Phytochemical studies of the leaves and stem have led to the identification of the known coumarins isooxypeucedanin, oxypeucedanin hydrate, xanthotoxin, isopimpinellin, 8-methoxymarmesin and marmesin, flavonoids quercetin-3-O-α-L-rhamnopyranoside, myricetin-3-O-α-Lrhamnopyranoside and hesperidin, alkaloids skimmianine and N-methylflindersine and limonoid limonin. The compounds isolated and the chemical profile of Euxylophora obtained from the literature clearly indicate its phytochemical affinities with other Rutoideae species.Keywords: Euxylophora paraensis; Rutaceae; chemosystematics. INTRODUÇÃOA família Rutaceae constitui o maior grupo de plantas da ordem Sapindales, possuindo cerca de 160 gêneros, com 1900 espécies amplamente distribuídas nas regiões tropicais e temperadas do globo terrestre, sendo mais abundante na América tropical, sul da África e Austrália.1,2 No Brasil existem cerca de 200 espécies já descritas. 2As plantas desta família são muito conhecidas pela presença de uma ampla diversidade de metabólitos secundários, destacando-se os alcaloides, especialmente aqueles derivados do ácido antranílico, as cumarinas, as lignanas, os flavonoides e os limonoides. 1,3,4 Existem relatos de largo espectro de atividades biológicas associadas a estas classes de metabólitos. 1,3 O gênero Euxylophora é monotípico, apresentando somente a espécie Euxylophora paraensis de ocorrência no norte do Brasil. 5Ela é conhecida popularmente como pau-amarelo devido à cor de sua madeira, a qual é de excelente qualidade, encontrando aplicação na confecção de diversos móveis. Suas árvores de grande porte constam na lista de espécies da flora brasileira ameaçadas de extinção desde 1993.6 Segundo a literatura, foram isolados de E. paraensis somente alcaloides do tipo 2(1H)-quinolinona, 4(1H)-quinolinona, 7-9 indolopiridoquinazolínico e bis-2(1H)-quinolinona. [8][9][10][11][12][13][14][15][16] Estas substâncias resultaram do estudo químico da casca do caule e do cerne, não havendo, até o momento, citação sobre fitoquímica das folhas de E. paraensis. As cumarinas são de ocorrência ampla nos gêneros de Rutaceae, ou seja, são marcadores químicos da família. A ausência de citações destes compostos para este gênero despertou o interesse no estudo de outros órgãos na tentativa de se buscar as cumarinas. A análise preliminar via cromatografia liquida de alta eficiência (CLAE-UV) de extratos das folhas de E. paraensis indicou a presença das cumarinas, o que levou ao presente estudo. Assim, este trabalho relata o primeiro estudo químico das folhas de E. paraensis que levou ao isolamento das cumarinas iso-oxipeucedanina, oxipeucedanina hidratada, xantotoxina, isopimpenilina, dos flavonoides quercetina-3-O-α-L-ramnopiranosídeo, miricetina-3-O-α-L-ramnopiranosídeo e hesperidina e do alcaloide esquimmianina. Bacterial spot, which is the main disease occurring in passion fruit trees, is caused by the bacterium Xanthomonas axonopodis pv. passiflorae, leading to large annual losses in passion fruit crops. This study aims to find extracts and/or bioactive compounds of endophytic fungi of noni (Morinda citrifolia) to treat bacterial spot in passion fruit trees. Nine fungi isolated from a specimen of M. citrifolia from the Brazilian Amazon are studied. The fungus Guignardia mangiferae NF17 shows the best inhibition results and is selected for the isolation of its secondary metabolites by chromatography techniques. The isolated compounds Sydowinol (S1) and Sydowinin A (S2) are identified by nuclear magnetic resonance (NMR). Compounds S1 and S2, as well as the acetonitrile extract from the biomass of G. mangiferae NF17, are tested against four strains of X. axonopodis pv. passiflorae obtained from plants infected by bacterial spot, and which inhibited bacterial growth up to the lowest concentration tested (3.125 µg/mL). This study reports, for the first time, the antibacterial activity against X. axonopodis pv. passiflorae by the compounds Sydowinol and Sydowinin A. Compounds S1 and S2 are reported for the first time for the genus Gignardia.
O descarte inadequado de resíduos industriais ou acidentes ambientais tem causado grandes impactos ambientais, como a contaminação por metais pesados. Processos biotecnológicos envolvendo microrganismos têm sido utilizados como estratégia para recuperar ambientes impactados. O objetivo deste trabalho foi avaliar a influência de metais pesados essencial e não essencial no crescimento e na produção de surfactante da linhagem Pseudomonas fluorescens Slim15. Os testes de concentração inibitória mínima (CIM) e concentração bactericida mínima (CBM) foram utilizados para avaliar a tolerância aos metais Pb e Zn. Foram realizados testes de atividade emulsificante e tensão superficial para avaliar a influência dos metais na produção de biossurfactante. A linhagem P. fluorescens Slim15 apresentou crescimento na presença de 31,25 mg/L de Zn e de 1000 mg/L de Pb no teste CIM e de 4000 mg/L no teste CBM para os metais testados. Os metais não apresentaram influência significativa na produção de biossurfactante. Esta pesquisa mostrou que a linhagem P. fluorescens Slim15 apresentou rendimento satisfatório nos testes de tolerância e produção de surfactante, mesmo em altas concentrações de metais. Esta linhagem bacteriana é uma promissora agente biorremediadora de ambientes contaminados por metais pesados.
Cancer is one of the most deadly diseases in the world, being related to the disordered growth of defective cells which produces harmful substances to the body. In this context, one of the great challenges of science is finding an effective bioactive compound for treating of this disease and microorganisms from restrictive environments, such as caves, are indicated as a promising source of these compounds. The objective of this work was to evaluate the extracts cytotoxic effects obtained from cultures of two bacterial strains isolated from Pedra da Cachoeira cave, Altamira-PA, located in a preserved forest fragment in the Eastern Amazon. Bacteria were identified by DNA sequencing analysis encoding ribosomal RNA. The choice of the ideal period of microbial development, aiming at the biocompounds production, was based in a kinetic study realized from a growth curve. Organic extracts were produced after 30 hours (stationary phase) and 40 hours (decline phase) of cultivation, obtaining four extracts by liquid -liquid partition of culture medium and four extracts from the bacterial cells. In vitro cytotoxicity tests were performed with B16F10 tumor cells using the MTT method. From the phylogenetic analyzes of the DNA sequences of both strains, a cluster with the species Bacillus subtilis was verified. The extracts obtained were subjected to cytotoxic assays and their cell viability was evaluated based on the minimum inhibitory concentration (IC50) and selectivity index (IS), with the CV26AI30H extract having an IC50 value of 83.99 and IS greater than 4.70.
