Immobilized metal ion affinity chromatography (IMAC) has been widely used for the enrichment of phosphopeptides, whereas no report exists describing the use of IMAC columns for the enrichment of sulfopeptides. In this study, we used IMAC-Ga microcolumns for the enrichment of sulfopeptides from a complex mixture of peptides, extracted from skin secretions of the Pachymedusa dacnicolor frog. The enriched fraction obtained by IMAC-Ga was analyzed by liquid chromatograpy/electrospray ionization mass spectrometry (LC/ESI-MS) in an Orbitrap XL and by matrix-assisted laser desorption/ionization time-of-flight time-of-flight (MALDI-TOF/TOF) in an ABI 4800 instrument. From this fraction, different sulfated and non-sulfated peptides belonging to the caerulin and bradykinin families were structurally characterized. Other interesting negatively charged groups, such as phosphate adducts of dermaseptins and pyridoxal phosphate attached to a protease inhibitor, were also characterized. Unexpectedly, some dermaseptin antimicrobial peptides were also enriched by IMAC-Ga and a Sauvatine-like peptide was also fully sequenced. Furthermore, neutral loss of sulfated peptides and their fragmentation patterns in the gas phase were also compared using collision-induced dissociation (CID) and high-energy collision dissociation (HCD). Our present study provides evidence that IMAC-Ga enrichment is a fast, useful and promising method for high-throughput analysis of sulfated-peptides, since high-resolution mass spectrometers can be used for this purpose.
High-resolution mass spectrometry-based peptidomics has been used to characterize several components in electro-stimulated skin secretions of the endemic Mexican frog Pachymedusa dacnicolor. Peptide mass screening performed in an Orbitrap-XL mass spectrometer showed that P. dacnicolor skin secretions possess 194 different components with molecular masses ranging mainly from 500 to 6,000 Da. Dozens of molecules were partially sequenced including two novel protease inhibitors. Additionally, one posttranslationally modified bradykinin and two novel dermaseptin-like antimicrobial peptides were fully sequenced. The novel peptide named here DMS-DA5 was fully characterized and showed potent antibacterial activity against various bacteria such as Escherichia coli, Bacillus subtilis, Salmonella enterica serovar typhimurium, and Pseudomonas aeruginosa with minimal inhibitory concentrations from 3.10 to 25.0 microM.
Se estudió el ciclo de reproducción de Sceloporus ochoterenae en una población que vive en un relicto de selva baja caducifolia en el centro de Morelos. Los machos con longitud hocico-cloaca (LHC) promedio de 49.53 ± 2.40 mm, son más grandes y más pesados (x = 3.66 ± 0.68 g) que las hembras (LHC 46.37 ± 2.08 mm, peso 3.34 ± 1.23 g), lo que revela dimorfismo sexual como ocurre en otras especies de lagartijas sceloporinas. Existen diferencias en los ciclos de reproducción entre los sexos, los machos inician la actividad reproductora en marzo con la máxima actividad en mayo, y las hembras al inicio de la temporada de lluvias (mayo), con el periodo de mayor actividad durante la temporada de lluvias (julio). La temperatura y precipitación están correlacionadas positivamente con los ciclos de reproducción de machos y hembras. El tamaño promedio de la nidada, fue de 5 huevos y se relaciona positivamente con el tamaño de las hembras. En contraste, el tamaño y peso de las hembras no tuvieron efecto significativo sobre la masa relativa de la nidada (promedio = 0.245). En general, las características reproductoras de la población de Morelos son diferentes a las observadas en Guerrero.
El dimorfismo sexual, es un fenómeno común en lagartijas del género Sceloporus, lo que permite identificar diferencias en el tamaño de cuerpo, y diseño morfológico, entre machos y hembras. Lo más común es que ocurran variaciones en el color disociadas de la morfología. Utilizando ANDEVA de una vía y análisis discriminante, obtuvimos diferencias significativas en seis caracteres morfológicos en correlación con color de la garganta; lo que permite reconocer tres diferentes fenotipos (amarillo,azul y naranja). Sin embargo, no observamos diferencias en el uso de microhábitats. Los machos con garganta de color azul (44.35 %) fueron los más grandes y robustos, y tienen similar abundancia que los machos con garganta de color amarillo (48.5%). Los machos con garganta naranja solo representaron el 8 % del total. Sugerimos que los mecanismos de selección de machos por parte de las hembras y las oportunidades de realizar cópula, por parte de los tres morfos, pueden inducir variaciones en la inversión reproductora entre localidades. Sugerimos que las características de cada morfo, son un factor importante a considerar en estudios sobre inversión reproductoras de las especies.
Se estudió la composición de la dieta de una población de lagartijas de Sceloporus horridus horridus del centro de Morelos, México. Se analizó la diversidad del alimento, la variación entre las estaciones húmeda y seca del año, y el traslape de la dieta entre hembras y machos. Los datos fueron comparados con los de otras poblaciones de Jalisco y Puebla, México. La dieta estuvo formada por 18 grupos de insectos, donde destacan por su valor de importancia los coleópteros (69.3%), hemípteros (68.0%), formícidos (38.7%) y larvas (37.3%). Hubo variación en la frecuencia de ocurrencia y en el porcentaje de consumo de alimento entre los periodos de lluvias y secas. Un ANOVA para comparar la dieta entre hembras (H′ = 2.14) y machos (H′ = 2.08) mostró diferencias significativas entre los dos sexos. Se concluye que en lo general la dieta de la población estudiada es similar a la observada en otras poblaciones de esta especie.
