ABSTRAKKepiting bakau Scylla olivacea merupakan salah satu komoditas perikanan yang memiliki nilai ekonomis yang sangat tinggi. Kendala yang dihadapi dalam kegiatan perbenihan kepiting bakau adalah masih tingginya tingkat mortalitas. Tujuan penelitan untuk mengevaluasi pengaruh hormon ecdyson terhadap sintasan dan periode moulting pada larva kepiting bakau Scylla olivacea. PENDAHULUANKepiting bakau Scylla olivacea merupakan salah satu komoditas perikanan yang memiliki nilai ekonomis yang sangat tinggi. Kepiting bakau memiliki nilai gizi yang tinggi dan kualitas cita rasa yang disukai konsumen baik di dalam negeri maupun pasaran internasional. Seiring meningkatnya permintaan konsumen, membawa konsekuensi terhadap tuntutan pengembangannya yakni salah satunya melalui budidaya kepiting bakau secara intensif. Kepiting bakau telah dibudidayakan di beberapa daerah di Indonesia, akan tetapi kendala utama yang dihadapi adalah ketersediaan benih yang terbatas. Salah satu solusi yang dapat dilakukan untuk mengatasi ketersediaan benih guna meningkatkan produksi kepiting bakau adalah melalui kegiatan perbenihan.
<p><em>Brachionus</em><em> plicatilis</em> is a widely used as natural feed in hatchery. Characteristics of rotifers as biocapsule can improve the quality of rotifers in a practical way. <em>Bacillus</em><em> </em>sp. is capable of improving the nutrition rotifers to produce vitamins, food detoxication or through enzymatic activity such as protease, lipase, and amylase. This research aimed at observing the influence of the enrichment length of <em>Bacillus </em>sp. on the quality of nutrition and growth of rotifers. The research container was a <em>polyearbonate</em> vessel of one ton volume which contained <em>Nannochloropsis</em> sp. with the density of 10<sup>5</sup> cell/mL, it was then scattered with rotifer with the density of 1,000 ind./mL. The types of bacteria used were <em>Bacillus subtilis</em>, <em>B. pumilus</em> and <em>B. licheformis</em> with the density of 2x10<sup>10</sup> cfu/g. The experiments tried with different enrichment length were A = 0 hour, B = 5 hours, C = 10 hours and D = 10 hours with triplicate. As the comparing data, rotifer culture experiment was carried out with <em>Nannochlorophsis </em>sp.as the control. The number of containers were 24 which consists of 12 for threatment and 12 pieces as control. Variables measured were proximate to each treatment and the growth of rotifers. The result of the research revealed the enrichment length of rotifers with <em>Bacillus</em> sp. has significantly (P<0,05) towards increased protein, crude fiber, ash, BETN, and growth. Optimum enrichment time was obtained at 5 hours.</p> <p>Key words: rotifers, <em>Bacillus</em>, enrichment period, proximate</p> <p> </p> <p>ABSTRAK</p> <p><em>Brachionus plicatilis</em> merupakan pakan alami yang banyak digunakan dalam pembenihan. Karakteristik rotifer sebagai biokapsul secara praktis dapat meningkatkan kualitas rotifer. <em>Bacillus </em>sp<em>.</em> mampu memperbaiki nutrisi rotifer dengan memproduksi vitamin, detoksikasi pangan maupun melalui aktivitas enzimatis seperti <em>protease</em>, <em>lipase</em>, dan <em>amilase</em>. Penelitian ini bertujuan untuk melihat pengaruh pengkayaan <em>Bacillus </em>sp<em>.</em> terhadap kualitas nutrisi dan pertumbuhan rotifer. Wadah penelitian menggunakan bak fiber volume 1 ton yang berisikan <em>Nannochloropsis</em> sp<em>.</em> dengan kepadatan 10<sup>5</sup> cell/mL, kemudian ditebari rotifer dengan kepadatan 1.000 ind./mL. Jenis bakteri yang digunakan berupa <em>Bacillus</em> <em>subtilis</em>, <em>B. Pumilus </em>dan<em> B. Licheformis </em>dengan kepadatan 2x10<sup>10</sup> cfu/g. Perlakuan yang diujikan adalah lama pengkayaan probiotik <em>Bacillus </em>sp<em>.</em> yang berbeda, yaitu perlakuan A (0 jam), perlakuan B (5 Jam), perlakuan C (10 jam) dan perlakuan D (15 jam). Sebagai data pembanding dilakukan percobaan kultur rotifer dengan <em>Nannochloropsis</em> sp<em>.</em> sebagai kontrol. Jumlah wadah penelitian yang dipergunakan sebanyak 24 buah yang terdiri atas 12 buah untuk perlakuan dan 12 buah sebagai kontrol. Peubah yang diukur adalah proksimat pada masing-masing perlakuan dan pertumbuhan rotifer. Hasil penelitian menunjukkan bahwa perlakuan lama pengkayaan rotifer dengan <em>Bacillus</em> sp<em>.</em> meningkatkan (P<0,05) terhadap kadar protein, serat kasar, abu, BETN, dan pertumbuhan rotifer. Penelitian ini bertujuan untuk menganalisis kandungan vitamin C rotifer, artemia dan larva rajungan setelah diperkaya dengan vitamin C dan menganalisis pemberian vitamin C terhadap sintasan, rasio RNA/DNA, kecepatan metamorfosis dan ketahanan stres larva rajungan stadia zoea. Serta menentukan dosis optimum vitamin C yang menghasilkan sintasan, rasio RNA/DNA, ke-cepatan metamorfosis dan ketahanan stres larva rajungan stadia zoea yang terbaik. Penelitian ini dilaksanakan pada bulan Mei sampai Juni 2020 di unit pembenihan kepiting dan rajungan Balai Perikanan Budidaya Air Payau (BPBAP) Takalar. Analisis Rasio RNA/DNA dilakukan di Laboratorium Uji Fisika Kimia BPBAP Takalar. Rancangan penelitian menggunakan Rancangan Acak Lengkap (RAL). Hasil penelitian menunjukkan bahwa pemberian vitamin C pada rotifer dan artemia dapat meningkatkan kandungan rotifer, artemia dan larva rajungan. Pemberian vitamin C pada rotifer dan artemia terhadap larva rajungan stadia zoea dapat meningkatkan sintasan, kecepatan metamorfosis dan ketahanan stres larva rajungan dan Dosis vitamin C 250 ppm yang terbaik pada pemeliharaan larva rajungan stadia zoea. This study aimed to analyze the content of vitamin C rotifer, artemia and small crab larvae after enriched with vitamin C and to analyze the provision of vitamin C to survival, RNA / DNA ratio, metamorphosis rate and stress resistance of crab larvae in zoea stage. Also, it aimed to determine the optimum dose of vitamin C which produces the best survival rate, RNA/DNA ratio, metamorphosis speed and stress resistance of crab larvae in zoea stage. This research was conducted from May to June 2020 in the hatchery unit of the Brackish Water Cultivation Fishery Center (BPBAP) Takalar. RNA / DNA ratio analysis was carried out at the Laboratory of Chemical Physics Testing of BPBAP Takalar. The research design used a completely randomized design (CRD). The results showed that giving vitamin C to rotifers and brine shrimp increased the content of rotifers, brine shrimp and crab larvae. Giving vitamin C to rotifers and artemia to larvae of zoea stadia crabs can increase survival, metamorphosis speed and stress resistance of crab larvae and the best dose of vitamin C 250 ppm in the maintenance of zoea stadia crab larvae.
Penelitian ini bertujuan untuk menganalisis rasio RNA/DNA, pertumbuhan dan tingkat kelangsungan hidup larva ikan bandeng yang disuplementasi Vitamin C pada rotifer dan artemia dengan dosis yang berbeda, perlakuan ini diharapkan dapat meningkatkan performa fisiologis larva ikan bandeng. Penelitian ini dilakukan pada bulan September sampai Oktober 2021 di unit pembenihan ikan bandeng Balai Perikanan Budidaya Air Payau (BPBAP) Takalar. Analisis laboratorium untuk beberapa parameter uji di laksanakan di Laboratorium Uji Fisika Kimia BPBAP Takalar. Pada penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Hasil Penelitian Menunjukkan bahwa Suplementasi vitamin C pada rotifer dan artemia dapat meningkatkan rasio RNA/DNA, pertumbuhan dan tingkat kelangsungan hidup larva ikan bandeng. Dosis suplementasi rotifer dan artemia dengan vitamin C terbaik untuk rasio RNA/DNA, pertumbuhan dan tingkat kelangsungan hidup yang diperoleh pada penelitian ini adalah pada dosis 200 mg/L dengan tingkat kelangsungan hidup 35,64 ± 0,125c% diikuti dosis 150 mg/L sebesar 34,45 ± 0,49c% dan dosis 100 mg/L 32,17 ± 0,79b%. untuk yang terendah pada perlakuan kontrol 0 mg/L 29,31 ± 1,189a%. The study aims to analyze the RNA/DNA ratio, growth, and survival rates of vitamin C supplemented milkfish larvae in rotifers and artemia at different doses, This treatment is expected to improve the physiological performance of the milkfish larvae. This research was conducted from September to October 2021 at the fish hatchery unit of The Brackish Aquaculture Fisheries Center (BPBAP) Takalar. Laboratory analysis for several test parameters was carried out at the BPBAP Takalar Chemical Physics Test Laboratory. The study used a Complete Randomized Design (RAL). The results showed that vitamin C supplementation in rotifers and artemia could improve RNA/DNA ratio, growth, and survival rate of milkfish larvae. The best dose of rotifer and artemia supplementation with vitamin C for RNA/DNA ratio, growth, and survival rate obtained in this study was at 200 mg/L with a survival rate of 35.64 ± 0.125c% followed by a dose of 150 mg/L of 34.45 ± 0.49c% and a dose of 100 mg/L of 32.17 ± 0.79b%. For the lowest on the control treatment was 0 mg / L 29.31% ± 1,189a.
Penelitian ini bertujuan untuk meningkatkan nilai nutrisi pakan alami rotifer dan artemia melalui penambahan beta karoten yang bersumber dari wortel. Penelitian ini dilaksanakan pada bulan Mei 2020 di unit pembenihan kepiting dan rajungan Balai Pe-rikanan Budidaya Air Payau (BPBAP) Takalar. Rancangan penelitian yang digunakan adalah rancangan acak lengkap (RAL) sebanyak 4 perlakuan dan 3 ulangan. Hewan uji yang digunakan adalah larva rajungan (Portunus Pelagicus) stadia megalopa yang ditebar dengan kepadatan 5 ekor/l dan dipelihara sampai memasuki stadia crab. Luaran penelitian yang ditargetkan adalah teknologi pengayaan pakan alami rotifer dan artemia menggunakan beta karoten yang berasal dari wortel. Hasil penelitian menunjukkan bahwa Pemberian frekwensi pakan yang tepat setelah dikayakan dengan beta caroten akan meningkatkan sintasan dan pertumbuhan pada larva rajungan. Frekwensi pakan yang tepat diberikan pada larva rajungan yang terbaik untuk kelang-sungan hidup larva rajungan adalah pemberian pakan 3 kali/hari. This study aims to increase the nutritional value of natural rotifer and artemia feed by adding beta carotene from carrots. This research was conducted in May 2020 at the hatchery unit of the Brackish Water Cultivation Fishery Center (BPBAP) Takalar. The research design used was a completely randomized design (CRD) with 4 treatments and 3 replications. The experimental animals used were small crab larvae (Portunus pelagicus) in the megalopa stage which were stocked with a density of 5 individuals/l and they were kept until entering the crab stage. The target output of this research is natural feed enrichment technology for rotifer and artemia using beta carotene derived from carrots. The results showed that giving the right frequency of feed after being enriched with beta carotene would increase survival and growth in small crab larvae. The best frequency of feed given to small crab larvae for the survival of small crab larvae is feeding 3 times/day.
