Expression levels of the NAC gene family were studied in rice infected with Rice dwarf virus (RDV), Rice black-streaked dwarf virus (RBSDV), Rice grassy stunt virus (RGSV), Rice ragged stunt virus (RRSV), and Rice transitory yellowing virus (RTYV). Microarray analysis showed that 75 (68%) OsNAC genes were differentially regulated during infection with RDV, RBSDV, RGSV, and RRSV compared with the control. The number of OsNAC genes up-regulated was highest during RGSV infection, while the lowest number was found during RTYV infection. These phenomena correlate with the severity of the syndromes induced by the virus infections. Most of the genes in the NAC subgroups NAC22, SND, ONAC2, ANAC34, and ONAC3 were down-regulated for all virus infections. These OsNAC genes might be related to the health stage maintenance of the host plants. Interestingly, most of the genes in the subgroups TIP and SNAC were more highly expressed during RBSDV and RGSV infections. These results suggested that OsNAC genes might be related to the responses induced by the virus infection. All of the genes assigned to the TIP subgroups were highly expressed during RGSV infection when compared with the control. For RDV infection, the number of activated genes was greatest during infection with the S-strain, followed by the D84-strain and the O-strain, with seven OsNAC genes up-regulated during infection by all three strains. The Os12g03050 and Os11g05614 genes showed higher expression during infection with four of the five viruses, and Os11g03310, Os11g03370, and Os07g37920 genes showed high expression during at least three viral infections. We identified some duplicate genes that are classified as neofunctional and subfunctional according to their expression levels in different viral infections. A number of putative cis-elements were identified, which may help to clarify the function of these key genes in network pathways.
An assessment of morphological variation among 70 accessions of greater yam (Dioscorea alata L.) collected throughout Malaysia was made. Data of 47 morphological variables measured from the accessions were subjected to multivariate analysis using principal component (PCA) and cluster analysis (CA). The PCA results indicated that the characters contributing largely to the species variability were those related to the shape, size and flesh colour of underground tubers; shape and colour of aerial tubers; position, shape, size and vein colour of the leaves; petiole colour; shoot growth rate; and number of days for shoots to germinate. The two-dimensional plot of the first two PCs showed a separation between accessions of purple tuber groups and those of white tuber groups, but was unable to distinguish accessions according to tuber shape groups, i.e. irregular, oblong and round, as revealed by visual observation. The dendrogram of CA revealed four major groups of D. alata in Malaysia, which supported the PCA grouping. This study demonstrated that D. alata in Malaysia consists of numerous genotypes revealing wide inter- and intra-group variability.
Abstract.Mushroom, as believed by many people, grows on specific time and condition as in the wild it grows after a heavy rain falls. The effects of lightning and thunderstorm may suppress the mychorrizal structure to grow and eventually forming fruiting body. This study was conducted to determine the effect of different acoustic sound treatments on the growth and yield of grey oyster mushroom (Pleurotussajor-caju). Five different acoustic sound treatments had been applied during spawning period which were thunder storm, hardcore music, soothing instrumental, Quranic recital and without any sound treatment applied which served as control. The parameters studied were mycelium growth rate, days of mycelium filled up the bags, days of pinhead emergence, days of fruiting body formation, total weight, percentage biological efficiency, pileus color and texture. There were significant differences (P<0.05) in the mycelium growth among different acoustic sound treatments where sound treated mushroom showed faster growth than control. Similar trend was also observed in yield (total weight and biological efficiency) where there were significant differences (P<0.05). However, there were no significant differences (P>0.05) observed in other parameters, such as pinhead emergence, fruiting bodies formation, pileus color and texture. In summary, treatments using different acoustic sound at 75 dB could be considered as better treatment to enhance the mycelium growth thus accelerate the mushroom cultivation process as well as increasing the mushroom productivity. This research could help farmers to grow and harvest their mushroom at specific time frame and fulfill customer's demand.
