Trichuris nematodes were isolated from roe deer (Capreolus capreolus). At first, nematodes were determined using morphological and biometrical methods. Subsequently genomic DNA was isolated and the ITS1-5.8S-ITS2 segment from ribosomal DNA (RNA) was amplified and sequenced using PCR techniques. With u sing morphological and biometrical methods, female nematodes were identified as Trichuris globulosa, and the only male was identified as Trichuris ovis. The females were classified into four morphotypes. However, analysis of the internal transcribed spacers (ITS1-5.8S-ITS2) of specimens did not confirm this classification. Moreover, the female individuals morphologically determined as T. globulosa were molecularly identified as Trichuris discolor. In the case of the only male molecular analysis match the result of the molecular identification. Furthermore, a comparative phylogenetic study was carried out with the ITS1 and ITS2 sequences of the Trichuris species from various hosts. A comparison of biometric information from T. discolor individuals from this study was also conducted.
The occurrence of Hypoderma diana (Diptera: Hypodermatidae) in a herd of 15 horses was observed during a 4-year period (2010-2013) in northwest Bohemia. During this period, infestation by the warble fly H. diana increased from 12.5% in 2010 to 53.3% in 2013. Nodules were observed on the neck, back, stomach, legs, and head. The incidence of H. diana was probably influenced by the presence of a specific host mouflon (Ovis orientalis musimon).
Heavy metal (Cd, Cr, Cu, Fe, Mn, Ni, Pb and Zn) levels in red fox small intestine samples with or without Echinococcus multilocularis infection were studied. The red foxes were taken from the open countryside of northwest Bohemia (CR). Red foxes with E. multilocularis infection had lower levels of toxic metals (Cd, Pb); cadmium levels in infected foxes (0.0052 mg/kg) were twice as low as in uninfected foxes (0.0106 mg/kg). This was the same case for lead: 0.0288 mg/kg infected red foxes (inf.) and 0.0413 mg/kg uninfected (uninf.). Conversely, red foxes with E. multilocularis infection yielded higher concentrations in comparison to their uninfected counterparts: Cr (0.0087 mg/kg uninf. and 0.0116 mg/kg inf.), Cu (0.2677 mg/kg uninf. and 0.3205 mg/kg inf.), Fe (6.46 mg/kg uninf. and 10.89 mg/kg inf.), Mn (0.1966 mg/kg uninf. and 0.2029 mg/kg inf.), Ni (0.0415 mg/kg uninf. and 0.064 mg/kg inf.) and Zn (16.71 mg/kg uninf. and 20.25 mg/kg inf). This could support the hypothesis that tapeworms are able to absorb toxic heavy metals from the host body into their tissues, as well as to modify other element concentrations in the host body.
Nine ponies (20 months of age) with naturally acquired cyathostome and pinworm infections were treated orally with ivermectin at a dosage of 400 μg /kg. Ivermectin was highly effective on the cyathostome infection. However, adult Oxyuris equi were present in six horses. In recent years, anthelmintic treatment with macrocyclic lactones have not appeared to deliver the expected efficacy against equine pinworms (Oxyuris equi) in the USA. This is the first European study to demonstrate anthelmintic resistance in Oxyuris equi to macrocyclic lactones in naturally infected ponies (Czech Republic).
Trichuris sp. individuals were collected from Myocastor coypus from fancy breeder farms in the Czech Republic. Using morphological and biometrical methods, 30 female and 30 male nematodes were identified as Trichuris myocastoris. This paper presents the first molecular description of this species. The ribosomal DNA (rDNA) region, consisting of internal transcribed spacer (ITS)-1, 5.8 gene and ITS-2, was sequenced. Based on an analysis of 651 bp, T. myocastoris was found to be different from any other Trichuris species for which published sequencing of the ITS region is available. The phylogenetic relationships were estimated using the maximum parsimony methods and Bayesian analyses. T. myocastoris was found to be significantly closely related to Trichuris of rodents than those of ruminants.
SummaryNine ponies were experimentally infected with third stage cyathostome larvae. These animals were examined post-mortem for small strongyle infections from October 2011 to January 2012. Seventeen species of the following Cyathostominae genera were identifi ed: Coronocyclus, Cyathostomum, Cylicocyclus, Cylicostephanus, Gyalocephalus and Petrovinema. Of the 17 species of Cyathostominae recovered, Cyathostomum catinatum, Cyathostomum alveatum and Cylicocyclus nassatus were the most prevalent (100 %). From the entire population of Cyathostominae, 65.6 % were located in the caecum and only 34.4 % in the colon. Only 1,736 (24,6 %) females and 5,329 (75,4 %) males were observed. There was a signifi cant difference between the number of males and females in the genus Cyathostomum, Cylicocyclus and Gyalocephalus.
Various laboratory animals -mice (Mus musculus) of six strains, rabbits (Oryctolagus cuniculus), guinea pigs (Cavia porcellus), rats (Rattus norvegicus), and Mongolian gerbils (Meriones unguiculatus) were experimentally infected with larvae of small strongyles (Cyathostominae), obtained from horse faeces and cultured to the infective larval stage L3. The attempt to transfer cyathostome larvae was aimed at developing a model for the investigation of different aspects of the life cycle and biology of these nematodes in the laboratory. Some animals were immunized (hydrocortisone) for the duration of the study. The laboratory animals were orally infected with 2-10 thousand sheathed or ex-sheathed L3 larvae of mixed cyathostome species. All attempts to inoculate any animal failed; there was no larval development in the experimental rodents and it can be stated that none of the investigated animals may serve as a suitable model host for horse nematodes of the subfamily Cyathostominae.
