O objetivo deste estudo foi detectar Streptococcus mutans e Streptococcus sobrinus na saliva e biofilme dentáriode famílias que possuem crianças com idade pré-escolar pelas técnicas da PCR e pela fenotipagem. Foi realizadoisolamento de estreptococos do grupo mutans em seis famílias, de amostras de biofilme dentário e saliva, atravésde semeadura em meio de cultura SB20. Essas amostras de Streptococcus mutans e Streptococcus sobrinus foramidentificadas por meio das características das colônias em ágar sangue, esfregaços corados pelo Gram e testesbioquímicos. A reação em cadeia da polimerase (PCR) foi realizada utilizando primers para amplificação do geneda glicosiltransferase. Streptococcus mutans foi detectado no biofilme dentário pela técnica de PCR de vinte(90,9%) membros e não foi encontrado em apenas duas (9,0%) pessoas, que apresentaram Streptococcus sobrinus.Na saliva, pela técnica do PCR foram identificadas vinte (90,9%) pessoas com Streptococcus mutans e três(13,6%) pessoas com Streptococcus sobrinus. Pelos resultados pode-se concluir que o método do PCR (PCRmultiplex)pode ser mais uma ferramenta na identificação dos estreptococos do grupo mutans. Também poderáser utilizado em estudos da transmissão intrafamiliar destes microrganismos. A especificidade da PCR em relaçãoao método de cultivo para amostras de biofilme dentário que continham Streptococcus mutans foi de 66,67% e asensibilidade de 100%, e para amostras de saliva a sensibilidade foi de 95% e a especificidade de 50%. A especificidadepara detecção de Streptococcus sobrinus para amostras de saliva foi de 95% e de biofilme dentário de100%, enquanto que a sensibilidade para amostras de saliva foi 100% e de biofilme dentário foi 50%. Foi verificadotambém que há uma correlação positiva entre o número de Streptococcus do grupo mutans na saliva dasmães e seus respectivos filhos (r=0,7015, p< 0,01).
