Edited by Roger J. Colbran Sonic hedgehog (SHH) is important for organogenesis during development. Recent studies have indicated that SHH is also involved in the proliferation and transformation of astrocytes to the reactive phenotype. However, the mechanisms underlying these are unknown. Involvement of SHH signaling in calcium (Ca) signaling has not been extensively studied. Here, we report that SHH and Smoothened agonist (SAG), an activator of the signaling receptor Smoothened (SMO) in the SHH pathway, activate Ca oscillations in cultured murine hippocampal astrocytes. The response was rapid, on a minute time scale, indicating a noncanonical pathway activity. Pertussis toxin blocked the SAG effect, indicating an involvement of a G i coupled to SMO. Depletion of extracellular ATP by apyrase, an ATP-degrading enzyme, inhibited the SAG-mediated activation of Ca oscillations. These results indicate that SAG increases extracellular ATP levels by activating ATP release from astrocytes, resulting in Ca oscillation activation. We hypothesize that SHH activates SMO-coupled Gi in astrocytes, causing ATP release and activation of G q/11 -coupled P2 receptors on the same cell or surrounding astrocytes. Transcription factor activities are often modulated by Ca patterns; therefore, SHH signaling may trigger changes in astrocytes by activating Ca oscillations. This enhancement of Ca oscillations by SHH signaling may occur in astrocytes in the brain in vivo because we also observed it in hippocampal brain slices. In summary, SHH and SAG enhance Ca oscillations in hippocampal astrocytes, G i mediates SAG-induced Ca oscillations downstream of SMO, and ATP-permeable channels may promote the ATP release that activates Ca oscillations in astrocytes.
With the rapid growth in triboelectric nanogenerator (TENG) technology, selecting appropriate tribomaterial (TM) with high electrical output performance and a cheap fabrication technique is vital for sustainable nanogenerator operations. Thus, we prepared an electrostatic bonded thin poly(diallyldimethylammonium chloride) and poly(4styrene-sulfonic acid) multilayered-based film as a potential positive TM via facile and cheap layer-by-layer (LbL) self-assembly fabrication technique. The high electrical output performance of the LbL-TENGs depends on the number of BL films was optimized by changing the number of bilayers (BLs) from 10 to 50. The 30 BL TENG shows a significant voltage of 189 V and a current density of 17 mA m −2 under the harsh environment of 65% RH, indicating the sustainability of the LbL-TENG device. Furthermore, the single-electrode mode of the optimized TENG exhibited an impressive TENG performance of 215 V and 79 mA m −2 and lit up 39 green light-emitting diodes just by tapping the TENG by hand.
This study was conducted to investigate the effect of feeding the electrolytic materials on blood and carcass traits of broiler during transportation exposed under intense heat condition in summer. The broilers were selected on the day when the outside temperature was about 32℃ to provide heat stressed environment. Broilers reared for 33 d were selected and fed with the electrolytic materials (NaHCO 3, NaCl, KCl) for 2 days. Treatments were as follows; feeding the underground water for control, NaHCO 3 (1.0%) + NaCl (0.5%) for treament 1, KCl (0.5%) + NaCl (0.5%) for treatment 2, KCl (1.0%) + NaCl (0.5%) treatment 3, KCl (0.5%) + NaHCO 3 (1.0%) + NaCl (0.5%) for treatment 4 and KCl (1.0%) + NaHCO 3 (1.0%) + NaCl (0.5%) for treament 5. pH of chicken meat increased for treatments group of electrolytic material, especially, that of treatment 3 was highest when compared to the other treatments. The frequency rate (%) of 1 + quality grade were 33.3, 60.0 and 83.3% at control, treatment 3, 4 and treatment 5, respectively. Occurrence rates of PSE were 50% for control and 13.3% for treatment 5. Corticosterone increased at the post-harvest period compared to the pre-harvest period of broiler and have small disparity between pre-and post-harvest only except treatment 3 when compared to control. pCO 2 partial pressure of blood at the pre-harvest period was low in all treatments by heat stress, the disparity value of control was high for control, and those of treatment 4 and 5 were low compared to other treatments.
ObjectiveWe investigated the efficacy and tolerability of ziprasidone combined with divalproex to determine the relationship between the initial dose of ziprasidone and the treatment effect among Korean patients with acute bipolar manic or mixed disorders.MethodsThis study was a 6-week, open-label, prospective investigation of Korean patients with an acute manic or mixed episode of bipolar disorder. Sixty-five patients were recruited. The patients were categorized based on the initial dose of ziprasidone as follows: low (20-79 mg/day) and standard (80 mg/day). Ziprasidone was given in combination with divalproex in flexible doses, according to the clinical response and tolerability.ResultsThe response and remission rates were significantly higher in the standard-dose group than the low-dose group. The combination of ziprasidone and divalproex was well-tolerated and adverse events were mostly mild with no statistically significant increase in body weight.ConclusionThe results of this study showed that a standard starting dose of ziprasidone in combination with divalproex for bipolar disorder is more effective than a low starting dose.
