RésuméL'étude phytochimique des extraits de Bridelia ferruginea a montré que cette plante contient des quinones, des tanins catéchiques et galliques, des alcaloïdes, des stérols, des polyterpènes, des polyphénols, des composés réducteurs, des flavonoïdes et des saponosides.L'extrait aqueux de Bridelia ferruginea provoque entre 10 -5 mg/mL et 10 -1 mg/mL une augmentation des contractions rythmiques de muscle lisse Taenia coli de cobaye. Ces effets inotropes positifs antagonisés par l'Atropine (10 -6 mg/mL) sont totalement abolis en milieu pauvre (0Ca 2+ ) et dépourvus de calcium (0Ca 2+ + EDTA 10 -5 M).Ces résultats suggèrent la présence de substances cholinomimétiques dans l'extrait brut. Ces substances favoriseraient l'entrée de calcium dans la cellule musculaire lisse par stimulation des récepteurs muscariniques. Ces substances cholinomimétiques justifieraient l'usage traditionnel de cette plante comme laxatif ou purgatif. Mots-clés :Bridelia ferruginea, Taenia coli, récepteur muscarinique. AbstractPhytochemical and pharmacological studies of bridelia ferruginea benth (euphorbiaceae) on guinea pig taenia coli motility.The phytochemical study of Bridelia ferruginea extracts revealed that the plant contained quinones, catechic and gallic tannins, alkaloids, sterols, polyterpenes, polyphenols, reducing compounds, flavonoids and saponins. The aqueous extract of Bridelia Afrique SCIENCE 05 (2) (2009) 305 -320 Semi Anthelme NENE BI et al. 306ferruginea, for concentrations ranging from 10 -5 mg/mL to 10 -1 mg/mL, caused an increase of the rhythmical contraction of guinea pig Taenia coli smooth muscle. These positive inotropic effects inhibited by Atropine (10 -6 mg/mL) were completely abolished in poor calcium mediums (0Ca 2+ ; 0Ca 2+ + EDTA 10 -5 M). The results suggested the presence of cholinomimetic substances in the crude extract of Bridelia ferruginea. Therefore, the inward calcium flow into the smooth muscle cell would be due to the stimulation of muscarinic cholinoceptors induced by these substances. In conclusion, the traditional use of this plant as a laxative is justified by the presence of these cholinomimetic substances.
Costus afer, couramment utilisée pour ses vertus médicinales, est une plante de la pharmacopée traditionnelle africaine. Elle intervient dans le traitement de nombreuses maladies telles que le paludisme, la diarrhée et l'hypertension. L'objectif de la présente étude a été d'évaluer les effets de l'extrait aqueux de cette plante sur le système cardiovasculaire et l'activité respiratoire chez les mammifères. L'étude des effets pharmacologiques de l'extrait aqueux total de Costus afer (Caf) sur l'activité cardiaque et la pression artérielle couplée à la respiration a révélé que Caf aux doses comprises entre 2,94.10-3 et 4,71.10-2 g/kg de poids corporel (PC) induit une diminution de l'amplitude des ondes P et QRS ainsi que la fréquence cardiaque du Lapin. Cet extrait aux doses comprises entre 2,94.10-3 et 3,82.10-2 g/kg de PC provoque une hypotension et une baisse de l'amplitude de l'activité respiratoire chez le lapin. En présence de l'atropine (10-6 mg/ml), l'effet inotrope négatif de Caf est aboli. La persistance de l'effet chronotrope négatif en présence de cet antagoniste des récepteurs cholinergiques de type muscarinique, suggère également la présence de substances cardioinhibitrices non cholinergiques. Pour des concentrations comprises entre 10-12 et 10-6 mg/ml, Caf induit sur le coeur isolé de rat des effets inotropes positifs qui seraient dus à la présence de substances cardiotoniques. Caf, aux concentrations supérieures à 10-6 mg/ml, provoque des effets inotrope et chronotrope négatifs sur l'activité contractile du coeur isolé de rat. Les résultats obtenus lors de cette étude suggèrent donc que cet extrait brut contiendrait des principes actifs cholinomimétiques et non cholinomimétiques et des principes actifs adrénomimétiques.
Heliotropium indicum Linn (Boraginaceae) est une plante usuellement employée dans la pharmacopée africaine pour le traitement de la diarrhée et des maladies cardiovasculaires en particulier l'hypertension. Des études précédentes ont mis en évidence cette propriété antihypertensive chez le lapin. L'objectif de cette présente étude a été d'examiner les mécanismes d'action des principes actifs, de l'extrait aqueux de Heliotropium indicum (HEL) sur le coeur isolé de rat et l'aorte isolée de cobaye, à l'origine de son effet antihypertensif. HEL, de 10-25 à 10-15 mg /ml, induit sur le coeur isolé des effets chronotrope et inotrope négatifs, et aux concentrations comprises entre 10-8 à 10-2 mg/ml, entraîne des effets chronotrope et inotrope positifs. L'inhibition de ces effets, par l'atropine à 10-8 mg/ml et par le propranolol à 10-10 mg/ml, suggère la présence concomitante de substances cholinomimétiques et adrénomimétiques de type β dans HEL. Sur l'aorte isolée, ce concentré naturel provoque une vasorelaxation endothélium-dépendante entre 10-6 et 10-1 mg/ml. L'extrait aqueux de Heliotropium indicum contiendrait de façon concomitante des substances cholinomimétiques et adrénomimétiques.
, Tel : (225) 05 068 986 RESUME L'objectif de ce travail est d'évaluer l'effet d'un extrait aqueux de l'écorce de tige de Bridelia ferruginea (SEA) sur le système nerveux central chez le rat. L'extrait aqueux de cette plante est administré aux animaux par la voie intrapéritonéale 30 minutes avant l'évaluation. L'actographe a permis de quantifier l'activité spontanée alors que l'activité locomotrice a été évaluée grâce à la roue d'activité et la traction. SEA, à la dose de 10 mg/kg de poids corporel réduit de manière significative l'activité spontanée de 63,26% (P<0,001) et de 81,52% (P<0,01) chez les rats pour les intervalles de temps respectifs de 5 et 10 minutes. L'activité locomotrice chez ces animaux est réduite de 78,08% (P< 0,0001) 15 minutes après l'enregistrement de cette activité. Cette dose augmente la latence du réflexe de rétablissement chez le rat. Ces résultats suggèrent que SEA possède un effet sédatif. Cet effet serait lié à la présence d'alcaloïde.
