5-Azacytidine combined with 2,4-D increases the number of Acca sellowiana somatic embryos. Global DNA methylation is directly affected by these compounds.
Comparative analysis of zygotic and somatic embryogenesis of Acca sellowiana showed higher amounts of sucrose, fructose, raffinose, and myo-inositol in zygotic embryos at different developmental stages than in corresponding somatic ones. These differences were mostly constant. In general, glucose levels were significantly lower than the other soluble carbohydrates analyzed, showing minor variation in each embryo stage. Despite the presence of sucrose in the culture medium, its levels conspicuously diminished in somatic embryos compared with the zygotic ones. Raffinose enhanced parallel to embryo development, regardless of its zygotic or somatic origin. Analysis of the soluble carbohydrate composition of mature zygotic cotyledon used as explant pointed out fructose, glucose, myoinositol, sucrose, and raffinose as the most important. Similar composition was also found in the corresponding somatic cotyledon. Total soluble carbohydrates varied inversely, decreasing in zygotic embryos and increasing in somatic embryos until the 24th d, at which time they increased rapidly about sixfold in zygotic embryos until the 27th d, a period coinciding with the zygotic proembryos formation. Such condition seems to reflect directly the variation of endogenous sucrose level, mainly because glucose and fructose diminished continuously during this time period. This means that, in terms of soluble sugars, zygotic embryo formation occurred under a situation represented by high sucrose amounts, simultaneously with low fructose and glucose levels, while in contrast, somatic embryo formation took place under an endogenous sugar status characterized by a substantial fructose enhancement. Starch levels increased continuously in zygotic embryos and decreased in somatic ones, the reverse to what was found in fructose variation. Starch accumulation was significantly higher in somatic torpedo and cotyledonary embryos than in the corresponding zygotic ones.
No Laboratório de Fisiologia do Desenvolvimento e Genética Vegetal do CCA-UFSC, nos anos de 1993 a 1996, foram testadas 24 combinações de tratamentos envolvendo dois genótipos de abacaxizeiro (Ananas comosus (L.) Merr.), seis combinações dos fitorreguladores ácido naftalenoacético (ANA) e 6-benzilaminopurina (BAP), e os meios de cultura líquidos e geleificados, com o objetivo de estabelecer um protocolo regenerativo para a micropropagação do abacaxizeiro. A taxa de regeneração comportou-se de forma quadrática para a maioria das combinações testadas. O meio de cultura MS (Murashige & Skoog, 1962), líquido, adicionado de ANA (2,7 µM) e BAP (4,4 µM), proporcionou uma taxa média de regeneração de 19,7 brotos/explante. A magnitude do efeito genotípico exibido pelas cultivares utilizadas foi intermediária entre o efeito das combinações dos níveis de ANA e BAP (o maior) e o efeito da constituição física do meio de cultura. A melhor combinação de tratamentos foi testada em 17 acessos coletados no Estado de Santa Catarina, demonstrando-se a eficiência do protocolo regenerativo. A taxa média de regeneração foi de 15,3 brotos/explante, dos quais 40% apresentaram altura igual ou inferior a 3 cm. Brotos enraizados ou não, com altura igual ou superior a 3 cm, apresentaram valores médios de 95,5% de sobrevivência.
-(Anomalous somatic embryos in Acca sellowiana (O. Berg) Burret (Myrtaceae). Somatic embryogenesis represents a valuable tool for the studies on the basic aspects of plant embryo development. Today this process is used as a potencial technique for large-scale plant micropropagation although, so far, it has been applied to only a small number of species. However, when somatic embryos are malformed they are considered economically useless. In Acca sellowiana (O. Berg) Burret, an important fruit-producing crop, large amounts of anomalous somatic embryos (76.3%) were found just after 40 days of culture of explants in a 2,4-D containing medium. Among the anomalous forms found in the cotiledonary stage, 12.2% consisted of fused embryos, 40.4% displayed fused cotyledons, 13.0% presented supernumerary cotyledons, and 10.7% showed absence or poorly developed cotyledons, including those without the shoot apical meristem. Histological analyses indicated that the altered embryos were formed either directly from cotyledons, hypocotyl and radicle of the zygotic embryos used as explants, or indirectly from calli formed from these tissue parts. It is suggested that the formation of anomalous somatic embryos, as well as a low frequency of conversion into emblings reflect physiological and/or genetic disturbances triggered by the presence of 2,4-D in the medium. In vitro experimental alternative approaches are discussed in order to lessen the occurrence of malformed somatic embryos.Key words -2,4-D, feijoa, morphohistological analysis, somaclonal variation, somatic embryogenesis RESUMO -(Embriões somáticos anômalos em Acca sellowiana (O. Berg) Burret (Myrtaceae)). A embriogênese somática representa uma ferramenta importante para estudos básicos do desenvolvimento das plantas. Atualmente este processo é utilizado como uma técnica com potencial para a micropropagação de plantas em grande escala, embora ainda com aplicação restrita a poucas espécies. Entretanto, quando são formados embriões somáticos anômalos a aplicação econômica é inviabilizada. Em Acca sellowiana (O. Berg) Burret, uma importante frutífera nativa, uma elevada quantidade de embriões somáticos com fenótipos alterados (76,3%) foi encontrada já no 40 o dia de cultura na presença de 2,4-D. Entre esses, 12,2% consistiam de embriões fundidos, 40,4% apresentavam cotilédones concrescidos, 13% possuíam mais de dois cotilédones e 10,7% não tinham cotilédones ou eles eram pouco desenvolvidos, incluindo os embriões sem meristema apical caulinar. As análises histológicas mostraram que os embriões malformados originavam-se tanto diretamente dos cotilédones, hipocótilo e radícula dos embriões zigóticos utilizados como explantes, como indiretamente de calos formados a partir dessas estruturas. In general, plant material grown in vitro has low photosynthetic ability to achieve positive carbon balances. Therefore, a continuous supply of carbohydrates from the culture medium is required, and sucrose has been the most commonly used carbon source. In this paper, we investigate the effects of different sucrose concentrations and the presence and absence of light on the endogenous levels of soluble carbohydrates and starch as well as on the proliferation and growth of Dendrobium Second Love (Orchidaceae) in vitro. The possibility of using etiolated stem segments as a means for micropropagating this hybrid was also verified. The results obtained indicated that the presence and absence of light and the sucrose concentrations used influenced the amounts of soluble carbohydrates and starch and the proliferation of D. Second Love shoots and roots. An increase in sucrose concentration caused a progressive increase in the amounts of total carbohydrates and starch. Under both light conditions, sucrose was the main sugar found in the shoots followed by glucose and fructose. The addition of sucrose to the culture medium up to 2% and 4% was advantageous to the number of shoots produced per explant and the root longitudinal growth in the presence and absence of light, respectively. Shoot and root dry matter and the number of roots formed per explant increased as sucrose concentration was raised up to 6% in both light treatments. The use of dark-grown shoot segments proved to be a useful and reliable alternative for the micropropagation of this hybrid.
-(In vitro germination and growth of Cattleya bicolor Lindley (Orchidaceae)). The technique of germinating orchid seeds in vitro has been used since the beginning of the last century. Nevertheless, the available knowledge regarding the nutritional composition of culture media that favor in vitro germination and growth of orchids is still scarce. Cattleya bicolor is a Brazilian threatened orchid species and very little is known about its seed germination and growth in vitro. For that reason, this study aimed at analyzing the influence of the culture medium on the in vitro germination of Cattleya bicolor seeds, on the initial development of its protocorms until they form seedlings, and on seedling growth during the first twelve months of in vitro culture. The results revealed that the Vacin & Went is the ideal culture medium for seed germination and initial seedling development of Cattleya bicolor in vitro. It is recommended that after six months the seedlings be transferred to MS or VW media, since they promote seedling development. They should be kept in one of those media until acclimatization. These results are very important for the optimization of propagation techniques of Brazilian orchid species. A germinação de sementes de orquídeas in vitro vem sendo utilizada desde o início do século passado. Apesar disso, o conhecimento disponível a respeito da composição nutricional dos meios de cultura que favorecem a germinação e o crescimento in vitro de orquídeas ainda é bastante escasso. Diante da ameaça de extinção da Cattleya bicolor e devido à escassez de conhecimento a respeito da germinação e do crescimento in vitro dessa espécie, este trabalho teve como objetivo avaliar a influência do meio de cultura na germinação in vitro de suas sementes, no desenvolvimento inicial dos protocormos até a formação das plântulas, bem como avaliar o crescimento das plântulas até os primeiros doze meses de cultivo in vitro. Os resultados mostraram que o meio de cultura ideal para a germinação in vitro das sementes e para o desenvolvimento inicial das plântulas de Cattleya bicolor é o de Vacin & Went. Recomenda-se que após seis meses de cultivo as plântulas sejam transferidas para o meio MS ou VW, que promovem o desenvolvimento das plântulas, e mantê-las em um destes meios de cultura até o início da aclimatização. Tais informações são de grande importância para a otimização do processo de propagação das espécies de orquídeas nativas do Brasil. Germination, growth and development in vitro of orchids of the genus CattleyaOrchids are over-exploited in their natural environment due to their ornamental importance, which is leading some species to extinction. In vitro culture is an alternative for ex-situ conservation. Our objective was to determine an efficient medium for in vitro seed germination and initial seedling growth of Cattleya forbesii and Cattleya harrisoniana. Seeds of Cattleya forbesii were incubated in Murashige & Skoog (MS) medium (T1) and MS added with 2,5 g L -1 of activated charcoal (T2). For the last, plants with 1 ± 0,2 cm of height were submitted to the following treatments: T1, T2, MS reduced to the half strenght (T3) and MS reduced to the half strenght supplemented with 1,25 g L -1 of activated charcoal (T4). Thirty days after seed inoculation of C. forbesii, it was verified 45% and 90% of germination in T1 and T2, respectively. The addition of activated charcoal also promoted an increase in the height of the C. forbesii seedlings Germinação, crescimento e desenvolvimento in vitro de orquídeas (Cattleya spp., Orchidaceae)As orquídeas no ambiente natural sofrem exploração devido a sua importância ornamental, levando algumas espécies à extinção. O cultivo in vitro é uma forma alternativa para a conservação ex-situ. Procurou-se determinar um meio de cultura eficiente para a germinação in vitro de sementes e o crescimento inicial de plântulas de Cattleya forbesii, bem como para o crescimento de plântulas in vitro de Cattleya harrisoniana. No primeiro caso, sementes foram inoculadas em meio de cultura básico de Murashige & Skoog (MS) = T1 e MS básico acrescido de 2,5 g L -1 de carvão ativado (CA) = T2. No segundo, plântulas com 1 ± 0,2 cm de altura foram submetidas aos tratamentos T1, T2, MS com a metade da concentração original de macro-micronutrientes (T3) e MS com a metade da concentração original de macro-micronutrientes suplementado com 1,25 g L -1 de CA (T4). Verificou-se aos 30 dias em C. forbesii uma porcentagem de germinação de 45% em T1 e 90% em T2. A adição de CA ao meio de cultura trouxe aumento na altura de plântulas de C. forbesii de acordo com análises realizadas aos 180 dias de cultivo. Em relação ao crescimento de C. harrisoniana, aos 240 dias observou-se que todos os parâmetros médios avaliados (altura da parte aérea, massa de matéria fresca total, número de raízes e folhas, comprimento da maior raiz e diâmetro do pseudocaule) foram significativamente maiores em T2. Dessa forma, sugere-se o uso do meio MS acrescido de 2,5 g L -1 de CA (T2), uma vez que é significativamente favorável tanto para a germinação de sementes quanto para o crescimento de ambas as espécies.Palavras-chave: carvão ativado, meio de cultura, micropropagação, orquídeas.
