The bacurizeiro (Platonia insignis Mart.) is a tree native to the Amazon whose fruit is much used in the gastronomy in the North and Northeast regions of Brazil. Due to its great economic potential for these regions, the species is being conserved in germplasm banks to support genetic breeding programs. The aim of this work was the molecular characterization of P. insignis accessions belonging to the germplasm bank of the Embrapa Eastern Amazon research unit using ISSR (Inter Simple Sequence Repeat) markers. Seventy-eight accessions of P. insignis belonging to 16 progenies were sampled in two different localities on Marajó Island, state of Pará, Brazil. Among the 16 progenies, seven were collected in Soure and nine in Salvaterra. The 78 accessions were genotyped with 23 pre-selected primers. We obtained 121 amplified products, of which 54 were polymorphic. The most polymorphic primers were UBC 834, UBC 899 and UBC 900. Primers UBC810 and UBC884 did not amplify polymorphic bands. Forty-nine markers out of 54 were selected for genetic analyses. AMOVA within and among progenies showed low genetic differentiation (Φ PT = 0.064, P<0.001) with higher diversity within progenies (96%), low genetic differentiation among sampling localities (Φ PT = 0.025, P<0.013), and higher diversity within (98%) than among localities. Clustering by UPGMA based on Jaccard similarities among pairs of accessions did not separate genotypes according to progeny or sampling localitiy. We recommend that new germplasm surveys consider a greater sampling effort within sampling localitites. Platonia insignis is a fruit tree native of Brazil with allogamous and asexual reproduction. The production of fruits is mainly obtained by exploitation of natural populations and the impact of genetic structuring on plant production may be evaluated. For this purpose, codominant and multiallelic markers such as microsatellite are the most suitable, but they need to be developed for this species. Thus, the aim of this work was to develop and validate microsatellite markers for P. insignis. We used Roche 454 GS FLX sequencing platform of a single P. insignis genotype and 1702 microsatellite sequences were identified. Based on some pre-requisites, we could develop 50 primer pairs to be tested. Twenty-two primer pairs successfully amplified fragments and they were tested in 31 genotypes of P. insignis that belong to a germplasm bank and were sampled in the northeast of Pará State, Brazil. Thirteen primers were polymorphic and the number of alleles per loci varied from 5 (PI18 and PI27) to 2 (PI08, PI25, PI31, PI33 and PI 37). Expected heterozygosity (H E ) varied from 0.74 (PI27) to 0.12 (PI31) and observed heterozygosity (H O ) varied from 1.00 (PI25) to 0.00 (PI08, PI31, PI33 and PI37). Principal coordinates could separate the genotypes of P. insignis in clusters and we can conclude that the primers can estimate the genetic diversity of P. insignis populations.
Culturalmente, o estacão de madeira é usado para o cultivo da pimenteira-do-reino no campo, contudo, o tutor de gliricídia mostrou-se como uma opção sustentável e viável para uso no sistema de produção da espécie. Apesar da redução dos custos para implantação dos pimentais, há pouca adesão da tecnologia pelo agricultor, em decorrência de escassez de informações sobre o comportamento de cultivares. Dispondo-se avaliar cultivares sob aspectos morfológicos de crescimento e produção em tutor vivo de gliricídia, comparativamente ao tutor morto, foi realizado um estudo de caso instalando um experimento em área de produtor em Baião - PA em DBC com cinco cultivares e cinco repetições considerando quatro plantas/cultivar. A altura das plantas, o diâmetro do caule e o número de ramos ortotrópicos foram tomados com 12 meses, assim como o comprimento do pecíolo, largura, comprimento e espessura de folhas adultas, como também o tamanho, peso e número de frutos/espiga, submetidos à ANOVA e teste de Tukey a 5%. A cultivar Kuthiravally apresentou maior média para Número de Ramos Ortotrópicos nos dois tipos de tutores. Bragantina, Clonada e Cingapura apresentaram maiores comprimentos do pecíolo em tutor vivo, e Bragantina e Uthirankotta apresentaram espigas de tamanho maiores quando cultivadas em gliricídia. Logo, há influência do tipo de tutor nos caracteres morfológicos de crescimento e produção de cultivares dessa espécie, com indicação da possibilidade do uso de gliricídia com sucesso no campo.
