Experimental and field studies were conducted to evaluate the effects of NH4+ enrichment on growth and distribution of the submersed macrophyte, Vallisneria natans L, in lakes of the Yangtze River in China, based on the balance between free amino acids (FAA) and soluble carbohydrates (SC) in the plant tissue. Increase of NH4+ rather than NO3– concentrations in the water column caused FAA accumulation and SC depletion of the plant. The plant showed a unimodal pattern of biomass distribution along both FAA/SC ratios and external NH4+ concentrations, indicating that a moderate NH4-N concentration (<0.3 mg L–1) benefited the plant, whereas the high NH4-N concentration (>0.56 mg L–1) eliminated the plant completely. Therefore, 0.56 mg NH4-N mg L–1 in the water column was taken as the upper limit for V. natans in lakes of the Yangtze River basin. The mesocosm experiment showed that at a high external NH4-N (0.81 mg L–1), V. natans failed to propagate with a loss of half SC content (5 mg g–1 DW) in the rhizomes, indicating that the consumption of carbohydrates for detoxification of excess NH4+ into non-toxic FAA significantly diminished carbohydrate supply to the rhizomes. This might consequently inhibit the vegetative reproduction of the plant, and also might be an important cause for the decline and disappearance of the plant with eutrophication. The present study for the first time reports substantial ecophysiological evidences for NH4+ stress to submersed macrophytes, and indicates that NH4+ toxicity arising from eutrophication probably plays a key role in the deterioration of submersed macrophytes like V. natans.
SUMMARY1. Published research suggests that the total phosphorus (TP) thresholds for the regime shifts between a clear-water state dominated by submersed macrophytes and a turbid-water state dominated by phytoplankton in shallow lakes vary with forms of lake basins and climates. However, such hypotheses remain untested by direct field evidence. We therefore tested the hypotheses with empirical data from subtropical lakes on the Yangtze floodplain and also from other lakes in temperate to tropical zones. 2. TP thresholds were found to vary little at moderate depths, but to decrease notably when depth exceeds a level of probably 3-4 m, and increase sharply when depth is below a level of around 1-2 m. 3. TP thresholds were found to be nearly equal in shallow lakes 4. The threshold of turbidity for the forward shift was found to be higher than that for the backward shift, amending the previous hypothesis of the equality of turbidity thresholds for both shifts. 5. Our findings suggest that according to the subequality of TP thresholds, similar target concentrations for in-lake TP can be set in most shallow lakes world-wide to mitigate eutrophication.
A novel fish chemokine receptor gene, chemokine (C-X-C motif) receptor 3 (CXCR3)-like was isolated from the grass carp Ctenopharyngodon idella, with its full-length genomic sequence. The cDNA of grass carp CXCR3-like (gcCXCR3-like) consists of 1261 bp with a 49 bp 5 -UTR and a 189 bp 3 -UTR. An open reading frame of 1023 bp encodes a 341-amino acid peptide, with seven transmembrane helices. The deduced amino acid sequence showed the same sequence identities (37.8%) with its counterparts in goat and human. The gcCXCR3-like gene consists of two exons, with one intervening intron, spaced over approximately 2 kb of genomic sequence. Phylogenetic analyses clearly demonstrated that the gcCXCR3-like resembles the CXCR3s of other vertebrates. Real-time PCR analysis showed that gcCXCR3-like was expressed in all tested organs except heart and the expression level of gcCXCR3-like was highest in brain. Flow cytometric analyses showed the positive rate of labelled leukocytes from the healthy grass carp was 17.3%, and the labelled leukocytes were divided into three types by cell sorting. Immunohistochemical localization revealed that gcCXCR3-like expressed in whole brain regions including cerebel, diencephalon, medulla oblongata, optic lobe, and rhinencephalon, and that the labelled leukocytes are actually populations of monocyte and/or phagocyte, lymphocyte and the granulocyte. It is considered that fish CXCR expression and their function may need to be investigated in both nervous and immune systems.
