Fissiparous strains of freshwater triclads of the Dugesia gonocephala group were collected from 12 localities in Sardinia all situated not more than 5 kilometers from the coast. Some environmental factors and the sexual status of the specimens were noted at the time of collection. During the laboratory rearing 30% of individuals of each strain became sexual (ex-fissiparous individuals). All the examined strains showed common karyological and morphological characteristics suggestive of the species Dugesia sicula Lepori.The chromosome complement, which was a constant 27+2-3 B chromosomes, was classified as aneutriploid, due to its clearly documented characteristics. The fissiparous populations of D. sicula appear to have a high degree of tolerance to variations in environmental factors, especially temperature. Within the D. gonocephala group this species has the broadest distribution in the Mediterranean region.
Il DNA mitocondriale in alcuni generi di Protozoi (Kinetopla stida) Istituto Superiore di Sanità, Laboratorio di Parassitologia.All'ordine dei Kinetoplastida appartengono Protozoi per 10 piu parassiti,caratterizzati dalla presenza di un unico mitocondrio contenente una inusuale quantita di DNA extranuclearetkDNA contenuto nel kinetoplasto).Tale DNA e stato pajr ticolarmente studiato nei generi Trypanosoma,Leishmania,e Crithidia (Stuart,K.,1983).La sua struttura presenta una organizzazione del tutto peculiare,essendo simile a quella di una ma tassa costituita da centinaia di molecole circolari di DNA,di piccole dimensioni(l-2.5 kbp) e da altre molecole circolari piu grand! di DNA(20-4o kbp) poste alia periferia della maca£ sa stessa.Questa struttura e stata studiata con metodi molec£ lari per comprenderne il significato biologico. L l uso di tecniche di ingegneria genetica ha permesso di caratterizzare e sequenziare alcuni di questi circoletti iden tificando la funzione codificante di alcuni tratti dei circo-11 grand! (maxi-circles) e particolari omologie e differenze di sequenze nei circdli piccoli (mini-circles).II kDNA ha fornito comunque un prezioso strumento di indagine, non solo dal punto di vista strettamente molecolare, ma anche come contributo ad una identificazione tassonomica di protozoi parassiti morfologicamente indistinguibili.Come esempio delle tecniche di analisi del kDNA per fini tassonomici vengono qui riportati alcuni risultati di una indaglne effettuata su stocks e ceppi di riferimento di Leishmania isolati da casi singoli di leishmaniosi viscerale e cutanea da mammiferi serbatoi della malattia.Dopo aver digerito il kDNA con endonucleasi di restrizic ne e stata osservata la migrazione elettroforetica dei frammenti ottenuti.I dati ottenuti sono stati confrontati con que! 11 ricavati dalla analisi degli isoenzimi (Gramiccia et al., 1985) e messi in relazione con alcune caratteri estrinseci del parassita,quali patologia nell'ospite,comportamento in natura e .distribuzione geografica.F. Abbiamo isolato tre di tali famiglie, caratterizzato due di esse, e, preliminarmente, la terza,dalla s.specie vulgaris meridionalis.
