We studied the leaves of Begonia grandis Dryand. subsp. grandis, the northernmost and most cold-resistant representative of the predominantly tropical genus Begonia, by histochemical methods. In glandular and nonglandular trichomes as well as in the epidermal cells of B. grandis Dryand. subsp. grandis leaves, phenolic compounds, including flavonoids, as well as terpenoids and carbonyl compounds were detected. The patterns of phenolic compounds in the acetone and ethanol leaf exudates and in leaves as a whole were similar and contained oxalic, citric, and gallic acids, isoquercitrin, and orientin. Concentrations of phenolic compounds in the acetone and ethanol exudates constituted 0.10 % and 2.59 % of all phenolic compounds in the leaves, respectively. Antimicrobial effects of the aqueous ethanol extract and of the ethanol exudate against reference strains of Bacillus subtilis, Staphylococcus aureus, and Candida albicans were detected at the disc con tents of 50.0 and 45.8 μg, respectively. The observed set of characteristics can be used in a targeted search for highly antimicrobial species of Begoniaceaе.
Динамика содержания пигментов в листьях Begonia grandis Dryander subsp. grandis при интродукции в Западной Сибири (г. Новосибирск) Впервые изучена динамика содержания пигментов листьев (хлорофиллов a и b, каротиноидов, антоцианов) и показателей физиологического состояния (соотношений хлорофилл a / хлорофилл b, хлорофилл / каротиноиды, хлорофилл / антоцианы) в течение сезона вегетации у растений Begonia grandis subsp. grandis, интродуцированных в Западной Сибири в условиях оранжереи и открытого грунта. В весенний период активного роста выявлены максимальные показатели содержания хлорофилла (8-9 мг/г абсолютно сухой массы), каротиноидов (4-5 мг/г) и антоцианов (18-20 мг/г). Установлено, что характер динамики пигментов с мая по конец сентября аналогичен в оранжерее и в открытом грунте. Для периодов адаптации характерны более низкие величины соотношений хлорофилл / каротиноиды (1,3-4,4) и хлорофилл / антоцианы (0,3-1,0) по сравнению с благоприятными периодами (5,5-6,2 и 2-2,1 соответственно). В условиях воздействия низкой температуры значительно больше антоцианов содержат листья оранжерейных растений, находящихся в более благоприятных условиях и имеющих более высокий физиологический статус. Переход к состоянию покоя B. grandis subsp. grandis при интродукции в оранжерее не сопровождается значительным уменьшением содержания хлорофилла и накоплением антоцианов. The dynamics of the seasonal activity of exometabolites was studied for the lianas from the families: Araceae — Epipremnum aureum G.S. Bunting, Monstera deliciosa Liebm. var. borsigiana h Araliaceae — Hedera helix L. and Schefflera octophylla (Lour.) Harms. The antimicrobial activity of intact plants was evaluated by treating inoculated microbial test cultures Staphylococcus epidermidis, Escherichia coli and Candida albicans. in Petri dished with the volatile emissions of plants. Antimicrobial action was measured in comparison with the reference using the scale of phytoncide activity. The species and seasonal specificity of antimicrobial activity of exometabolites of intact plant leaves was revealed. All the studied plants exhibited moderate seasonal activity against S. epidermidis. Long-term seasonal activity was detected for S. octophylla. Pronounced activity against E. coli was detected in the species H. helix, M. deliciosa var. borsigiana and S. octophylla, while antifungal action was detected in the species E. aureum and S. octophylla. Substantial antifungal and antibacterial activity against S. epidermidis was observed during intense growth during summer and in the autumn-winter period. These experimental data provide evidence of species-specific antimicrobial activity during different periods of seasonal development of plants against the test objects.
; 2 Сибирский государственный университет геосистем и технологий (СГУГИТ), Новосибирск Для подготовки специалистов по направлению 05.03.06 Экология и природопользование, профиль «Природопользование», проведен опыт внедрения в обучающий процесс навыков полевых исследований и обработки полученных данных совместно с научными сотрудниками ЦСБС СО РАН г. Новосибирска. Студенты под руководством ученых провели исследование растительного мира при инженерно-экологических изысканиях на модельном участке площадью 1 га береговой зоны Обского водохранилища в районе города Бердска. Работа выполнена в три этапа: 1) изучение литературных источников, подбор топографических карт и данных дистанционного зондирования Земли; 2) геоботанические описания растительности, выявление флористического состава; 3) создание карты растительности, списка растительных сообществ, разработка рекомендаций по охране редких и охраняемых видов. Студенты во время практики проанализировали взаимосвязь между дисциплинами «Экология», «Биология», «Картография» и «Рациональное природопользование». В ходе практики обучающиеся подготовили аннотированный список флористического состава растений; собрали, определили и подшили гербарий; создали атлас-определитель растительности модельного участка; составили карту растительности. Полученные практические навыки позволят будущим специалистам после завершения обучения в вузе быстро включиться в процесс работы как в научноисследовательской, так и проектно-производственной деятельности. The unique protected ecosystems created by the Central Siberian Botanical Garden (CSBG) researchers on the basis of ecological – geographical principle allowed the establishment of exact soil and microclimate conditions, favorable to the vigorous growth of many terrestrial Begonias. Begonias are successfully introduced in the CSBG as an understorey component of the pluristratal tropical forest and well adapted to the shady, humid conditions. Maintenance of rich Begoniaceae collection was based on the concept of imaging the natural habitat of the Begonias to determine environmental factors such as illuminance, temperature, humidity and substrate preference. In vitro regeneration of 4 Begonia genotypes was carried out starting from female flower segments and peduncles as explants. These regenerants have a better capability to grow than traditionally obtained ones and their flowers developed to anthesis in vitro. The techniques open a new way for an efficient micropropagation protocol for Begonia species conservation in vitro.
The paper continues multi-year integrated studies of the numerous taxons of tropical Begonia genus, which allowed us to evaluate the adaptive capacity of the representatives of this genus relying on the revealed morphological, rhythmological and biochemical features. The investigation of antimicrobial activity of intact plants and different fractions of plant extracts allowed us to distinguish several promising representatives of this genus exhibiting activity against a broad range of test microorganisms. The involvement of these plants within phytomodules in child care centers caused a substantial decrease in total microbial contamination of the air. The paper contains results of the experimental investigation of seasonal antimicrobial activity of 13 species, 8 hybrids. The phytoncide activity was measured by exposing the streak cultures of microbial test species bacteria Staphylococcus epidermidis, Esсherichia coli and yeast-like fungi Candida albicans to the volatile emissions of plants. It has been established that 95% of begonias are distinguished by the pronounced activity against S. epidermidis bacteria, 48% against E. coli bacteria and 43% against fungi Candida albicans. The seasonal specificity of the antimicrobial action of begonias was detected: the growth of the colonies of S. epidermidis bacteria is inhibited to the highest extent during spring and summer, at the phase of intense growth, while the activity against E. coli is the highest during autumn and winter at the phase of moderate growth. The taxons with clearly pronounced, long-term and universal antimicrobial action to the studied test objects were revealed and recommended for practical planting in various types of indoor environments. The data obtained in the study may serve as the basis for further investigation of the chemical composition of volatile exometabolites by the example of the representatives of Gireoudia and Coelocentrum sections that have exhibited high antimicrobial activity against S. epidermidis and E. coli.
The effect of three phyto modules – different combinations of indoor plants with the pronounced phytoncide activity – on the qualitative and quantitative composition of microflora in the air and on a decrease in the risk of acute respiratory diseases (ARD) in children was studied during autumn and winter in 2018-2020 in child care preschool institutions. Sanitary descriptive, epidemiological, sanitary bacteriological and statistical methods were involved. Observation revealed higher attendance rate in the observation group, in combination with lower recorded incidence of disease in comparison with the reference group. The fraction of facultative microflora (FM) with respect to the total microbial count (TMC) was about 30 % in the rooms equipped with phyto modules, while the ratio was 60% in the rooms without plants. The degree of antimicrobial activity depended on the assortment of plants and on the total leaf area per unit room volume. The phytoncide effect of the plants was traced to the most remote point of the room under investigation – 3 m. A favorable factor was an increase in the relative air humidity from 26 to 40%.
Изучение адаптационного потенциала четырёх таксонов из рода Begonia (B. rockii Irmscher, B. sutherlandii J.D. Hooker, B. variegate Y.M. Shui & W.H. Chen, B. 'Gloire de Lorraine') способствовало выделению факторов, влияющих на длительное сохранение коллекции бегоний in vitro и ex vivo в Центральном сибирском ботаническом саду (ЦСБС). Выявлена взаимосвязь экологических условий произрастания данных бегоний в природе с ритмами роста в условиях оранжерей. Составлены феноспектры, отражающие фазы развития растений, что позволяет организовать необходимые агротехнические мероприятия для бегоний в течение года. Проанализирована продуктивность размножения изученных бегоний различными способами. Изучено влияние различных концентраций экзогенных регуляторов роста и состава питательных сред на регенерационную способность флоральных эксплантов 4 таксонов бегоний для моделирования их пролиферации в культуре in vitro. Предложен состав модифицированной среды N 6 , включавшей цитокинины TDZ и БАП в концентрациях 0,5 мг/л и 1,0 мг/л соответственно, что позволило получить путем прямого органогенеза многочисленные регенеранты из фрагментов цветков бегоний, относящихся к разным генотипам. Жизнеспособность микрорастений была сохранена при культивировании в коллекции in vitro благодаря применению приема чередования питательных сред с различным соотношением макроэлементов и регуляторов роста. Подобраны субстраты для выращивания бегоний в условиях оранжерей и для адаптации микроклонов при переводе их в условия ex vitro. Предложенные способы длительного поддержания коллекции представителей рода Begonia предполагают сочетание стандартных методик и микроклонального размножения, при применении которых происходит реювенилизация растительного материала.
