we found a graded independent relation between higher RDW and adverse outcomes in ICU patients. RDW has the potentially clinical utility to predict outcome in ICU patients.
AIM:To study the relationship between the polymorphisms in some cytokines and the outcome of hepatitis B virus (HBV) and hepatitis C virus (HCV) infection. METHODS:Samples were obtained from 203 patients infected with HBV and/or HCV while donating plasma in 1987, and 74 controls were obtained from a rural area of North China. Antibodies to HBV or HCV antigens were detected by enzyme-linked immunoassay. The presence of viral particles in the serum was determined by nested reverse-transcriptase polymerase chain reaction (PCR). Hepatocellular injury, as revealed by alanine aminotransferase (ALT) and aspartate aminotransferase level, was detected by a Beckman LX-20 analyzer. DNA was extracted from blood cells. Then, the single nucleotide polymorphisms of IL-2-330, IFN-γ+874, IL-10-1082/-592 and IL-4-589 were investigated by restriction fragment length polymorphism-PCR or sequence specific primer-PCR. CONCLUSION:These results suggest that polymorphisms in some cytokine genes influence persistent HBV and HCV infection, clinical outcome, HCV replication, and liver damage.
The phase transition from vegetative to reproductive growth is a critical event in the life cycle of flowering plants. FLOWERING LOCUS T (FT) plays a central role in the regulation of this transition by integrating signals from multiple flowering pathways in the leaves and transmitting them to the shoot apical meristem. In this study, we characterized FT homologs in the temperate grasses Brachypodium distachyon and polyploid wheat using transgenic and mutant approaches. Downregulation of FT1 by RNAi was associated with a significant downregulation of the FT-like genes FT2 and FT4 in Brachypodium and FT2 and FT5 in wheat. In a transgenic wheat line carrying a highly-expressed FT1 allele, FT2 and FT3 were upregulated under both long and short days. Overexpression of FT1 caused extremely early flowering during shoot regeneration in both Brachypodium and hexaploid wheat, and resulted in insufficient vegetative tissue to support the production of viable seeds. Downregulation of FT1 transcripts by RNA interference (RNAi) resulted in non-flowering Brachypodium plants and late flowering plants (2–4 weeks delay) in wheat. A similar delay in heading time was observed in tetraploid wheat plants carrying mutations for both FT-A1 and FT-B1. Plants homozygous only for mutations in FT-B1 flowered later than plants homozygous only for mutations in FT-A1, which corresponded with higher transcript levels of FT-B1 relative to FT-A1 in the early stages of development. Taken together, our data indicate that FT1 plays a critical role in the regulation of flowering in Brachypodium and wheat, and that this role is associated with the simultaneous regulation of other FT-like genes. The differential effects of mutations in FT-A1 and FT-B1 on wheat heading time suggest that different allelic combinations of FT1 homoeologs could be used to adjust wheat heading time to improve adaptation to changing environments.
IntroductionAlthough nonthyroidal illness syndrome is considered to be associated with adverse outcome in ICU patients, the performance of thyroid hormone levels in predicting clinical outcome in ICU patients is unimpressive. This study was conducted to assess the prognostic value of the complete thyroid indicators (free triiodothyronine (FT3), total triiodothyronine; free thyroxine, total thyroxine, thyroid-stimulating hormone and reverse triiodothyronine) in unselected ICU patients.MethodsA total of 480 consecutive patients without known thyroid diseases were screened for eligibility and followed up during their ICU stay. We collected each patient's baseline characteristics, including the Acute Physiology and Chronic Health Evaluation II (APACHE II) score and thyroid hormone, N-terminal pro-brain natriuretic peptide (NT-proBNP) and C-reactive protein (CRP) levels. The primary outcome was ICU mortality. Potential predictors were analyzed for possible association with outcomes. We also evaluated the ability of thyroid hormones together with APACHE II score to predict ICU mortality by calculation of net reclassification improvement (NRI) and integrated discrimination improvement (IDI) indices.ResultsAmong the thyroid hormone indicators, FT3 had the greatest power to predict ICU mortality, as suggested by the largest area under the curve (AUC) of 0.762 ± 0.028. The AUC for FT3 level was less than that for APACHE II score (0.829 ± 0.022) but greater than that for NT-proBNP level (0.724 ± 0.030) or CRP level (0.689 ± 0.030). Multiple regression analysis revealed that FT3 level (standardized β = -0.600, P = 0.001), APACHE II score (standardized β = 0.912, P < 0.001), NT-proBNP level (standardized β = 0.459, P = 0.017) and CRP level (standardized β = 0.367, P = 0.030) could independently predict primary outcome. The addition of FT3 level to APACHE II score gave an NRI of 54.29% (P < 0.001) and an IDI of 36.54% (P < 0.001). The level of FT3 was significantly correlated with NT-proBNP levels (r = -0.344, P < 0.001) and CRP levels (r = -0.408, P < 0.001).ConclusionIn unselected ICU patients, FT3 was the most powerful and only independent predictor of ICU mortality among the complete indicators. The addition of FT3 level to the APACHE II score could significantly improve the ability to predict ICU mortality.
Spherical silver and gold nanoparticles with narrow size distributions were conveniently synthesized in aqueous solution by a novel electrochemical method. The technological keys to the electrochemical synthesis of monodispersed metallic nanoparticles lie in the choice of an ideal stabilizer for the metallic nanoclusters and the use of a rotating platinum cathode. Poly(N-vinylpyrrolidone) (PVP) was chosen as the stabilizer for the silver and gold clusters. PVP not only protects metallic particles from agglomeration, but also promotes metal nucleation, which tends to produce small metal particles. Using a rotating platinum cathode effectively solves the technological difficulty of rapidly transferring the (electrochemically synthesized) metallic nanoparticles from the cathode vicinity to the bulk solution, avoiding the occurrence of flocculates in the vicinity of the cathode, and ensuring the monodispersity of the particles. The particle size and particle size distribution of the silver and gold nanoparticles were improved by adding sodium dodecyl benzene sulfonate (SDBS) to the electrolyte. The electrochemically synthesized nanoparticles were characterized by TEM and UV/Vis spectroscopy.
A high-resolution pollen record of the past 13000 a from Huguangyan Maar Lake reveals the vegetation and environment changes in southern China during the Holocene. It shows that (i) pollen percentage of trees and shrubs reached 56% during the early Holocene (11600-7800 cal a BP), of which the pollen percentage of tropical trees reached a maximum at 9500-8000 cal a BP, reflecting a hot and wet environment; (ii) during the mid-Holocene (7800-4200 cal a BP), the pollen percentage of montane coniferous trees and herbs increased, while the percentage of tropical-subtropical trees decreased, indicating lower temperature and humidity; (iii) in the late Holocene spanning from 4200 to 350 cal a BP, the pollen percentage of herbs and montane conifer increased significantly, indicating a marked decrease of temperature and humidity. Our pollen data reveal that the time period 9500-8000 cal a BP in southern China represents a climatic optimum for the Holocene characterized by hot and wet conditions. This is consistent with the Holocene optimum found in lower latitude regions globally. We speculate that strong insolation might cause the northward migration of the ITCZ and subtropical summer monsoon front, which resulted in an early Holocene optimum in the Huguangyan area. The dry tendency and climate fluctuations of the middle and late Holocene could be associated with a decrease in solar insolation and frequent ENSO event.
Cubic Pt nanoparticles were prepared by NaBH4 reduction of K2PtCl6 in an aqueous medium. Under the effect of phase-transfer inducer HCl, Pt nanoparticles were transferred to a toluene solution containing dodecanethiol, forming a toluene organosol of cubic Pt nanoparticles protected by dodecanethiol. A monolayer of cubic Pt nanoparticles is obtained on carbon film by self-assembly using this organosol. The stability of the organosol is checked; after being aged for 10 months, the same monolayer of cubic Pt nanoparticles can be obtained using the organosol. The organosol, Pt nanoparticle self-assembled monolayer, and phase transfer were characterized by UV−vis spectra and transmission electron microscopy.
Mechanical properties of methane hydratebearing-sediments (MHBS) are basic parameters for safety analysis of hydrate exploration and exploitation. Young's modulus, cohesion, and internal friction angle of hydratebearing sediments synthesized in laboratory, are investigated using tri-axial tests. Stress-strain curves and strength parameters are obtained and discussed for different compositions and different hydrate saturation, followed by empirical expressions related to the cohesion, internal friction angle, and modulus of MHBS. Almost all tested MHBS samples exhibit plastic failure. With the increase of total saturation of ice and methane hydrate (MH), the specimens' internal friction angle decreases while the cohesion increases.
