Abstract:The southern United States is characterized by a humid, subtropical climate and consists of 16 states (Texas, Oklahoma, Arkansas, Louisiana, Mississippi, Tennessee, Kentucky, Alabama, Florida, Georgia, South Carolina, North Carolina, Virginia, West Virginia, Delaware, and Maryland) and Washington DC. Currently this region is experiencing the largest net population growth in the U.S. Over the last century, the expansion of large urban centers and impervious area in the region has altered the hydrologic cycle. This review synthesizes regional research that shows how watershed hydrology, groundwater recharge, stream geomorphology, climate, biogeochemistry, and stream ecology have been affected by urbanization and the expansion of watershed impervious area.

We report the results of pyrosequencing of DNA collected from the activated sludge basin of a wastewater treatment plant in Charlotte, NC. Using the 454-FLX technology, we generated 378,601 sequences with an average read length of 250.4 bp. Running the 454 assembly algorithm over our sequences yielded very poor assembly, with only 0.3% of our sequences participating in assembly of significant contigs. Of the 117 contigs greater than 500 bp long that were assembled, the most common annotations were to transposases and hypothetical proteins. Comparing our sequences to known microbial genomes showed nonspecific recruitment, indicating that previously described taxa are only distantly related to the most abundant microbes in this treatment plant. A comparison of proteins generated by translating our sequence set to translations of other sequenced microbiomes shows a distinct metabolic profile for activated sludge with high counts for genes involved in metabolism of aromatic compounds and low counts for genes involved in photosynthesis. Taken together, these data document the substantial levels of microbial diversity within activated sludge and further establish the great utility of pyrosequencing for investigating diversity in complex ecosystems.Although largely invisible in the urban landscape when they are functioning well, wastewater treatment plants are integral to the municipal obligation to protect public health, aquatic ecosystems, and the quality of life. At the heart of wastewater treatment plants is a process whereby a dense microbial consortium is employed to remove organic and nutrient contaminants. The microbes used to treat wastewater are a crucial tool in environmental protection. The current use of molecular techniques that do not require the isolation and cultivation of microorganisms (1, 33), including 16S rRNA analysis (6,13,20) and fluorescent in situ hybridization (8), have greatly expanded our understanding of wastewater microbial communities. Researchers have identified many bacteria of importance to wastewater treatment, including the bacteria involved in biological phosphorus removal (5, 16, 29), nitrifiers (8, 19, 25), denitrifiers (3, 12, 17), and methanogens (18, 36). Molecular techniques have also improved our understanding of fundamental processes such as nitrification and denitrification, as well as plant upsets, such as foaming (9, 24), which can decrease treatment efficiency.In this paper, we apply recently developed pyrosequencing technology to probe the molecular diversity of the aerobic basin of a wastewater treatment plant in Charlotte, NC. In line with other studies of complex microbial communities (28, 32), we observed astounding levels of diversity. We found that substantial regions of the genomes of the most prevalent microbes in the wastewater treatment plant are poorly described by existing sequence databases. Women are underrepresented in a number of science, technology, engineering, and mathematics (STEM) disciplines. Limited diversity in the development of the STEM workforce has negative implications for scientific innovation, creativity, and social relevance. The current study reports the first-year results of the PROmoting Geoscience Research, Education, and SuccesS (PROGRESS) program, a novel theory-driven informal mentoring program aimed at supporting first- and second-year female STEM majors. Using a prospective, longitudinal, multi-site (i.e., 7 universities in Colorado/Wyoming Front Range & Carolinas), propensity score matched design, we compare mentoring and persistence outcomes for women in and out of PROGRESS (N = 116). Women in PROGRESS attended an off-site weekend workshop and gained access to a network of volunteer female scientific mentors from on- and off-campus (i.e., university faculty, graduate students, and outside scientific professionals). The results indicate that women in PROGRESS had larger networks of developmental mentoring relationships and were more likely to be mentored by faculty members and peers than matched controls. Mentoring support from a faculty member benefited early-undergraduate women by strengthening their scientific identity and their interest in earth and environmental science career pathways. Further, support from a faculty mentor had a positive indirect impact on women’s scientific persistence intentions, through strengthened scientific identity development. These results imply that first- and second- year undergraduate women’s mentoring support networks can be enhanced through provision of protégé training and access to more senior women in the sciences willing to provide mentoring support.

Stormwater control measures (SCMs) are designed to mitigate deleterious effects of urbanization on river networks, but our ability to predict the cumulative effect of multiple SCMs at watershed scales is limited. The most widely used metric to quantify impacts of urban development, total imperviousness (TI), does not contain information about the extent of stormwater control. We analyzed the discharge records of 16 urban watersheds in Charlotte, NC spanning a range of TI (4.1 to 54%) and area mitigated with SCMs (1.3 to 89%). We then tested multiple watershed metrics that quantify the degree of urban impact and SCM mitigation to determine which best predicted hydrologic response across sites. At the event time scale, linear models showed TI to be the best predictor of both peak unit discharge and rainfall-runoff ratios across a range of storm sizes. TI was also a strong driver of both a watershed's capacity to buffer small (e.g., 1-10 mm) rain events, and the relationship between peak discharge and precipitation once that buffering

Insights into the effects of stormwater control measures (SCMs) on urban stream hydrology and in-stream processes are required to understand their effectiveness in mitigating the environmental problems associated with urbanization. Stable water isotopes were applied to understand processes occurring within SCMs and their effects on water sourcing in urban streams. We sampled ten events from June to November 2013 at four locations along a 360-m headwater stream reach in North Carolina and at four SCMs (two ponds, one wetland and one bioretention) that contribute to the reach. We used streamflow upstream of the SCMaffected reach and outflow from an intensively sampled retention pond as endmembers to quantify contributions of this pond's outflow to streamflow. Synchronous sampling revealed that SCM outflows have different isotopic signatures, likely a function of evaporation and mixing within the storage volume of each SCM. The SCMs also have distinctive isotopic signatures relative to the receiving stream. The isotopic signature of discharge from the intensively sampled pond reveals varying residence times (hours to weeks) within the structure. At sampled timepoints during ten events, this pond, which drains 25% of the watershed's impervious area, contributed an average of 10% (0-21%) of the streamflow on the rising limb and 12% (0-19%) of discharge at peak flow. During recession, this pond contributed an average of 32% (11-54%) of the stream's discharge, reflecting the SCM's design goals of temporarily storing and delaying run-off, mitigating the effects of impervious surface on peak flows. Based on this study, isotopes appear to be a robust tool for examining stormwater-stream dynamics.Where values are not provided, uncertainty bands spanned 0-100%.
