A series of 3-substituted-7-aminoalcoxy-coumarin was designed and evaluated as cholinesterase inhibitors and antioxidants. All compounds were effective in inhibiting AChE with potencies in the nanomolar range. The 3-(4-(dimethylamino)phenyl)-7-aminoethoxy-coumarin (6a) was considered a hit, showing good AChE inhibition potency (IC50 = 20 nM) and selectivity (IC50 BuChE/AChE = 354), quite similar to the reference drug donepezil (IC50 = 6 nM; IC50 BuChE/AChE = 365), also presenting antioxidant properties, low citotoxicity and good-predicted ADMET properties. The mode of action (mixed-type) and SAR analysis for this series of compounds were described by means of kinetic and molecular modeling evaluations.
Espécies reativas de oxigênio são formadas nas células durante as reações metabólicas aeróbicas e quando produzidas em grande quantidade ou não eliminadas de forma eficaz podem prejudicar o funcionamento ou danificar a célula causando sua morte. Para manter a viabilidade celular é indispensável a manutenção do equilíbrio entre a quantidade de espécies reativas de oxigênio e a ação das defesas que a célula dispõe. No aparato de defesa celular estão compostos e enzimas com atividade antioxidante. Entre as enzimas está a catalase, que catalisa a reação que inativa o peróxido de hidrogênio (H2O2) impedindo a formação do radical hidroxila (OH-), uma das espécies reativas que mais danos causa a célula. Compostos bioativos com capacidade de modulação da atividade enzimática podem aumentar a capacidade de defesa celular contra espécies reativas de oxigênio a partir do aumento da atividade de enzimas como a catalase. O objetivo deste trabalho foi avaliar a influência de extratos obtidos de açaí e pitanga, contendo compostos bioativos, sobre a atividade da enzima catalase. Os extratos das duas frutas foram capazes de modular a atividade enzimática, porém só os extratos etéreo e aquoso de pitanga aumentaram essa atividade, sendo a maior proporcionada pelo extrato etéreo. Os resultados indicam a possibilidade de modular a atividade da catalase e, com isso, permitir maior capacidade de defesa celular contra espécies reativas de oxigênio.
<p><span style="font-size: medium;">Glyphosate, a non-selective herbicide with systemic action, is one of the most widely used substances in modern agriculture. With the objective of comparing the efficiency of glyphosate previously complexed with copper (Cu421) with that of the commercial product (Roundup WG®) and the purified substance (purified glyphosate) in controlling <em>Brachiaria decumbens </em>Stapf (signal grass), tests were conducted of the <em>in vitro </em>and <em>in vivo </em>activity of the enzymes 5-enolpyruvylshikimate-3-phosphate synthase (EPSPs) and phenylalanine ammonia lyase (PAL). The Cu421 complex significantly inhibited the EPSPs activity, as did the commercial product. <em>In vivo </em>test showed that treatments increased PAL activity more than double. The Cu421 complexed caused a substantial decline in the Falker chlorophyll index, as well as, the greatest reduction in the fresh mass yield and total protein concentration. In addition, the Cu 421 complexed application resulted in an increase in N-NO3- content in <em>B. decumbens</em>. The Cu421 complex had similar action as Roundup WG® and the purified glyphosate, indicating the possibility of using this complex to control weeds.</span></p>
Este trabalho pretende contribuir para o estudo de formas alternativas de controle de Rhipicephalus microplus através do fungo Metarhizium anisopliae. Com o objetivo de avaliar o efeito deste fungo sobre a atividade de proteases do carrapato bovino, 80 fêmeas ingurgitadas de R. microplus foram divididas em grupo controle e grupo de tratamento (48h de tratamento). As fêmeas tratadas foram imersas nas suspensões conidiais e os controles em água destilada estéril acrescida de Tween 80 0,1%. Foi feita a dissecção das fêmeas e os intestinos foram lavados, secos em papel de filtro para a retirada do excesso de líquidos e pesados. Foram então homogeneizados e a concentração protéica foi determinada. A presença da atividade proteolítica em extrato intestinal foi dosada pela hidrólise do substrato protéico hemoglobina e pela quantificação da tirosina. Nos valores de pH 4,0 e 5,0 foi encontrado um aumento da atividade frente o uso do fungo. Nestes valores de pH há presença principalmente de cisteíno e aspártico protease. Ao fazer uso de um inibidor de cisteíno protease (ditiotreitol) houve inibição de 74% e 54% da atividade proteolítica nos grupos tratado e controle, respectivamente. Enquanto isso, o outro inibidor de cisteíno proteases, phenylmethanesulfonyl fluoride, inibiu 86% e 80% da atividade proteolítica em grupos tratado e controle, respectivamente. Foi possível concluir com este estudo que há uma atividade majoritária de cisteíno proteases atuando em intestino de R. microplus e que esta atividade pode ser aumentada na presença do fungo entomopatogênico. Este aumento, por sua vez, pode ser decorrente de proteases do fungo ou do mecanismo de defesa do carrapato, o que sugere que M. anisopliae pode vir a ser uma forma de controle eficaz para este carrapato.

Durante esse processo, a ação da CYP pode ativar compostos potencialmente tóxicos prejudicando o funcionamento celular. Melhores resultados de desintoxicação podem ser alcançados diminuindo a ação da CYP e aumentando a ação da GST, responsável pela eliminação da maioria destes compostos. Estudos têm sido realizados no intuito de avaliar efeitos que componentes bioativos de vegetais realizam sobre a atividade das enzimas de biotransformação. Há varias frutas ricas em compostos que, isolados, são reportados como moduladores de atividade enzimática incluindo enzimas biotransformadoras. O objetivo deste trabalho foi avaliar a capacidade de compostos de açaí e pitanga em inibir a atividade de CYP e aumentar a atividade de GST hepática de ratos, buscando otimizar o processo de biotransformação. A atividade de enzimas da família de CYP (PROD, EROD, MROD e ECOD) foi determinada fluorimetricamente utilizando substratos específicos, e a atividade de GST foi realizada por ensaio espectrofotométrico utilizando o substrato CDNB. Foram utilizados nos ensaios extratos de etéreos de açaí e pitanga. Dentre os extratos testados, apenas o extrato etéreo de açaí promoveu modulação parcialmente satisfatória, já que o extrato de pitanga não demostrou influência sobre as enzimas. O extrato de açaí inibiu expressivamente a atividade de CYP, possivelmente diminuindo a ativação de compostos tóxicos, porém também foi inibidor da GST diminuindo a capacidade celular de eliminação destes compostos. ABSTRACT The Citocromo P450 enzymes (CYP) and Glutathione S-transferase (GST) participate in the process of cellular metabolism of xenobiotic compounds making them less toxic and facilitating their elimination (biotransformation). During this process, the action of CYP can activate potentially toxic compounds impairing the cellular function. Better detoxification results can be achieved by decreasing the action of CYP and increasing the action of GST, responsible for the elimination of most of these composts. Studies have been carried out to evaluate the effects of bioactive components of plants on the activity of biotransforming enzymes.
