Two mononuclear fluorophore-labeled copper(II) complexes [Cu(nip)(acac)](+)(2) and [Cu(nip)2](2+) (3), where fluorophore is 2-(naphthalen-1-yl)-1H-imidazo[4,5-f][1,10]phenanthroline (nip) (1) and acac is acetylacetone, have been synthesized and characterized by various techniques. The ligand 1 and complex 2 are structurally characterized by single-crystal X-ray diffraction. The coordination geometries around the copper are square planar in solid as well as solution state as evidenced by electron paramagnetic resonance (EPR) spectroscopy. The density functional calculations carried out on 1-3 have shown that electron-rich regions in the highest occupied orbital are localized on the naphthalene and partly on the phenanthroline moiety. Both complexes 2 and 3 in dimethyl sulfoxide (DMSO) exhibit near square planar structure around the metal ion in their ground state. Time-dependent density functional theory (TD-DFT) calculations reveal that Cu(II) ion in complex 2 shows tetrahedral coordination around the metal while 3 retains its square planar geometry in the lowest excited state. The interaction of complexes with calf-thymus DNA (CT DNA) has been explored by using absorption, emission, thermal denaturation, and viscosity studies, and the intercalating mode of DNA binding has been proposed. The complexes cleave DNA oxidatively without any exogenous additives. The protein binding ability has been monitored by quenching of tryptophan emission in the presence of complexes using bovine serum albumin (BSA) as model protein. The compounds showed dynamic quenching behavior. Further, the anticancer activity of the complexes on MCF-7 (human breast cancer), HeLa (human cervical cancer), HL-60 (human promyelocytic leukemia), and MCF-12A (normal epithelial) cell lines has been studied. It has been observed that 3 exhibits higher cytotoxicity than 2, and the cells undergo apoptotic cell death.
Ruthenium mononitro and mononitroso terpyridine complexes incorporating strong π-acidic azopyridine ligands of the types [Ru II (trpy)(L)(NO 2 )]ClO 4 6-10 and [Ru II (trpyThe single crystal X-ray structure of the nitro complex 7 has been determined. In acetonitrile solvent the nitro complexes exhibit strong MLCT transitions near 500 nm and the same MLCT bands are observed to be blue shifted near 370 nm in the case of the nitroso derivatives. The nitrosyl complexes exhibit ν NO in the range 1960-1950 cm Ϫ1 . The nitro (6-10) and nitroso (11-15) complexes exhibit ruthenium()-ruthenium() oxidation in the ranges 1.33-1.47 V and 1.62-1.72 V versus SCE respectively. The nitroso complexes exhibit two successive one-electron reductions near 0.7 and 0.0 V due to reductions of coordinated NO ϩ → NO and NO → NO Ϫ respectively. All the complexes (6-15) systematically display three ligand based (trpy and L) reductions to negative potentials of the SCE. The nitro complexes (6-10) display weak emissions near 700 nm (Φ = 0.83 × 10 Ϫ2 -2.98 × 10 Ϫ2 ). The electrochemically generated 6 ϩ -10 ϩ show LMCT transitions near 490 nm and rhombic EPR spectra in acetonitrile at 77 K. The pseudo first order rate constants (k) and the thermodynamic parameters (∆H ‡ , ∆S ‡ and K) for formation of the nitro complexes, [Ru II (trpy)(L)(H 2 O)] 2ϩ → [Ru II (trpy)(L)(NO 2 )] ϩ , and the conversion [Ru II (trpy)(L)(NO)] 3ϩ → [Ru II (trpy)(L)(NO 2 )] ϩ have been determined.
The symmetrically dinuclear title compounds were isolated as diamagnetic [(bpy)2Ru(mu-H2L)Ru(bpy)2](ClO4)2 (1-(ClO4)2) and as paramagnetic [(acac)2Ru(mu-H2L)Ru(acac)2] (2) complexes (bpy=2,2'-bipyridine; acac- = acetylacetonate = 2,4-pentanedionato; H2L = 2,5-dioxido-1,4-benzoquinonediimine). The crystal structure of 22 H2O reveals an intricate hydrogen-bonding network: Two symmetry-related molecules 2 are closely connected through two NH(H2L2-)O(acac-) interactions, while the oxygen atoms of H2L2- of two such pairs are bridged by an (H2O)8 cluster at half-occupancy. The cluster consists of cyclic (H2O)6 arrangements with the remaining two exo-H2O molecules connecting two opposite sides of the cyclo-(H2O)6 cluster, and oxido oxygen atoms forming hydrogen bonds with the molecules of 2. Weak antiferromagnetic coupling of the two ruthenium(III) centers in 2 was established by using SQUID magnetometry and EPR spectroscopy. Geometry optimization by means of DFT calculations was carried out for 1(2+) and 2 in their singlet and triplet ground states, respectively. The nature of low-energy electronic transitions was explored by using time-dependent DFT methods. Five redox states were reversibly accessible for each of the complexes; all odd-electron intermediates exhibit comproportionation constants K(c)>10(8). UV-visible-NIR spectroelectrochemistry and EPR spectroscopy of the electrogenerated paramagnetic intermediates were used to ascertain the oxidation-state distribution. In general, the complexes 1n+ prefer the ruthenium(II) configuration with electron transfer occurring largely at the bridging ligand (mu-H2Ln-), as evident from radical-type EPR spectra for 13+ and (+. Higher metal oxidation states (iii, iv) appear to be favored by the complexes 2m; intense long-wavelength absorption bands and RuIII-type EPR signals suggest mixed-valent dimetal configurations of the paramagnetic intermediates 2+ and 2-.
This article describes novel conformationally ordered alpha/beta-hybrid peptides consisting of repeating l-proline-anthranilic acid building blocks. These oligomers adopt a compact, right-handed helical architecture determined by the intrinsic conformational preferences of the individual amino acid residues. The striking feature of these oligomers is their ability to display an unusual periodic pseudo beta-turn network of nine-membered hydrogen-bonded rings formed in the forward direction of the sequence by 1-->2 amino acid interactions both in solid-state and in solution. Conformational investigations of several of these oligomers by single-crystal X-ray diffraction, solution-state NMR, and ab initio MO theory suggest that the characteristic steric and dihedral angle restraints exerted by proline are essential for stabilizing the unusual pseudo beta-turn network found in these oligomers. Replacing proline by the conformationally flexible analogue alanine (Ala) or by the conformationally more constrained alpha-amino isobutyric acid (Aib) had an adverse effect on the stabilization of this structural architecture. These findings increase the potential to design novel secondary structure elements profiting from the steric and dihedral angle constraints of the amino acid constituents and help to augment the conformational space available for synthetic oligomer design with diverse backbone structures.
Reaction of 3,6-diaryl-1,2,4,5-tetrazines (aryl = R = phenyl, 2-furyl or 2-thienyl) with 2 equiv of Ru(acac)2(CH3CN)2 results in reductive tetrazine ring opening to yield diruthenium complexes [(acac)2Ru(III)(dih-R(2-))Ru(III)(acac)2] bridged by the new 1,2-diiminohydrazido(2-) (dih-R(2-) = HNC(R)NNC(R)NH(2-)) ligands. rac/meso diastereoisomers could be detected and separated for the compounds with R = phenyl and 2-thienyl, all species are diamagnetic and were characterized by 1H NMR spectroscopy. Crystal structure determination of the meso isomers with R = phenyl and 2-thienyl confirmed the 1,2-diiminohydrazido formulation through long N-N (approximately 1.40 A) and short C=N(H) bonds (approximately 1.31 A), implying two bridged ruthenium(III) centers at about 4.765 A distance with strong antiferromagnetic coupling. The complexes undergo two reversible and well-separated one-electron reduction and oxidation processes, respectively. EPR Spectroscopy of the paramagnetic intermediates with comproportionation constants K(c) > 10(12) and UV-vis-NIR spectroelectrochemistry were used to identify the accessible redox states as [(acac)2Ru(II)(dih-R(2-))Ru(II)(acac)2]2-, [(acac)2Ru(II)(dih-R(*-))Ru(II)(acac)2]-, [(acac)2Ru(III)(dih-R(2-))Ru(III)(acac)2], [(acac)2Ru(III)(dih-R(*-))Ru(III)(acac)2]+, and [(acac)2Ru(III)(dih-R)Ru(III)(acac)2]2+. While the UV-vis-NIR spectroscopic response of [(acac)2Ru(dih-R)Ru(acac)2](0/-/2-) is very similar to that of [(bpy)2Ru(adc-R)Ru(bpy)2](4+/3+/2+), adc-R(2-) = 1,2-diacylhydrazido(2-), the EPR result indicating ligand-centered spin for [(acac)2Ru(II)(dih-R(*-))Ru(II)(acac)2]- despite deceptive NIR absorptions around 1400 nm reveals distinct differences in the electronic structures.
The complex bioactive natural and unnatural benzopyran congeners have been synthesized using one-/two-step approaches in very good yields from the reactions of two different dihydroxyphthalides, natural resorcyclic acid derivative, and trihydroxybenzophenone with citral and/or farnesal, via the phenol-driven intramolecular diastereoselective thermal/base-catalyzed dipolar [2+2] cycloaddition reactions and three different thermal intramolecular cyclization reactions. The effects of the nature and the position of phenolic groups in the starting materials on the course of these cycloaddition reactions have also been described. Depending upon the absence or presence of intramolecular hydrogen bonding of the phenolic group with the carbonyl moiety in the starting materials, these phenol-driven intramolecular thermal/base-catalyzed dipolar [2+2] cycloaddition reactions either furnished the kinetically controlled products or directly formed the thermodynamically controlled rearranged products, respectively.
