Penelitian ini bertujuan mengetahui profil biokimia darah pada sapi perah yang mengalami kawin berulang. Sapi yang digunakan dalam penelitian ini adalah dua puluh ekor sapi perah peranakan Friesian Holstein, berumur 3-8 tahun sudah pernah beranak minimal satu kali, mempunyai siklus reproduksi normal, dan kondisi tubuh sehat. Seluruh sapi dibagi ke dalam dua kelompok yang masing-masing terdiri atas 10 ekor sapi. Kelompok I adalah sapi fertil sedangkan kelompok II adalah sapi yang telah diinseminasi lebih dari tiga kali tetapi belum atau tidak bunting. Profil biokimia darah yang diukur adalah kadar total protein, total kolesterol, glukosa, dan kalsium. Data dianalisis menggunakan t-test. Hasil penelitian menunjukkan bahwa total protein; total kolesterol; glukosa; dan kalsium pada kelompok I vs kelompok II masing-masing adalah 6,77±0,38 vs 6,82±0,821 g/dl (P>0,05); 166,08±37,06 vs 125,95±38,108 mg/dl (P<0,05); 68,40±9,60 vs 48,58±6,675 mg/dl (P<0,01); dan 9,90±1,43 vs 9,23±0,94 mg/dl (P>0,05). Disimpulkan bahwa sapi-sapi perah yang mengalami kawin berulang mempunyai total kolesterol dan kadar glukosa yang lebih rendah dibanding sapi-sapi perah yang fertil.
Background:Testicular dysfunction is a degenerative disorder characterized by failure in the synthesis of reproductive hormones and spermatogenesis. Secretome derived from the human umbilical mesenchymal stem cell (MSC) has been reported to repair some degenerative disorders.Aim:This study aimed to investigate the effect of secretome derived from the human umbilical MSCs on cisplatin-induced testicular dysfunction in rats.Materials and Methods:Thirty-six male Wistar rats were divided into the control and secretome-treated groups. In the secretome-treated group, testicular dysfunction was induced by 3 mg/kg BW of cisplatin intraperitoneally 3 times with 3-day intervals. The secretome-treated group was divided according to dose: Low-dose (0.2 mL/kg BW) and high-dose (0.5 mL/kg BW) groups. Secretomes were injected intraperitoneally once a week for 3 weeks. 1 week after the injection of secretome, the cauda epididymis of the rats was removed for spermatozoa evaluation and histological examination.Result:After the injection of secretome, the sperm motility of the high-dose group showed thin wave-like, rare, and slow movements. No abnormal sperm morphology was observed in all the treated groups. The number of spermatozoa increased gradually in the high-dose group after the injection of secretome. The developmental stages of the spermatogenic cells were complete in both spermatozoa groups after the injection of secretome. However, the spermatozoa in the seminiferous tubules of the high-dose group were denser. Vimentin and cytokeratin immunoreactivities were very strong in the high-dose group 1 week after the second secretome injection.Conclusion:High-dose secretome derived from the human fetal umbilical cord could increase the number and motility of sperms in rats with cisplatin-induced testicular dysfunction. The administration of high-dose secretome was effective 1 week after the second dose, as indicated by very strong immunoreactivity for vimentin and cytokeratin. Moreover, secretome could promote the regeneration of the seminiferous tubules of both the groups.
Objective: This study was carried out to find out the immunolocalization of Interleukin 6 (IL-6) and Interleukin 10 (IL-10) in the testicular tissue of testicular dysfunction rat treated with secretome from human umbilical stem cells. Materials and methods: Rats were induced with cisplatin for testicular dysfunction condition. After that, the rats were grouped into two categories and were treated with secretome at 0.2 and 0.5 ml/kg BW once every week for 4 weeks. One week later, after the secretome treatment, the rats were sacrificed for histological evaluation using the immunohistochemical method. The preparation slides were examined using a light microscope and were analyzed descriptively and quantitatively. Results: There were no IL-6 and IL-10 immunoreactivities seen in the testicular tissue after cisplatin induction. However, the immunoreactivities of IL-6 and IL-10 were detected after secretome treatment, with both dosages of 0.2 and 0.5 ml/kg BW. These immunoreactivities were detected in the spermatogonia, spermatid/luminal tissue of seminiferous tubule, spermatogenic cells, and Leydig cells. In the cell calculation, the numbers of IL-6 immunoreactive cells were higher at the lower secretome dosage. In contrast, the numbers of IL-10 immunoreactive cells were higher at the higher secretome dosage. Conclusion: IL-6 and IL-10 can be localized in the testicular tissue of testicular dysfunction rat after secretome treatment.
Background and Aim: Reproductive disorders keep the beef cattle population in Jepara Regency, Central Java, Indonesia, from increasing. Ovarian hypofunction and repeat breeding are the most common reproductive disorders, leading to large economic losses for traditional breeders. However, the impact of poor reproductive performance among traditional breeders is not well-known. This study aimed to investigate the reproductive performance of beef cattle with ovarian hypofunction and repeat breeding in Jepara Regency. Materials and Methods: We determined cattle's reproductive status by rectal examination and anamnesis, assessing reproductive performance in 28 cows with repeat breeding and 27 cows with ovarian hypofunction. The following parameters were measured: Postpartum estrous (PPE), days open (DO), service per conception (S/C), and calving interval (CI). The data came from livestock records from animal recording cards, iSIKHNAS, and estimated births from the insemination date that produced a pregnancy. Results: In beef cattle diagnosed with ovarian hypofunction, S/C, PPE, DO, and CI were 1.28, 257 days, 265 days, and 18 months, respectively. The length of CI caused by long PPE may be due to improper nutrition and calf weaning delays. In beef cattle with repeat breeding, S/C, PPE, DO, and CI were 4.15, 106 days, 210 days, and 16 months, respectively. The length of CI was caused by long DO due to pregnancy failure at the first estrus. Conclusion: Hypofunction and repeat breeding reduce the reproductive performance of beef cattle in the Jepara Regency. The cows' health conditions pre- and postpartum can be optimized by providing high-quality feed to enhance reproductive performance.
Background and Aim: Various management practices may cause the occurrence of reproductive failure indicated by repeat breeding in beef cattle. This study aimed to estimate the prevalence and the risk factors of repeat breeding in beef cattle in Sleman Regency, Indonesia. Materials and Methods: Observational and cross-sectional studies were used to determine the prevalence and the risk factors of repeat breeding. Sampling was conducted using a multistage cluster design. The sample size was determined using a sampling formula (n=4 PQ/L2). Questionnaire and interview data were evaluated descriptively. Chi-square analysis and odds ratio (OR) test were conducted to determine the association and association strength with a confidence level of 95%. Univariate, bivariate, and multivariate analysis through multivariate logistic regression test was done using Statistical Package for the Social Sciences, version 21.0 software. Results: The results indicated that the prevalence of repeat breeding in beef cattle in the Sleman Regency was 30.4%. Multivariate analysis indicated that risk factors that significantly affected the repeat breeding were breeding experience (p=0.000; OR=3.378), knowledge of estrus cycle (p=0.000; OR=5.263), feed type (p=0.001; OR=6.061), feeding frequency (p=0.003; OR=2.77), shed hygiene (p=0.33; OR=2,381), and drainage system (p=0.000; OR=4,484). Conclusion: Various husbandry management significantly influence the incidence of repeat breeding in beef cattle in Sleman Regency with the type of feed, which was the highest risk factor. Hay should not be used as the main feed source since it might increase the incidence of repeat breeding. However, the other environmental factors such as season and presence of infection or parasite also need to be investigated further.
