Bu çalışmada Karadeniz'de Sinop yarımadası civarında yakalanan izmarit balığı (Spicara smaris) etinin amino asit içeriği ve bu balığın boy ağırlık ilişkisi belirlenmiştir. Amino asit içeriğinin belirlenmesinde; bir karışımdaki istenen maddeyi ileri düzeyde ayırıp tanımlayabilir ve ölçebilir özellikteki LC-MS / MS (Sıvı Kromatografi Kütle Spektrometresi) cihazı kullanılmış ve bu yöntemle izmarit balıkları için sonuçlar ilk kez rapor edilmiştir. Bu çalışmada esansiyel (EAA) ve esansiyel olmayan (NEAA) toplam 19 adet amino asit tespit edilmiştir. Esansiyel amino asitler toplam amino asit miktarının %52,44'ünü oluşturmuştur. İzmarit etinde en yüksek oranda tespit edilen EAA miktarları sırasıyla lysin (2,10±0,215 g/100 g), leucine (1,68±0,115 g/100 g), arginine (1,18±0,030 g/100 g) ve valine (1,09±0,070 g/100 g) şeklindedir. İzmarit etinde en yüksek oranda tespit edilen NEAA miktarları ise sırasıyla glutamic asit (2,88±0,010 g/100 g), aspartik asit (2,16±0,280 g/100 g), alanine (1,32±0,000 g/100 g) ve glycine (1,05±0,060 g/100 g) şeklindedir. Araştırmada incelenen balıkların ağırlık-boy ilişkisi W = 0,0044TL 3,3423 , R 2 = 0,990, N = 45, P < 0,05 şeklinde hesaplanmıştır. Ağırlık boy-ilişkisi (WLR) denkleminin korelasyon katsayısı (r) istatistiksel olarak sıfırdan farklı (P < 0,05), b değeri ise istatistiksel olarak b = 3 den (izometrik büyüme) farklı bulunmuştur (Pauly t test P < 0,05). Bu sonuçlar Karadeniz'de uzatma ağlarıyla avlanan izmarit balıklarının AA bakımından zengin ve pozitif allometrik büyüme özelliği gösterdiğini ortaya koymuştur.
The study was achieved between May 2014 and April 2015 at the Karasu Creek located in the province of Sinop. It was conducted to determine anionic detergent pollution and some physicochemical properties (pH, temperature, conductivity, salinity, dissolved oxygen, total hardness, chemical oxygen demand, phosphate PO4-3, total nitrogen). The anionic detergent concentration of the stations was determined on a monthly basis. Seasonally averaged values of the anionic detergent was measured as the highest value in the autumn season. The lowest values of anionic detergent were found in stations in winter and spring. The increase in the concentration of anionic detergent is caused by population growth in residential areas, increased agricultural activities and rains, and that chemicals move to riverbed from terrestrial areas with rain water.
A B S T R A C TLC-MS/MS was used to determine the amino acid composition in muscle, ovary and liver of whiting, Merlangus merlangus euxinus, caught off the coast of Sinop province in the Black Sea. A total of 19 amino acids (AA) were found in the different samples. The essential amino acids (EAA) in the different tissues of whiting were 55.9% in the meat, 54.8% in the ovary and 51.7% in the liver of the total amino acids. The AA contents except for Arg, Glu, Pro and Tau in meat and ovary of whiting were not significantly different (P > 0.05), but the AA contents except for Cys, Tyr, Asp, Orn and Tau of these two tissues were significantly (P < 0.05) higher than the AA contents of the liver. These results showed that whiting ovaries have approximately as much protein as the meat.
Özet Bu çalışmada, Karadeniz'de Sinop kıyılarından avlanan hamsi balığı etinin esansiyel amino asit (EAA) ve esansiyel olmayan amino asit (NEAA) profili, LC-MS/MS cihazıyla ve bu cihaza uygun olarak modifiye edilmiş yöntem ile ilk kez analiz edilmiştir. Sonuçlar daha önce diğer balıklar üzerine yapılmış çalışmalarla kıyaslanarak, hamsi balığı etinin amino asit içeriği ve diğer balıklardan farkı tartışılmıştır. Hamsi etinde toplam 19 adet amino asit tespit edilmiş olup arginine, cystine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, serine, threonine, tyrosine ve valine esansiyel amino asitleri oluşturmuştur. Bu esansiyel amino asitler toplam amino asit (TAA) miktarının %57.6'sını oluşturmuştur. Hamsi etinde en yüksek oranda tespit edilen EAA miktarı sırasıyla lysin (2,74±0,180 mg/100 g), leucine (2.13±0.230 mg/100 g), arginine (1.54±0.025 mg/100 g) ve valine (1.20±0.080 mg/100 g) şeklinde olduğu tespit edilmiştir. Hamsi etinde en yüksek oranda tespit edilen NEAA miktarının ise sırasıyla, glutamic asit (3.68±0.135 mg/100 g), aspartik asit (2.84±0.270 mg/100 g), alanine (1.52±0.005 mg/100 g) ve glycine (1.22±0.095 mg/100 g) şeklinde olduğu tespit edilmiştir. Bu çalışmada elde edilen sonuçlar önceki çalışmalardan elde edilen sonuçlarla kıyaslandığı zaman, hamsi etinin EAA ve NEAA bakımından zengin olduğu, elde edilen farklı AA profili sonuçlarının ise balık türü, mevsim ve uygulanan AA belirleme yöntemine göre farklılıklar gösterebileceği görülmüştür.
Increasing urbanization and socioeconomic activities day by day cause pollution and degradation of natural resources in the universe. The study was carried out to determine the water quality of Karasu Stream, which originates from the Gündüzlü Forests of Küre Mountains in the Black Sea Region and flows from the Akliman coast to the Black Sea, 8 km west of Sinop. Methods and Results: Karasu Stream's temperature, carbon dioxide, pH, alkalinity, dissolved oxygen, conductivity, oxidation-reduction potential, chloride, biological oxygen demand, ammonium nitrogen, hydrogen sulfide, organic matter, total suspended solids parameters, and total coliform and fecal coliform values were investigated. The seasonal condition was revealed by monitoring every month from May 2014 to April 2015. The total coliform and fecal coliform load of Karasu Stream was determined in the lowest winter season (station 2) and the highest summer season (station 1). The analysis of water samples showed that the mean values of BOI5, NH4 +-N, OM, Toplam coliform ve Fekal coliform were statistically significant both between seasons and between stations (P <0.05). Also, while temperature, CO2, pH, DO, alkalinity and H2S values were differed significantly from season to season (P <0.05), there was no difference between stations (P >0.05). Conclusions: When the research results are compared with the Surface Water Quality Regulation, the classes of Total coliform (Class II-III) and Fecal coliform (Class II-III) load of river water are determined. It was determined that the water source's DO value was low and H2S, F. Coliform values were high. This strengthens the likelihood that eutrophication may start or increase in the water supply. Significance and Impact of the Study: In order to ensure the sustainability of water resources, it is necessary to prevent further deterioration of aquatic environments, and ensuring the protection of water resources may be possible by bringing the issue to the fore.
