Anencephaly is a congenital disease manifesting with the absence of the brain due to the failure of the cranial part of the neural tube to close during the embryonic stage. The disease may be accompanied by other anomalies and usually results in premature death. A stillborn puppy of a 2-year-old female Pomeranian dog is examined in this case. The lack of brain tissue and accompanying abnormal skull formation was noted macroscopically. The eyes were protruding out of their normal position (exophthalmos), and a bifid tongue together with a secondary cleft palate was present. On serial sections stained with Haematoxylin-Eosin, only the medulla spinalis among the central nervous system structures could be inspected microscopically. Immunohistochemistry staining revealed GFAP immunoreactivity in the astrocytic glial cells. NeuN immunoreactivity was detected in the neurons in the medulla spinalis and spinal ganglions. Incomplete retinal layers were observed on the eye sections stained with Haematoxylin-Eosin and NeuN. The case was concluded to be coherent with skull and nervous system congenital malformations rarely observed in dogs. To the best of our knowledge, this represents the first description of a dog with anencephaly, bifid tongue and cleft palate.
OBJECTIVE: One of the most important factors that adversely affects the outcome of peripheral nerve surgery is the formation of epineural and extraneural scar tissue after surgery. Many surgical methods and pharmacological and chemical agents have been used to prevent the formation of epineural scar tissue, but satisfactory results have not been achieved in clinical applications. The purpose of this study was to investigate the combined effect of fat graft and platelet-rich fibrin on the formation of epineural scar tissue and on nerve healing in the mature rat model. METHODS: A total of 24 female Sprague-Dawley rats were used. A circumferential segment of epineurium was excised from both bilateral sciatic nerves. The epineurectomized right nerve segment was wrapped with a combination of fat graft and platelet-rich fibrin (experimental group), while the left nerve segment did not receive any surgical procedure other than the epineurectomy (sham group). Notably, 12 randomly selected rats were sacrificed in the fourth week for histopathological examination of early results. The other 12 rats were sacrificed in the eighth week for late results. RESULTS: The formation of fibrosis, inflammation, and myelin degeneration were less common in the experimental group, while nerve regeneration was found to be higher at both 4 and 8 weeks. CONCLUSION: The intraoperative application of a combination of fat graft and platelet-rich fibrin appears to be effective on nerve healing after surgery at both the early and late periods.
Yarı ince kesitlerin kullanımı histolojik ve klinik tanı çalışmalarında önemli bir yere sahiptir. Fakat bu amaçla kullanılan plastik gömme ortamları hidrofobik oldukları için boyaların dokulara girişini sınırlandırmaktadırlar. Bu çalışmada; yarı ince epon kesitlerin boyanmasında daha çok detay sağlamak üzere bilinen epon solventleri ile asetonun karşılaştırılması ve daha önce denenmemiş toluidine blue-eozin ikili boyamasının değerlendirilmesi amaçlanmıştır. Çalışmada sıçan beyin, ince bağırsak ve pankreas doku örneklerine ait epon bloklardan ultramikrotom ile 1µm kalınlığında yarı ince kesitler alındı. Boyama öncesi epon kesitlere boyaların penetrasyonunu kolaylaştırmak amacı ile sodyum hidroksit, periyodik asit ve aseton ile üç ayrı etching uygulandı. Bir grup kesitte epon uzaklaştırılmadı. Toluidine blue ve toluidine blu-eozin ikili boyaması uygulanan kesitler ışık mikroskobunda değerlendirildi. Aseton ile etching sonrası üç dokuya ait toluidine blue boyama sonuçları, alkali ve asit etchingi sonrası boyamalara ve etching uygulanmadan yapılan boyamalara göre çok daha başarılı olarak değerlendirildi. Özellikle pankreasta daha berrak görüntüler sayesinde daha iyi detay sağladığı görüldü. Toluidine blue-eozin boyaması ile beyin ve pankreas dokularında olumlu sonuçlar elde edildi. Eponun uzaklaştırılması amacıyla ilk kez denenen aseton ile başarılı sonuçlar elde edildiği için diğer solventlere alternatif olarak asetonun rutinde kullanılabileceği düşünüldü. Yine ilk kez denenen toluidine blue-eozin ikili boyamasından epon kesitlerin değerlendirilmesinde yararlanılabileceği sonucuna varıldı.
Temporomandibular eklem (TME), memeli çenesinin hareketleri için gerekli olan sinoviyal bir eklemdir, vücutta en sık kullanılan eklemlerden biridir. Yaşlı hastalarda primer olarak görülebilen, bazı patolojik durumlarda (travma ve internal derangement) sekonder olarak gelişen osteoartrit TME'de en sık görülen artrit tipidir. TME patolojilerinin, kıkırdak dokunun histolojik yapısı gereği iyileşme potansiyeli düşük olduğu için, klinik tedavileri sınırlıdır. Bu çalışmada farklı hücrelere dönüşme potansiyeli olan yağ doku kaynaklı kök hücrelerin, birçok büyüme faktörü barındıran Trombositten Zengin Fibrin (PRF) ile uyarılarak, hasar oluşturulan TME kıkırdağı üzerindeki kombine etkilerinin araştırılması amaçlandı. Çalışmada tavşanların her iki TME eklem disk laterallerinde 3 mm'lik defektler oluşturuldu. Sağ tarafa ense-sırt bölgesinden alınan yağ grefti ve tavşanın kendi kanından hazırlanan PRF yerleştirildi, sol tarafı sham grubu olarak planlanıp başka işlem yapılmadan kapatıldı. 8 hafta sonunda eklem kondil ve diskleri çıkarıldı, Hematoksilen&Eozin (H&E) ve Safranin O-Fast Green metodları ile boyanarak incelendi. Her iki grup için eklem yüzey yapısı, kondrosit yoğunluğu, hücre kümelenmesi ve Safranin O-Fast Green boyanma kaybı histopatolojik olarak derecelendirildi. Değerlendirilen parametreler arasında anlamlı fark saptanmadığı için, deneysel TME defekt modelinin onarımında yağ grefti ve PRF kombinasyonu etkili bulunmadı.
