Numerous prevalence studies and outbreaks of Vibrio parahaemolyticus infection have been extensively reported in shellfish and crustaceans. Information on the quantitative detection of V. parahaemolyticus in finfish species is limited. In this study, short mackerels (Rastrelliger brachysoma) obtained from different retail marketplaces were monitored with the presence of total and pathogenic strains of V. parahaemolyticus. Out of 130 short mackerel samples, 116 (89.2%) were detected with the presence of total V. parahaemolyticus and microbial loads of total V. parahaemolyticus ranging from <3 to >105 MPN/g. Prevalence of total V. parahaemolyticus was found highest in wet markets (95.2%) followed by minimarkets (89.1%) and hypermarkets (83.3%). Pathogenic V. parahaemolyticus strains (tdh+ and/or trh+) were detected in 16.2% (21 of 130) of short mackerel samples. The density of tdh+ V. parahaemolyticus strains were examined ranging from 3.6 to >105 MPN/g and microbial loads of V. parahaemolyticus strains positive for both tdh and trh were found ranging from 300 to 740 MPN/g. On the other hand, antibiotic susceptibility profiles of V. parahaemolyticus strains isolated from short mackerels were determined through disc diffusion method in this study. Assessment of antimicrobial susceptibility profile of V. parahaemolyticus revealed majority of the isolates were highly susceptible to ampicillin sulbactam, meropenem, ceftazidime, and imipenem, but resistant to penicillin G and ampicillin. Two isolates (2.99%) exhibited the highest multiple antibiotic resistance (MAR) index value of 0.41 which shown resistance to 7 antibiotics. Results of the present study demonstrated that the occurrence of pathogenic V. parahaemolyticus strains in short mackerels and multidrug resistance of V. parahaemolyticus isolates could be a potential public health concerns to the consumer. Furthermore, prevalence data attained from the current study can be further used to develop a microbial risk assessment model to estimate health risks associated with the consumption of short mackerels contaminated with pathogenic V. parahaemolyticus.

Bacteriophages are ubiquitous in our world, mainly in the oceans, soil, the water and food we consume. They can be used efficiently in modern biotechnology, as well as alternatives to antibiotics for many antibiotic resistant bacterial strains. Phages can be used as vehicles for vaccines both DNA and protein, for the detection of pathogenic bacterial strain, as biocontrol agents in agriculture and food industry. This review outlines the properties as well as the influence of different external physical and chemical factors like temperature and acidity on phage persistence. A better understanding of the complex problem of phage sensitivity to external factors may be useful for other researchers working with phages. Furthermore, the applications of bacteriophages were described in this paper as well.

Salmonella is the well-recognized foodborne pathogen leading the most research and surveillance attention especially from government agencies as well as in food industry. In Malaysia, Salmonella is one of the main bacteria which monitored by the National Laboratory Surveillance System. Previously, standard culture methods have always been employed by many laboratories for Salmonella detection in Food Surveillance Programs. However, more advanced detection methods will be needed to improve the sensitivity and specificity of Salmonella identification. In this review, Salmonella detection methods including conventional and recent advances in molecular-based methods will be discussed.

Salmonellae are highly pathogenic foodborne bacteria able to cause infection even at low doses. Infection by Salmonella from contaminated foods leads to gastrointestinal disease known as salmonellosis. Raw beef can be a source of human infection if the meat products are not properly handled, stored or cooked. This study aimed to investigate the prevalence and concentration of Salmonella in the raw beef sold at wet markets and hypermarkets in Serdang, Selangor, Malaysia, using MPN-PCR and MPN-plating on Xylose Lysine Deoxycholate (XLD) medium. In addition, Salmonella isolates recovered from the samples were tested for antibiotics susceptibility using Kirby-Bauer antibiotic susceptibility testing. The incidence of Salmonella in the raw beef samples using plating and PCR methods were 64.63% (53/82) and 17.07% (14/82) respectively. The microbial concentration of Salmonella in raw beef samples ranged between 3-4600 MPN/g by MPNplating and 3-30 MPN/g by MPN-PCR approach. All isolates were found to be susceptible to imipenem, gentamicin, kanamycin, and chloramphenicol but resistant to cephalothin. It can be deduced from the results that raw beef can be a reservoir for Salmonella infection and the use of cephalothin (30 μg) in the treatment of infection due to these strains could be ineffective. Preventive measures such as proper temperature control as well as proper handling of raw beef in the market place are crucial to the minimization of any potential health hazard posed by this foodborne pathogen.
