Escherichia coli O157:H7 is one of the pathogenic bacteria causing foodborne disease. The use of lytic bacteriophages can be a good solution to overcome the disease. This study is aimed at isolating lytic bacteriophages from environmental sewage with E. coli O157:H7 bacterial cells. The sample used in this study was eight bacteriophages, and the technique used in identifying E. coli O157:H7 carriers of the stx1 and stx2 genes was PCR. The double layer plaque technique was used to classify bacteriophages. Plaque morphology, host specificity, and electron micrograph were used to identify the bacteriophages. The result obtained plaque morphology as a clear zone with the largest diameter size of 3.5 mm. Lytic bacteriophage could infect E. coli O157:H7 at the highest titer of 10 × 10 8 PFU / mL . Bacteriophages have been identified as Siphoviridae and Myoviridae. Phage 3, phage 4, and phage 8 could infect Atypical Diarrheagenic E. coli 1 (aDEC1) due to their host specificity. The Friedman statistical tests indicate that lytic bacteriophage can significantly lyse E. coli O157:H7 ( p = 0.012 ). The lysis of E. coli O157:H7 by phage 1, phage 2, phage 3, and phage 5 bacteriophages was statistically significant, according to Conover’s posthoc test ( p < 0.05 ). The conclusion obtained from this study is that lytic bacteriophages from environmental sewage could lyse E. coli O157:H7. Therefore, it could be an alternative biocontrol agent against E. coli O157:H7 that contaminates food causing foodborne disease.
AbstrakLatar belakang :Prevalensi anemia pada wanita hamil di Indonesia berkisar 20-80%, tetapi pada umumnya banyak penelitian yang menunjukkan lebih besar yaitu 50%. Di wilayah Indonesia bagian barat tergolong tinggi, Aceh 56,6%, Sumatera utara 77,9%, Sumatera Barat 8,9%, Riau 65,6%, Jambi 74,2%, Sumatera Selatan 58,3%, Lampung 60,7%.Tujuan penelitian : Untuk mengetahui faktor-faktor yang berhubungan dengan kejadian anemia pada ibu hamil di wilayah kerja Puskesmas Bandar Jaya Lahat Kabupaten Lahat tahun 2016.Metode : Penelitian ini menggunakan desain analitik, rancangan cross sectional dengan purposive sampling, dalam bentuk univariat dan bivariat (uji chi square). Sampel berjumlah 49responden. Dengan variabel independen adalah usia ibu hamil, paritas, jarak kelahiran, usia kehamilan, dan pengetahuan. Dan variabel dependen adalah kejadian anemia pada ibu hamil.Hasil penelitian: Ada hubungan antara usia ibu hamil (p = 0,000), jarak kelahiran (p = 0,000), usia kehamilan (p = 0,000), dan pengetahuan (p = 0,000), dengan kejadian anemia pada ibu hamil. Dan tidak ada hubungan paritas (p = 0,472) dengan anemia pada ibu hamil.Simpulan : Ada hubungan antara usia ibu hamil (p = 0,000), jarak kelahiran (p = 0,000), usia kehamilan (p = 0,000), dan pengetahuan (p = 0,000), dengan kejadian anemia pada ibu hamil. A good strategy to conquer the Escherichia coli-cause food-borne disease could be bacteriophages. Porins are a type of β-barrel proteins with diffuse channels and OmpA, which has a role in hydrophilic transport, is the most frequent porin in E. coli; it was also chosen as the potential receptor of the phage. And the Rz/Rz1 was engaged in the breakup of the host bacterial external membrane. This study aimed to analyze the amino acid of OmpA and Rz/Rz1 of lytic bacteriophage from Surabaya, Indonesia. This study employed a sample of 8 bacteriophages from the previous study. The OmpA analysis method was mass spectrometry. Rz/Rz1 was analyzed using PCR, DNA sequencing, Expasy Translation, and Expasy ProtParam. The result obtained 10% to 29% sequence coverage of OmpA, carrying the ligand-binding site. The Rz/Rz1 gene shares a high percentage of 97.04% to 98.89% identities with the Siphoviridae isolate ctTwQ4, partial genome, and Myoviridae isolate cthRA4, partial genome. The Mann–Whitney statistical tests indicate the significant differences between Alanine, Aspartate, Glycine, Proline, Serine ( p = 0.011 ), Asparagine, Cysteine ( p = 0.009 ), Isoleucine ( p = 0.043 ), Lysine ( p = 0.034 ), Methionine ( p = 0.001 ), Threonine ( p = 0.018 ), and Tryptophan ( p = 0.007 ) of OmpA and Rz/Rz1. The conclusion obtained from this study is the fact that OmpA acts as Phage 1, Phage 2, Phage 3, Phage 5, and Phage 6 receptors for its peptide composition comprising the ligand binding site, and Rz/Rz1 participates in host bacteria lysis.
ABSTRAK Latar belakang :Stunting adalah suatu masalah gizi yang berdampak buruk terhadap kualitas hidup anak dalam mencapai titik tumbuh kembang yang optional sesuai potensi genetiknya. Indonesia menduduki peringkat ke lima dunia untuk jumlah anak dengan kondisi stunting. Penyuluhan gizi dapat diartikan sebagai suatu pendekatan edukatif untuk menghasilkan perilaku individu atau masyarakat yang diperlukan dalam peningkatan derajat kesehatan dan mempertahankan gizi yang baik, salah satunya mencegah dan menanggulangi kejadian stunting.Tujuan : Mengetahui perbedaan tingkat pengetahuan sebelum dan sesudah penyuluhan.Metodologi penelitian : Penelitian ini melakukan studi analitik observasional dengan rancangan cross sectionaldengan jenis quasi experiment one group pretest-posttest. Penelitian dilakukan selama bulan oktober 2018. Pengambilan sampel sebanyak 60 sampel dengan teknik accidental sampling.Analisis data menggunakan uji paired t-test.Hasil : Rata-rata tingkat pengetahuan wali murid sebelum penyuluhan adalah 40,68. Rata-rata tingkat pengetahuan wali murid setelah penyuluhan adalh 72,70. Terdapat perbedaan merata yang signifikan antara tingkat pengetahuan wali murid tentang kriteria stuntingsebelum dan sesudah dilakukan penyuluhan ( p = 0.032 ).Kesimpulan : Terdapat perbedaan merata yang signifikan antara tingkat pengetahuan sebelum dan sesudah dilakukan penyuluhan.Kata kunci : Penyuluhan, pengetahuan stunting, wali murid, edukasi.Kepustakaan : 32, 2005-2017
Background: Diabetes Mellitus (DM) is a group of metabolic diseases characterized by an abnormality of insulin secretion, insulin work, or both. DM cases in Indonesia as many as 8,5 million cases in 2013. Prevalence based on diagnosis or symptoms according to the health departement of 2,1%.Purpose: To know the correlation of type 2 diabetes mellitus on triglyceride levels and blood pressureMethods: This study used secondary data derived from the medical record status of patients with diabetes mellitus. The method used is analytic observational with cross sectional approach. Sampling technique using purposive sampling. The population of this study is the diabetes mellitus type 2 in 2017 as many as 151. Sample of 109 respondents. Data analyzed by univariate and bivariate using Chi Square analysis.Results: The frequency of respondent characteristic on female gender as much 57,8%, based on age group >45 old counted 58,7%. Blood pressure frequency distribution found hypertension as much 63,3%. High frequency distribution of triglyceride levels is 67,9%. The Chi Square analysis result there is a significant correlation of type 2 diabetes mellitus on triglyceride levels and blood pressure with Odds Ratio 6,865 with p-value of 0,009.
Salmonella enterica merupakan salah satu bakteri patogen penyebab gastroenteritis yang ditransmisikan melalui air dan makanan terkontaminasi yang sering terjadi pada negara berkembang. Beberapa strain Salmonella enterica multi-resisten terhadap berbagai antibiotika. Bakteriofag litik pada famili Siphoviridae dapat menjadi solusi alternatif dalam mengurangi kejadian gastroenteritis oleh Salmonella enterica. Tujuan penelitian ini adalah untuk mengisolasi, mengetahui kemampuan bakteriofag litik Lytic Bacteriophage 1 (LB1) dalam melisis inangnya yaitu Salmonella enterica penyebab gastroenteritis, serta mengetahui karakterisasi bakteriofag litik LB 1 sebagai biokontrol penyakit gastroenteritis. Bakteriofag litik diisolasi dari pembuangan limbah domestik menggunakan teknik double layer plaque. Bakteriofag litik diidentifikasi berdasarkan morfologi plak, struktur litik, inang, aktivitas lisis sel bakteri Salmonella enterica, stabilitas dalam kondisi buffer yang berbeda dan karakterisasi protein. Bakteriofag litik LB1 hanya menginfeksi sel Salmonella enterica. Hasil pemeriksaan dengan menggunakan Transmission Electron Microscope (TEM), bakteriofag litik LB1 termasuk ke dalam famili Siphoviridae. Morfologi kepala hexagonal-icosahedral berdiameter 72.7 nm, dengan ekor non-kontraktil berdiameter 17.3 nm dan panjang 100 nm. Bakteriofag litik LB1 memiliki stabilitas terbaik dalam buffer Ringers suhu 4 oC yang ditunjukkan dengan penurunan plak sebesar 28% setelah 3 minggu penyimpanan. Hasil pengujian efektivitas menunjukkan bahwa bakteriofag litik LB1 dapat mengurangi populasi sel Salmonella sebanyak 67,12% setelah 8 jam inkubasi. Bakteriofag litik LB1 memiliki 8 protein yang berbeda dengan berat molekul yang beragam 11.4 kDa, 19.6 kDa, 23 kDa, 33 kDa, 58.3 kDa, 77 kDa, 94.5 kDa, 133 kDa. Studi ini menunjukkan bahwa bakteriofag litik LB1 yang diisolasi pembuangan limbah domestik dapat secara efektif mengurangi Salmonella enterica dengan cara melisis sel bakteri. Bakteriofag litik LB1 berpeluang dapat digunakan sebagai biokontrol penyakit gastroenteritis yang disebabkan oleh Salmonella enterica. Stabilitas terbaik bakteriofag litik LB1 pada penyimpanan dalam buffer Ringer di suhu dingin (4oC), memiliki karakterisasi famili Siphoviridae, dapat mengurangi Salmonella enterica sebanyak 67.12% setelah 8 jam inkubasi, dan memiliki berat molekul 11.4-133 kDa. Salmonella enterica is one of pathogenic bacteria causing gastroenteritis transmitted by water and food contamination which commonly occur in developing country. Some study reported that Salmonella serovar enterica strains were multi-resistant to various of antibiotics. Lytic bacteriophage in Siphoviridae family offered a good solution to reduce gastroenterytis disease caused by Salmonella enterica. This reseach aim was to isolate, effectivity test of LB 1 and to characterize lytic bacteriophage as biocontrol of gastroenterytis. Methodology and results were LB1 lytic bacteriophage was isolated from domestic waste using double layer plaque technique, was determined by the plaque morphology, the structure, the host range, the activity to lyse bacterial host cells, the stability of phage on different buffer conditions, and the protein characterization. The results showed that LB1 only infects Salmonella enterica. Based on Electron Microscope Observation showed that LB1 is grouped into Siphoviridae. It has hexagonal-icosahedral head with 72.7 nm in diameter and long-non contractile tail with 100 nm in diameter. LB1 had a good storage stability in Ringers buffer at low temperature (40C), with viability of bacteriophage decreased by 28% after 3 weeks of storage. The effectiveness showed that LB1 could reduce Salmonella enterica by 67.12% after 8 hours of incubation. LB 1 has different proteins with molecular weights: 11.4 kDa, 19.6 kDa, 23 kDa, 33 kDa, 58.3 kDa, 77 kDa, 94.5 kDa, and 133 kDa. The conclusion was LB1 was isolated from sewage water were identified to reduces Salmonella enterica effectively with concentration of 8.2x108 CFU/mL. LB 1 can be used as a biocontrol of gastroenterytis caused by Salmonella enterica, LB 1 has the best stability in buffer ringers in cold temperatures (4oC) and proven as Siphoviridae family, reduced Salmonella enterica by 67.12% after 8 hours of incubation, and has protein molecule with molecular weight 11.4 to 133 kDa.
