents.The antimalarial agent artemisinin (qinghaosu) was isolated in 1972 from Artemisia annua, an ancient Chinese herbal remedy for fever (for a review, see reference 26). Several artemisinin derivatives are currently undergoing phase I and II clinical studies, including artemether, arteether, and artesunate, which are the methyl ether, ethyl ether, and succinate esters, respectively, of dihydroartemisinin. Artemisinin and its derivatives have been widely used for the therapy of malaria in China, Vietnam, Thailand, and Myanmar; over 2 million doses of artemether have been administered in China alone. Artemisinin-type antimalarial agents are particularly useful against chloroquine-resistant Plasmodium falciparum strains and cerebral malaria (47).
RESUMOComo objetivo de avaliar a capacidade antioxidante de 15 hortaliças comercializadas na Cidade do Recife, extratos metanólicos foram testados quanto a atividade antioxidante em sistema modelo β-caroteno/ácido linoléico e a habilidade de seqüestrar o radical estável 1,1-difenil-2-picrilhidrazil (DPPH). Todas as hortaliças investigadas exibiram propriedade antioxidante, entretanto a ação foi diferenciada entre os vegetais. Os extratos metanólicos da couve folha, tomate, batata, couve-flor, repolho verde, espinafre e alface crespa, com percentual de inibição superior a 70%, foram os mais eficazes em seqüestrar o radical livre. Os extratos metanólicos da alface lisa, cebola branca e vagem apresentaram ação moderada (60-70% de inibição), enquanto que a cebola roxa, chuchu, pepino e cenoura exibiram a mais fraca capacidade de seqüestrar o radical DPPH. No sistema modelo β-caroteno/ácido linoléico, os extratos metanólicos do espinafre e couve-folha exibiram a mais elevada atividade antioxidante (superior a 70%). Ação antioxidante moderada (60-70%) foi exibida pelos extratos da alface lisa, cebola branca e couve-flor, enquanto que os do chuchu, cenoura, pepino, tomate e vagem, com atividade inferior a 60%, foram considerados com fraca ação antioxidante. As hortaliças testadas podem ser vistas como fontes dietéticas de antioxidantes que podem trazer benefícios à saúde, portanto o seu consumo deve ser estimulado. This study was carried out to determine the antioxidant capacity of 15 vegetables commonly consumed in Recife -PE, Brazil. Methanol extracts were screened for their antioxidant activity using two tests: DPPH free radical scavenging and β-carotene/linoléico acid assay. All vegetables showed antioxidant properties however the action was differentiated among the kinds of vegetables. The methanol extracts of collard greens, tomatoes, potatoes, cauliflowers, green cabbage, spinach and lettuce "crespa" had the highest percentage in terms of free radical scavenging activities (> 70%). A moderate activity (60-70%) was shown by lettuce "lisa", white onion and green beans while purple onion, chayote, cucumber and carrot had the lowest free radical scavenging activities. In the β-carotene/linoleic acid assay, spinach and collard greens extracts showed strong antioxidant activity (> 70%). The lettuce "lisa", white onion and cauliflower had a moderate action (60-70%) while the chayote, carrot, cucumber, tomato and green bean showed weak antioxidant activity. The results indicate that vegetables which were evaluated may provide a source of dietary antioxidant therefore their consumption must be encouraged. INTRODUÇÃOAs frutas, reconhecidas fontes de vitaminas, minerais e fibras, são alimentos nutricionalmente importantes da dieta. No entanto, nos últimos anos, maior atenção tem sido dada a estes alimentos uma vez que evidências epidemiológicas têm demonstrado que o consumo regular de vegetais está associado à redução da mortalidade e morbidade por algumas doenças crônicas não transmissíveis. O efeito protetor exercido por estes alimentos tem sido atribuído à presença de fitoquímicos com ação antioxidante, dentre os quais se destacam os polifenóis (Wang, Cao, Prior, 1996;Bravo, 1998;Martinez-Valverde, Periago, Ros, 2000;Kaur, Kapoor, 2002).Os polifenóis, produtos secundários do metabolismo vegetal, constituem um amplo e complexo grupo de fitoquímicos, com mais de 8000 estruturas conhecidas (Bravo, 1998; Martinez-Valverde, Periago, Ros, 2000).
Various organic and aqueous extracts of leaves of Indigofera suffruticosa Mill (Fabaceae) obtained by infusion and maceration were screened for their antibacterial and antifungal activities. The extracts were tested against 5 different species of human pathogenic bacteria and 17 fungal strains by the agar-solid diffusion method. Most of the extracts were devoid of antifungal and antibacterial activities, except the aqueous extract of leaves of I. suffruticosa obtained by infusion, which showed strong inhibitory activity against the Gram-positive bacteria Staphylococcus aureus with a minimal inhibitory concentration (MIC) of 5000 µg ml−1. The MIC values to dermatophyte strains were 2500 µg ml−1 against Trichophyton rubrum (LM-09, LM-13) and Microsporum canis. This study suggests that aqueous extracts of leaves of I. suffruticosa obtained by infusion can be used in the treatment of skin diseases caused by dermatophytes.
Em decorrência de uma ampla diversidade genética, a pitanga (Eugenia uniflora L.) apresenta cor que varia do alaranjado até o vermelho escuro quase negro. Em Pernambuco, no entanto, é comum encontrá-la com coloração laranja-avermelhada. Como muitos fitoquímicos presentes em frutos exibem propriedades antioxidantes, com destaque para carotenóides e compostos fenólicos, e frente a escassez de informações sobre seus teores em pitanga, objetivou-se quantificá-los nas seleções cujos frutos apresentam coloração vermelha e roxa. Assim, foram determinados espectrofotometricamente os teores de carotenóides e fenólicos totais nos frutos destas seleções nos estádios maduro e semi-maduro. Na pitanga roxa madura e em sua película também foram determinados os teores de antocianinas, flavonóis e carotenóides totais. Os compostos fenólicos e os carotenóides totais na pitanga roxa madura encontram-se em maiores teores do que na pitanga vermelha no mesmo estádio de maturação. A seleção roxa exibiu quantidade significante de antocianinas, cujo teor foi mais elevado no fruto maduro do que no semi-maduro. Os carotenóides, antocianinas e flavonóis encontram-se mais concentrados na película do que na polpa deste fruto maduro. A presença de antocianinas, flavonóis e carotenóides totais na pitanga roxa fazem deste fruto uma fonte promissora de compostos antioxidantes cujo cultivo deveria ser estimulado.
ABSTRACT-The aimed of this work was to investigate the effects of maturity stages on the physicochemical characteristics of bilimbi (Averrhoa bilimbi L.) which were cultivated in Recife-PE. During one year, the fruits were harvested from five different trees (unidentified variety) in mature and half-mature stages. They were analysed on total soluble solids (TSS), oxalic acid and vitamin C. Ripe fruits had the highest levels of TSS and vitamin C and lowest levels of oxalic acid, independently of weather conditions. The results showed that maturity stage influenced on physicochemical characteristics of bilimbi fruits.Index terms: Averrhoa bilimbi L., physicochemical characteristics, maturity stages. Os frutos foram colhidos de 05 árvores (variedade desconhecida), nos estádios de maturação maduro e semimaduro, durante um ano. Foram realizadas as determinações de sólidos solúveis totais (SST), ácido oxálico e vitamina C. Os frutos maduros apresentaram maiores valores de SST e vitamina C, e menores de ácido oxálico, independentemente das condições climáticas. Os resultados demonstraram que o estádio de maturação influenciou nas características físico-químicas do bilimbi.
