Acidobacteria have been described as one of the most abundant and ubiquitous bacterial phyla in soil. However, factors contributing to this ecological success are not well elucidated mainly due to difficulties in bacterial isolation. Acidobacteria may be able to survive for long periods in soil due to protection provided by secreted extracellular polymeric substances that include exopolysaccharides (EPSs). Here we present the first study to characterize EPSs derived from two strains of Acidobacteria from subdivision 1 belonging to Granulicella sp. EPS are unique heteropolysaccharides containing mannose, glucose, galactose and xylose as major components, and are modified with carboxyl and methoxyl functional groups that we characterized by Fourier transform infrared (FTIR) spectroscopy. Both EPS compounds we identified can efficiently emulsify various oils (sunflower seed, diesel, and liquid paraffin) and hydrocarbons (toluene and hexane). Moreover, the emulsions are more thermostable over time than those of commercialized xanthan. Acidobacterial EPS can now be explored as a source of biopolymers that may be attractive and valuable for industrial applications due to their natural origin, sustainability, biodegradability and low toxicity.

Rhizobium tropici, a member of the Rhizobiaceae family, has the ability to synthesize and secrete extracellular polysaccharides (EPS). Rhizobial EPS have attracted much attention from the scientific and industrial communities. Rhizobial isolates and R. tropici mutants that produced higher levels of EPS than the wild-type strain SEMIA4080 were used in the present study. The results suggested a heteropolymer structure for these EPS composed by glucose and galactose as prevailing monomer unit. All EPS samples exhibited a typical non-Newtonian and pseudoplastic fluid flow, and the aqueous solutions apparent viscosities increased in a concentration-dependent manner. These results serve as a foundation for further studies aimed at enhancing interest in the application of the MUTZC3, JAB1 and JAB6 strains with high EPS production and viscosity can be exploited for the large-scale commercial production of Rhizobial polysaccharides.

The members of subdivision 1 Acidobacterium were grown at different pH values in a new medium formulation named PSYL 5 includes sucrose, as a carbon source and other compounds (such as KHPO and MgSO7HO). Growth rate was nearly constant at pH 5 and declined at pH 3-4 and 6-7. However, it was found that the effects involving good C/N ratios and pH on the growth of members of subdivision 1 Acidobacterium were significant, and the strongest effect was given by this conditions at pH 5. In additional, incubation results of 48, 72, 96 and 120 h were relatively shorter as compared to other media earlier described for members of subdivision 1 of the phylum Acidobacteria on solid laboratory media.

SUMMARYAuxin-producing bacteria inhabit the roots of orchids and can bring benefits to the host plant. Plants of this family are multiplied by in vitro assimbiotic conditions and little is known about the role of these microorganisms for ex vitro acclimatization. Four auxin-producing rhizobacteria isolated from the specie Cattleya walkeriana were evaluated for their ability to promote survival and growth of in vitro germinated plantlets during ex vitro acclimatization. Partial sequencing of the 16S rRNA genes of bacteria cultures from root velamen of this epiphytic species identified them as Bacillus, Burkholderia, Enterobacter and Curtobacterium. The presence of indole compounds in the filtered supernatants of liquid cultures was quantified by colorimetric assay and confirmed by HPLC. Indole-3-lactic acid (ILA) and indole-3-acetaldehyde (IAAld) were present in high quantities, except in Enterobacter sp. cultures, where in indole-3-acetic acid (IAA) and indole-3-pyruvic acid (IPA) were more prevalent. These rhizobacteria were inoculated into asymbiotically-germinated plantlets of the host orchid, acclimatized in greenhouse for 90 days and assessed for their growth-promoting ability. The lowest ability to promote growth was observed for Burkholderia sp. and Curtobacterium sp., while Bacillus sp. and Enterobacter sp. improved growth in all evaluated characteristics and increased the percentage of plantlet survival. O objetivo deste trabalho foi avaliar o efeito de diferentes fontes de carbono na composição dos exopolissacarídeos produzidos por Rhizobium tropici SEMIA 4077 e SEMIA 4080, e o efeito da inoculação dessas estirpes na nodulação em feijoeiro. As fontes de carbono utilizadas interferiram nas concentrações de galactose e glicose dos exopolissacarídeos, que foram maior na SEMIA 4080 em meio com sacarose. Embora a SEMIA 4080 tenha liberado grande quantidade de exopolissacarídeos na cultura, não houve diferença entre as estirpes quanto ao número de nódulos do feijoeiro, independentemente da fonte de carbono utilizada no preparo do inóculo. No parâmetro matéria seca de nódulos, houve diferença entre as estirpes, independentemente da fonte de carbono utilizada no preparo do inóculo.

Exopolysaccharide (EPS) are produced by a diverse of rhizobia species and has been demonstrated to be a bioemulsifier with potential applications in the degradation of hydrocarbons. In the present study, attempts were made to obtain the new exopolysaccharide production by Rhizobium tropici (SEMIA 4080 and MUTZC3) strains during growth on hydrocarbon substrate. Under the different cultivation conditions, the high molecular weight exopolysaccharides from Rhizobium tropici strains cultivated for 96h mainly consisted of carbohydrates (79-85%) and a low percentage of protein. The EPSC3-D differed from the others, with only 60% of carbohydrate. However, all strains produced polymers with distinct rheology properties, such as viscosity of each EPS sample, suitable for different applications. In addition, RP-HPLC, FTIR and NMR studies revealed EPS produced by rhizobia strains were similar indicating minimal difference between EPS compositions.
