Durian is the most popular seasonal fruit in Indonesia. Only 1/3 of durian is edible, whereas the seeds are thrown away. This waste is potential of value-added such as seed gum. Mayonnaise is o/w emulsion based product use egg yolk as an emulsifier, but in this research, durian seed gum (2,5%, 3%, 3,5%, 4%, and 4,5%) has been used to replace on egg yolk in making vegan mayonnaise. The best formulation of vegan mayonnaise from 4% durian seed gum was compared to commercial and the control as well. This result is as good as mayonnaise with egg yolk, and the commercial mayonnaise.
AbstrakRumput laut dalam beberapa jenis dilaporkan mempunyai aktivitas antidiabet secara in vivo maupun secara in vitro, namun aktivitas antidiabet fraksi-fraksi polisakarida dari rumput laut belum banyak dilaporkan. Gracilaria gigas merupakan jenis rumput laut merah yang dapat menghasilkan ekstrak agar dan memiliki dua fraksi yaitu agarosa dan agaropektin. Tujuan dari penelitian ini adalah untuk mendapatkan ekstrak agar, fraksi agarosa, fraksi agaropektin dari Gracilaria gigas dan untuk menentukan aktivitas penghambatan α-glukosidase dari ekstrak agar dan fraksinya. Ekstraksi agar digunakan larutan etanol, dan untuk fraksinasi agarosa dan agaropektin digunakan larutan dimetil sulfoxide. Hasil penelitian menunjukkan bahwa fraksi agarosa memiliki kandungan sulfat lebih rendah (0,28%) dibandingkan dengan agar (5,91%) dan agaropektin (6,07%). Jenis sampel (agar, agarosa, dan agaropektin) dan dosis sampel berinteraksi dan berpengaruh nyata dalam menghambat aktivitas enzim α-glukosidase. Fraksi agarosa memiliki nilai IC 50 terhadap α-glukosidase terendah (96,86 ± 4,61 ppm), diikuti oleh ekstrak agar (116,63 ± 5,61 ppm), kemudian fraksi agaropektin (158,34 ± 1,77 ppm).Kata kunci: α-glukosidase, ekstrak, fraksi, Gracilaria gigas, IC 50 Abstract Some types of seaweed are reported to have antidiabet activity in vivo or in vitro, but the activity antidiabet fractions of polysaccharides from seaweed has not been widely reported. Gracilaria gigas is one type of red seaweed that can produce agar and it has two factions, namely agarose and agaropectin. The aim of this study was to obtain an agar extract, agarose fraction, agaropektin fraction of Gracilaria gigas and to determine the α-glucosidase inhibitory activity of extracts and the fractions. Extraction of agar that used an ethanol solution, and to fractionate agarose and agaropektin used dimetilsulfoxide solution. The results showed that the fraction of the agarose having lower sulfate content (0.28%) compared with agar (5.91%) and agaropektin (6.07%). Types of samples (agar, agarose, and agaropektin) and sample doses significantly in inhibiting α-glucosidase enzyme activity. Agarose fraction has IC 50 value against α-glucosidase lowest (96.86 ± 4.61 ppm), followed by the extract agar (116.63 ± 5.61 ppm), then the fraction agaropektin (158.34 ± 1.77 ppm).
