Introduction. México is one of the most important producers and exporters of blackberries in the world. During postharvest handling, blackberry fruits are exposed to the attack of phytopathogenic fungi. Materials and methods. To obtain Rhizopus stolonifer isolates, samples of leaves and rhizospheric soils were placed on Petri plates containing Potato Dextrose Agar (PDA). In addition, fruits were also placed in humidity chambers at 25°C. To describe the infection process on blackberry fruits by Rhizopus stolonifer, samples were taken at different intervals for a period of 72 h. All fungal isolates obtained were maintained on PDA. The bacterial colonies were isolated and purified by streaking on PDA. The antagonistic activity of the bacteria was assessed against R. stolonifer by dual culture technique on PDA. To detect the production of siderophores, the chrome azurol S assay was carried out. Results. The infection process of R. stolonifer on blackberry fruits was described for the first time in this work. Eighty-six bacterial isolates from different parts of the plant and rhizospheric soil were obtained. Bacterial isolates with antagonistic activity were identified and the production of siderophores was measured. Four isolates showed antagonistic activity against R. stolonifer. Bacillus subtilis obtained from soil was the most effective isolate. Conclusion. In this study we isolated and identified antagonistic bacteria with potential for biocontrol on Rhizopus stolonifer obtained from blackberry fruits. To our knowledge this is the first report regarding this topic. Résumé-Introduction. Le Mexique est l'un des plus importants producteurs et exportateurs de mûres dans le monde. Au cours de la manutention post-récolte, ces fruits sont exposés à l'attaque de champignons phytopathogènes. Matériel et méthodes. Pour obtenir des isolats de Rhizopus stolonifer, des échantillons de feuilles et de rhizosphère ont été placés en boîtes de Pétri contenant un milieu PDA (Potato Dextrose Agar). En outre, des fruits ont également été placés dans des chambres humides à 25°C. Pour décrire le processus d'infection des mûres par R. stolonifer, des échantillons ont été prélevés à des intervalles différents pendant une période de 72 h. Tous les isolats fongiques obtenus ont été maintenus sur milieu PDA. D'autre part, les colonies bactériennes ont été isolées et purifiées par stries sur le milieu PDA. L'activité antagoniste des bactéries contre R. stolonifer a été évaluée par la technique de double culture sur PDA. Pour détecter la production de sidérophores, un dosage à l'azurol de chrome a été effectué. Résultats. La fresa (Fragaria x ananassa) es un alimento exquisito, que proporciona beneficios a la salud, por lo que es la frutilla de mayor producción y exportación en México. Sin embargo, es altamente perecedera, susceptible de sufrir daños en poscosecha, por Botrytis cinerea y Rhizopus stolonifer, entre otros. El uso de plaguicidas en precosecha es la estrategia de control de estos patógenos; no obstante, está documentado que los plaguicidas dañan la salud humana y al ecosistema, lo que muestra la necesidad de estudiar alternativas amigables. El quitosano grado reactivo (QGR) es un polímero inocuo con actividad antifúngica ampliamente reportada, mientras que el octanoato de sodio (8:0) (OS) también tiene esta propiedad; sin embargo, hasta el momento se desconoce si el quitosano grado comercial (QGC) disponible en México, tiene el mismo efecto. Por lo tanto, el objetivo de esta investigación fue evaluar la efectividad de QGR y el compósito QGR-OS en la protección de fresa en poscosecha y compararla con la de QGC y el compósito QGC-OS. Los compuestos se asperjaron sobre los frutos y se incubaron simulando las condiciones de exportación. Los resultados mostraron reducción significativa de la severidad y la incidencia de las enfermedades fúngicas de fresa en poscosecha luego de la aplicación de QGR, QGC y QGR-OS, QGC-OS, no así del OS aplicado solo. El QGC y el compósito QGC-OS son excelentes candidatos para ser utilizados en un estudio comercial de mayor alcance.
Species of the genus Agave are distributed originally in the tropical and subtropical areas of the American continent with about 200 taxa and 136 species, and its center of origin is probably limited to México. These kind of plants usually grow and live in extreme environmental conditions such as heat and drought where their CAM pathway for fixing CO allow them to survive in conditions where other plants cannot survive. Although this kind of plants resist harsh environmental conditions, climate change is imposing stronger kinds of stress that diminish their productive potential and in some cases are cause of death. Because of this, genetic improvement becomes a need of fundamental importance in this kind of species. Despite their economic importance, Agave species have received scarce attention with regard to its genetic improvement, probably due to their unique botanical features such as plant architecture, spines, long life span, and monocarpy, among others, which make hybridization a difficult task for the intra- and interspecific gene transfer and creation of genetic variability among many other breeding techniques.The protocol here presented is a combination of a novel hybridization technique and biotechnological tools, and allows the use of several procedures for the genetic improvement of agaves such as pollen selection, clonal selection, and somatic cell selection, among others, since the rescued embryos can be used for micropropagation, for phenotype/genotype selection or the production of cell lineages for diverse genetic improvement purposes.
El frijol es una leguminosa, que además de fijar biologicamente el nitrógeno, en su cultivo intensivo demanda aplicaciones importantes de fertilización nitrogenada, siendo la urea el fertilizante más utilizado por su alto contenido de nitrógeno de 46%, y que es abastecida a las plantas a través del follaje por su alta solubilidad, lo cual facilita el óptimo manejo del nitrógeno. Methylobacterium es una -proteobacteria methilotrófica facultativa, colonizador persistente de la filósfera con un potencial metabólico importante, el cual ha demostrado que promueve el desarrollo vegetal, induce resistencia sistémica, atenúa el estrés vegetal y produce un espectro amplio de fitohormonas y enzimas como la ureasa necesaria para la hidrolisis de la urea. El objetivo de este trabajo, fue optimizar la absorción foliar de nitrógeno uréico, empleando de manera concomitante diferentes concentraciones de urea con M. extorquens y para ello se cuantificaron los aminoácidos libres y proteínas solubles, probando 5 tratamientos y un control con M. extorquens y urea al 3 y 4% de manera individual y combinados. Los resultados mostraron una diferencia estadísticamente significativa en la concentración del contenido de amino ácidos libres y proteínas solubles en los tratamientos combinados respecto al control y a los tratamientos individuales.
