Changes in the thermal transition parameters of the backbone proteins of the erythrocyte plasma membrane, particularly of spectrin and membrane domain of band 3 protein, in patients with hereditary hemochromatosis are demonstrated by differential scanning microcalorimetry. Electron microscopy (the freeze-fracture technique) reveals aggregation of intramembrane particles and their enlargement in erythrocyte membranes from patients in comparison with membranes from healthy donors. These structural changes in the erythrocyte membranes are assumed to contribute to hemolysis and iron overload of the organism. Key Words: hereditary hemochromatosis; differential scanning microcalorimetry; erythrocyte membraneHereditary hemochromatosis (HH) is a pathological state that results from disturbed iron metabolism and leads to intracellular accumulation and deposition of iron-containing pigments inducing irreversible structural and functional changes in cells [6,9,19]. The key role in iron metabolism is played by macrophages and erythrocytes [8,10,19]. The role of the macrophage system in the pathogenesis of HH is well established. Macrophages cannot catabolize and effectively store hemoglobin-derived iron [6][7][8][9]19]. The role of erythrocytes is poorly understood. However, splenomegaly [2,6,19] and hemolysis [5] in patients with HH point to some defects in the erythrocyte plasma membrane. An increase in serum iron and ferritin contents may result from excessive phagocytosis of defective erythrocytes. In the present study we compared thermal transitions in the erythrocyte membranes from patients with HH and healthy donors. MATERIALS AND METHODSThe study was performed on erythrocytes from 22 male patients with HH aged 17-65 years and two boys (9-and l 1-year-old). The duration of HH varied from 1 month to 6 years. All patients had symptoms typical of the late stage of the disease: cirrhosis of the liver, diabetes mellitus, splenomegaly, and melanoderma. Laboratory tests revealed specific signs of the disease in all cases: iron metabolism disturbances and the presence of A3 and B7 loci. In half of the patients, ferritin of erythrocyte lysate considerably surpassed the normal level, varying from 12 to 773.8 gg/g Hb (vs. 5.9 in the norm). Examination of HH patients revealed intravascular hemolysis usually accompanied by elevation of free plasma hemoglobin.

Алиментарно-функциональные диареи телят являются наиболее распространенными среди всех заболеваний молодняка крупного рогатого скота, при этом они могут осложняться условно-патогенными и патогенными микроорганизмами. Поэтому лабораторные исследования патологического материала -это неотъемлемая часть постановки точного диагноза, на что требуется несколько дней. В связи этим у ветеринарных специалистов существует необходимость в разработке схем терапевтических мероприятий, направленных на предупреждение развития условнопатогенной и патогенной микрофлоры и интоксикации организма, восстановления функции желудочно-кишечного тракта и работы печени с дальнейшей корректировкой на основании полученных результатов лабораторных исследований. С этой целью были отобраны телята с выраженными клиническими признаками алиментарно-функциональной диареи и сформированы контрольная и опытная группы. Животным двух групп применяли схему лечения принятую в хозяйстве. В опытной группе дополнительно, на фоне комплексного применения витаминов (Pyridoxine hydrochloride, Retinoli acetatis, alfa-Tocopherol acetate, Colecalciferolum), использовали препараты, оказывающие гепатопротекторное и спазмолитическое действия. После проведенных мероприятий у всех исследуемых животных клинические признаки заболевания отсутствовали, а в опытной группе была установлена положительная динамика показателей крови. Так, отмечали восстановление таких показателей, как холестерин, глюкоза, альбумины, билирубин, свидетельствующие о восстановлении процессов пищеварения, об активизации обменных процессов, нормализации работы печени. При этом ядерный индекс не превышал 0,1. Таким образом, применение гепатопротекторного средства, миотропного спазмолитика и комплекса витаминов в качестве дополнительной терапии при алиментарно-функциональной деареи телят способствовало детоксикации, восстановлению функции пищеварения, улучшению процессов регенерации и нормализации функции печени.
The article presents data on the study of metabolism in calves two weeks before diarrhea at the age of 19-52 days of life. For this purpose, three groups of conditionally healthy animals of the Holstein breed were formed at the age of 19, 30, 52 days of life. To assess the metabolic status of young cattle, biochemical and general clinical blood tests were carried out. In the course of a laboratory blood test in young cattle, two weeks before disease manifestation, disorders of mineral and protein metabolism, a decrease in liver function were established. At the same time, with the increase in the age of the calves, the imbalance in protein metabolism was aggravated against the background of a change in diet and mineral metabolism improved as a result of the maturation of the digestive system.
