We describe the development and scale-up of a nickel-catalyzed reductive cross-electrophile coupling reaction between a substituted 2-chloropyridine and ethyl 3-chloropropanoate using manganese dust as the terminal reductant. Several additives were screened for the activation of the manganese reductant in situ, and chlorotriethylsilane (TESCl) was found to provide the optimal conversion. A focused beam reflectance measurement (FBRM) probe was utilized to monitor particle attrition as well as manganese activation during the reaction. Modeling was employed to garner an understanding of mixing requirements that would ensure effective suspension of the manganese during scale-up. The process was successfully demonstrated on a 7 kg scale and afforded 2 in 64% yield.

A prominent member of the widely distributed class of guaianolides is arglabin, [1] which inhibits farnesyl transferase and thus activation of the RAS proto-oncogene, a process that is believed to play a pivotal role in 20-30 % of all human tumors. The natural product shows promising antitumor activity and cytotoxicity against different tumor cell lines (human tumor cell lines IC 50 = 0.9-5.0 mg mL À1 ). [2] Arglabin is isolated from Artemisia glabella and is transformed into the hydrochloride salt of the dimethylamino adduct 1·HCl to increase bioavailability. This compound 1·HCl has been successfully used in Kazakhstan for the treatment of breast, colon, ovarian, and lung cancers. [3,4] The heart of arglabin consists of a cycloheptane ring with five contiguous stereocenters, to which two five-membered rings are trans-annulated. The resulting strain can be released by ring opening of the g-butyrolactone at C-2, which makes arglabin and its derivatives prone to attack by nucleophilesa mode of action that plays an integral role in its biological activity. [2,5] The absence of other control factors should cause the planned epoxidation of 2 described in our retrosynthetic analysis (Scheme 1) to preferably occur from the top face of the tricycle to afford a less-strained but undesirable cis junction between the five-and seven-membered rings. Considering this, we reasoned that a hydroxy function at C-8 could direct a suitable epoxidation reagent to the desired lower face, and subsequently allow the installment of the double bond between C-8 and C-9 by elimination of water. Compound 2 should be accessible from 3 by allylation and subsequent ringclosing metathesis (RCM); the latter requires the formation of a tetrasubstituted double bond within a medium-sized seven-membered ring. Following the strategy developed in our research group for the enantioselective synthesis of trans-4,5-disubstituted g-butyrolactones, [6,7] the allylation of cyclopropane 4 with the allylsilane 5 should lead to the first key intermediate 3.Cyclopropanecarbaldehyde 8 was prepared in diastereoand enantiomerically pure form in two steps from methyl-2-furoate (6) on a 50 g scale (Scheme 2) following an analogous Scheme 1. The Lewis acid catalyzed, three-component reaction of isatin and two 1,3-dicarbonyl compounds is reported. Reactions proceed with high efficiency under mild reaction conditions and with good functional group tolerance to afford spirooxindole pyranochromenedione derivatives.

Die daraus resultierende Ringspannung kann durch Ringöffnung des g-Butyrolactons an C-2 abgebaut werden. Deshalb neigt Arglabin auch dazu, Nucleophile bevorzugt an dieser Stelle zu addieren, was für die biologische Aktivität eine wichtige Rolle spielt. [2,5] Die geplante Epoxidierung von 2 in der gezeigten retrosynthetischen Analyse (Schema 1) sollte ohne weitere dirigierende Faktoren bevorzugt von der Oberseite des Tricyclus erfolgen, was zu einer weniger gespannten, aber ungewollten cis-Verknüpfung im Guaiansystem führen würde. Deshalb sollte eine Hydroxygruppe an C-8 eingeführt werden, die ein entsprechendes Epoxidierungsreagens auf die gewünschte Unterseite dirigieren und anschließend die Einführung der Doppelbindung zwischen C-8 und C-9 mithilfe einer Eliminierung ermöglichen sollte.2 sollte ausgehend von 3 durch Allylierung und anschließende Ringschlussmetathese (RCM) zugänglich sein; letztere erfordert einen siebengliedrigen Ringschluss und Bildung einer tetrasubstituierten Doppelbindung. Mithilfe einer von uns entwickelten Strategie zur enantioselektiven Synthese von trans-4,5-disubstituierten g-Butyrolactonen [6,7] sollte die Allylierung von Cyclopropancarbaldehyd 4 durch das chirale Allylsilan 5 somit zum ersten wichtigen Zwischenprodukt 3 führen.Cyclopropancarbaldehyd 8 konnte diastereo-und enantiomerenrein im 50-g-Maßstab in zwei Schritten aus Furan-2-methylester (6) hergestellt werden (Schema 2) -auf analoge Weise wie bereits für dessen Enantiomer beschrieben wurde.[9] Das Allylsilan 11 wurde ausgehend von enantiomerenreinem 9[10] durch trans-selektive Methylcuprat-Addition und anschließende Ni II -katalysierte Kreuzkupplung [11] mit Trimethylsilylmethylenmagnesiumchlorid erhalten. Die durch Trifluorboretherat vermittelte Bildung von 12 erfolgte mit exzellenter doppelter Stereokontrolle, wobei sich das Allylsilan 11 von der entgegengesetzten Seite seines Methylsubstituenten der Carbonylgruppe in 8 in Überein-stimmung mit dem Felkin-Anh-Modell [12]
