Natural products produced by microorganisms have been utilized as sources of new drugs possessing a wide range of agrochemical and pharmacological activities. During our research on Actinomycetes from Brazilian mangroves, the ethyl acetate extract of Streptomyces sp. AMC 23 isolated from the red mangrove (Rhizophora mangle) rhizosphere produced a highly active compound against the microalga Chlorella vulgaris, often used to assess the phytotoxic activity. As a result, the bioassay-guided fractionation led to the isolation of the mixture of the known compounds bafilomycin B1 and bafilomycin B2. The chemical structures of bafilomycin B1 and bafilomycin B2 were established based on their spectroscopic data by infrared (IR), mass spectrometry (MS), (1)H nuclear magnetic resonance (NMR), gradient-enhanced heteronuclear multiple quantum coherence (gHMQC), and gradient-enhanced heteronuclear multiple-bond connectivity (gHMBC) as well as comparison with reference data from the literature. Moreover, it was also possible to identify other bafilomycins using non-chromatographic-dependent techniques (Tandem mass spectrometry). Additionally, this is the first report on the phytotoxic activity of bafilomycin B1.
The main aim was to identify the active compound against Rhizoctonia solani produced by the cassava endophyte Paenibacillus sp. IIRAC-30. The compounds produced were extracted with ethyl acetate and purified by Sephadex column prior to analysis by Q-TOF mass spectrometry. A C 15 -lipopeptide with an estimated molecular weight of 1036 Da and homologues were identified. The lipopeptide had a cyclic structure, which was deduced by interpreting the ESI-MS/MS spectra of main protonated homologues containing 15:0 FA, and the amino acid composition was Glu-Leu-Leu-Val-Asp-Leu-Leu. Therefore, the lipopeptides produced by isolate IIRAC-30 was characterized as a surfactin series. Thus, the main mechanism used by Paenibacillus sp. IIRAC-30 to suppress R. solani was elucidated. Furthermore, because lipopeptides active against phytopathogens generally show low toxicity to humans and the environment, the positive findings presented here suggest that the isolate IIRAC-30 could be a possible candidate for biocontrol of R. solani.
Mangroves are coastal ecosystems which occur in the areas of transition between marine and terrestrial environments throughout the tropical and subtropical regions, suffering direct tide influence. The diverse microbial communities are important to the nutrient cycling occurrence and consequently to the maintenance of these environments fauna and flora biodiversity. The Actinobacteria are a heterogeneous group of filamentous bacteria that naturally inhabit the soil. They can easily adapt to different environmental conditions, producing antibiotics and colonizing the rhizosphere and plants internal tissues. This study aims were: to allow the assessment of the diversity of rhizosphere Actinobacteria isolated from mangroves located in Cananeia and Bertioga, cities under the jurisdiction of Sao Paulo State; to evaluate the production of antifungal secondary metabolites of these microorganisms; to analyze the chemical profile of organic extracts of Actionobacteria by mass spectrometry, followed by the evaluation of the chemical components of theses extracts through thin-layer chromatography and preparative column for their separation. After this process minimum inhibitory concentration of the active fractions of the bioassays were determined.
