Die Urnsetzung von p-Benzochinon (1) mit 13 substituierten @-Arylarninocrotonestern 2 in Propionsaure fuhrte einheitlich zu den 6-Hydroxyindolen 5. Als Begleitprodukte werden isoliert:5-Hydroxy-indol-Derivate 3, 5-Acyloxy-4-hydroxy-indol-Abkommlinge 4 und als neuartige Reaktionsprodukte Arylamino-benzofurane 6, deren Struktur mit chemischen und spektroskopischen Methoden bewiesen wird. Der Reaktionsverlauf bei der 6-Hydroxy-indol-Bildung wird an Hand der Ergebnisse diskutiert.
Synthesis of CHydroxyindolesReaction of p-benzoquinone (1) with 13 substituted f3-arylaminocrotonates 2 in propionic acid yields 6-hydroxy-indoles 5. By-products are: 5-hydroxyindoles 3, 5-acyloxy-4-hydroxyindoles 4 and arylaminobenzofurans 6 as new products. The structures of the furans 6 were clarified by chemical and spectroscopical methods. The reaction path that leads to 6-hydroxyindols is discussed.Bei der Umsetzung von Enaminen 2 mit p-Benzochinon (1) nach dem Nenitzescu-Verfahren bilden sich in Essigsaure je nach den Reaktionsbedingungen die 5-Hydroxy-bzw. 4-Hydroxy-5-acetoxy-indole 3 bzw. 42)3). Die Synthese eines 6-Hydroxyindols 5 in Eisessig war 19714) erstmals zufallig gegluckt. Nach einem erfolglosen Versuch von anderer Seites) konnten in zwei P~blikationen~)') lediglich 4 weitere 6-Hydroxy-indole erhalten werden (Seite 5 16 oben).Da diese Substanzen ebenso wie ihre Umsetzungsprodukte interessante pharmakologische Eigenschaften a u f w e i~e n~)~) , erschien es lohnend, eine generelle Synthesemoglichkeit von 6-Hydroxy-indol-Derivaten zu erarbeiten. Gleichzeitig sollten Hinweise auf den bisher unbekannten Reaktionsverlauf erhalten werden.Da die bisher dargestellten 6-Hydroxy-indole 5 bei der Umsetzung in Essigsaure bei niedrigen Temperaturen erhalten worden waren, wurde nun als Reaktionsmedium Propionsaure gewahlt. Reaction of p-benzoquinone 3 and aminomethylene indanones 2 via easily oxidized benzocarbazoles 4/5 affords the heterocyclic quinones 6. The structure of 4/5 and 6 is proven by derivatization to 7b and 8b. The product 9 obtained by methylation of 6 is hydrogenated to 10 or debenzylated to 11. Ether cleavage yields 12. Reaction of 9 with lithium-methyl or NaBH4 affords 13 and the intermediates 14 and 15. Phenol 6a was alkylated to 16 or aminomethylated to 17 or 18. The indole quinones 17 and 18 show strong cytotoxic activity against colon cells and pulmonary carcinoma cells.
