Resumo -O objetivo deste trabalho foi determinar as variáveis que influenciam a produção simultânea de celulases e xilanase por Aspergillus aculeatus URM 4953 e A. phoenicis URM 4924, com uso de bagaço de cana-de-açúcar como substrato. As variáveis avaliadas foram pH inicial, tempo de cultivo, concentração do substrato, agitação, concentração do inóculo, temperatura, tipo de bagaço de cana-de-açúcar e espécie de Aspergillus. Foi utilizado o planejamento fatorial fracionário 2 , com três repetições no ponto central. Todas as variáveis analisadas influenciaram a produção de pelo menos uma das enzimas. As maiores atividades enzimáticas observadas foram: celulases totais, 0,45 UI mL . As melhores condições para produção das enzimas foram: pH 6,0; tempo de cultivo de 168 horas; bagaço de cana-de-açúcar residual como substrato; e A. aculeatus URM 4953, que produziu as celulases e a xilanase simultaneamente. It was used the 2 8-4fractional factorial design, with three replicates in the central point. All analysed variables influenced at least one of the enzyme production. The highest enzymatic activities were: total cellulases, 0.45 UI mL . The best conditions for enzyme production were: pH 6.0; cultivation time of 168 hours; residual sugarcane bagasse as substrate; and A. aculeatus URM 4953, which produced simultaneously all the cellulases and xylanase. The variables that influence the simultaneous production of cellulases and xylanase are pH, cultivation time, sugarcane bagasse type, and Aspergillus species.
This study aimed to produce cellulases and xylanase from Aspergillus species in submerged cultivation using sugarcane bagasse as substrate. Nine strains of Aspergillus were obtained from the Coleção de Culturas Micoteca [University Recife Mycology (URM)] and were cultivated in medium containing raw and processed sugarcane bagasse (RSB and PSC, respectively) for 5 days at 30°C and 90 rpm. The production of enzymes was evaluated by determining the activity of total cellulase, endoglucanase, exoglucanase, cellobiase, and xylanase. RSB was the best inducer for total cellulase and xylanase whereas PSB was best for exoglucanase. The production of endoglucanase and cellobiase at higher levels was independent of the type of sugarcane bagasse used. The strains that produced cellulases and xylanase at the highest levels were A. phoenicis URM 4924 and A. aculeatus URM 4953 using RSB and A. carbonarius URM 1546 using PSB medium.
Objetivo: Realizar uma revisão de literatura com os principais teores referentes à infecção pelo vírus da febre amarela: registros históricos e epidemiológicos, virologia, transmissão, aspectos clínicos, diagnóstico, tratamento e profilaxia, associando-os aos avanços e retrocessos no combate à doença no Brasil. Metodologia: A revisão fundamentou-se em artigos das bases on line National Center for Biotechnology, (NCBI Pubmed) e Scientific Eletronic Library Online (SCIELO), entre 1928-2016 e nas plataformas governamentais BRASIL, Centers for Disease Control and Prevention (CDC) e World Health Organization ( WHO), entre 2010-2019. Resultados: os episódios de febre amarela (FA) em humanos no país, após 1942, sobretudo nos últimos anos 2017-2018, decorreram do ciclo silvestre e sugerem uma relação direta com degradação e tragédias ambientais. Também, episódios de FA silvestre em humanos, somados ao retorno do vetor urbano e falhas na cobertura vacinal nacional, inferem uma ameaça de reurbanização da doença. Considerações finais: Dada a importância da FA para a saúde pública no Brasil, faz-se necessário o implemento de ações imediatas e a longo prazo. Imediata, com a ampliação da cobertura vacinal gratuita nacional em quantidade suficiente para evitar surtos; e em longo prazo, a implementação de políticas públicas direcionadas a preservação ambiental e ao controle vetorial urbano.
