Espécies reativas de oxigênio são formadas nas células durante as reações metabólicas aeróbicas e quando produzidas em grande quantidade ou não eliminadas de forma eficaz podem prejudicar o funcionamento ou danificar a célula causando sua morte. Para manter a viabilidade celular é indispensável a manutenção do equilíbrio entre a quantidade de espécies reativas de oxigênio e a ação das defesas que a célula dispõe. No aparato de defesa celular estão compostos e enzimas com atividade antioxidante. Entre as enzimas está a catalase, que catalisa a reação que inativa o peróxido de hidrogênio (H2O2) impedindo a formação do radical hidroxila (OH-), uma das espécies reativas que mais danos causa a célula. Compostos bioativos com capacidade de modulação da atividade enzimática podem aumentar a capacidade de defesa celular contra espécies reativas de oxigênio a partir do aumento da atividade de enzimas como a catalase. O objetivo deste trabalho foi avaliar a influência de extratos obtidos de açaí e pitanga, contendo compostos bioativos, sobre a atividade da enzima catalase. Os extratos das duas frutas foram capazes de modular a atividade enzimática, porém só os extratos etéreo e aquoso de pitanga aumentaram essa atividade, sendo a maior proporcionada pelo extrato etéreo. Os resultados indicam a possibilidade de modular a atividade da catalase e, com isso, permitir maior capacidade de defesa celular contra espécies reativas de oxigênio.

Durante esse processo, a ação da CYP pode ativar compostos potencialmente tóxicos prejudicando o funcionamento celular. Melhores resultados de desintoxicação podem ser alcançados diminuindo a ação da CYP e aumentando a ação da GST, responsável pela eliminação da maioria destes compostos. Estudos têm sido realizados no intuito de avaliar efeitos que componentes bioativos de vegetais realizam sobre a atividade das enzimas de biotransformação. Há varias frutas ricas em compostos que, isolados, são reportados como moduladores de atividade enzimática incluindo enzimas biotransformadoras. O objetivo deste trabalho foi avaliar a capacidade de compostos de açaí e pitanga em inibir a atividade de CYP e aumentar a atividade de GST hepática de ratos, buscando otimizar o processo de biotransformação. A atividade de enzimas da família de CYP (PROD, EROD, MROD e ECOD) foi determinada fluorimetricamente utilizando substratos específicos, e a atividade de GST foi realizada por ensaio espectrofotométrico utilizando o substrato CDNB. Foram utilizados nos ensaios extratos de etéreos de açaí e pitanga. Dentre os extratos testados, apenas o extrato etéreo de açaí promoveu modulação parcialmente satisfatória, já que o extrato de pitanga não demostrou influência sobre as enzimas. O extrato de açaí inibiu expressivamente a atividade de CYP, possivelmente diminuindo a ativação de compostos tóxicos, porém também foi inibidor da GST diminuindo a capacidade celular de eliminação destes compostos. ABSTRACT The Citocromo P450 enzymes (CYP) and Glutathione S-transferase (GST) participate in the process of cellular metabolism of xenobiotic compounds making them less toxic and facilitating their elimination (biotransformation). During this process, the action of CYP can activate potentially toxic compounds impairing the cellular function. Better detoxification results can be achieved by decreasing the action of CYP and increasing the action of GST, responsible for the elimination of most of these composts. Studies have been carried out to evaluate the effects of bioactive components of plants on the activity of biotransforming enzymes.
