Background: In rats, esophagogastroduodenal anastomosis (EGDA) without concomitant chemical carcinogen treatment leads to gastroesophageal reflux disease, multilayered epithelium (MLE, a presumed precursor in intestinal metaplasia), columnar-lined esophagus, dysplasia, and esophageal adenocarcinoma. Previously we have shown that columnar-lined esophagus in EGDA rats resembled human Barrett's esophagus (BE) in its morphology, mucin features and expression of differentiation markers (Lab. Invest. 2004;84:753-765). The purpose of this study was to compare the phenotype of rat MLE with human MLE, in order to gain insight into the nature of MLE and its potential role in the development of BE.
Background: Berberine, a well-known component of the Chinese herbal medicine Huanglian, has wide range of biochemical and pharmacological effects, including antineoplastic effect, but the exact mechanisms remain unclear. The aim of the present study was to evaluate the potential chemo-sensitization effect of berberine in ovarian cancer cell line A2780. Methods: The expression of miR-93 was measure by RT-PCR. The target of miR-93 was confirmed by luciferase activity assay. Hoechst 33258 staining, Annexin V and PI double staining were used for apoptosis analysis. Results: In this study, we found A2780/DDP cells that were incubated with berberine combined with cisplatin had a significantly lower survival than the control group. Berberine enhanced cisplatin induced apoptosis and induced G0/G1 cell cycle arrest in A2780 cells. Next, we observed that the miR-93 levels in cisplatin resistant cell lines were higher than that in cisplatin sensitive cell lines. Furthermore, our study found berberine could inhibit miR-93 expression and function in ovarian cancer, as shown by an increase of its target PTEN, an important tumor suppressor in ovarian cancer. A2780 cells that were treated with PTEN siRNA had increased survival compared to NC group and this could be partly alleviated by the AKT inhibitor Triciribine. More importantly, A2780 cells that were treated with PTEN siRNA had a survival pattern that is similar to cells with miR-93 overexpression. Conclusion: The results suggested that berberine modulated the sensitivity of cisplatin through miR-93/PTEN/AKT signaling pathway in the ovarian cancer cells.
Malignant glioma is the most common type of primary brain tumor in adults, characterized by rapid tumor growth and infiltration of tumor cells throughout the brain. Alterations in the activity of the 26S proteasome have been associated with malignant glioma cells, although the specific defects have not been identified. Recently, microRNA-326 (miR-326) was shown to play an important role in glioblastoma and breast cancer, but the underlying molecular mechanisms remain unclear. In the present study, the human Nin one binding protein (NOB1) was identified as a direct target of miR-326 and a potential oncogene in human glioma. Similar to NOB1 silencing by shRNA, overexpression of miR-326 in human glioma cell lines (A172 and U373) caused cell cycle arrest at the G1 phase, delayed cell proliferation and enhanced apoptosis. MiR-326 inhibited colony formation in soft agar and decreased growth of a xenograft tumor model, suggesting that miR-326 and NOB1 are required for tumorigenesis in vitro and in vivo. Furthermore, these processes were shown to involve the MAPK pathway. NOB1 overexpression in human glioma samples was detected by Affymetrix array analysis, and NOB1 mRNA and protein levels were shown to be increased in high-grade glioma compared to low-grade glioma and normal brain tissue. Furthermore, high levels of NOB1 were associated with unfavorable prognosis of glioma patients. Taken together, these results indicate that miR-326 and NOB1 may play an important role in the development of glioma.
Quaking (QKI) is an alternative splicing factor that can regulate circRNA formation in the progression of epithelialmesenchymal transition, but the mechanism remains unclear. High expression of QKI is correlated with short survival time, metastasis, and high clinical stage and pathology grade in hepatocellular carcinoma (HCC). Here we report that transcription of the QKI gene was activated by the Yin-Yang 1 (YY1)/p65/p300 complex, in which YY1 bound to the superenhancer and promoter of QKI, p65 combined with the promoter, and p300 served as a mediator to maintain the stability of the complex. This YY1/p65/p300 complex increased QKI expression to promote the malignancy of HCC as well as an increased circRNA formation in vitro and in vivo. Hyperoside is one of several plant-derived flavonol glycoside compounds. Through virtual screening and antitumor activity analysis, we found that hyperoside inhibited QKI expression by targeting the YY1/p65/p300 complex. Overall, our study suggests that the regulatory mechanism of QKI depends on the YY1/p65/p300 complex and that it may serve as a potential target for treatment of HCC. Significance: These findings identify the YY1/p65/p300 complex as a regulator of QKI expression, identifying several potential therapeutic targets for the treatment of HCC.
The effects of different concentrations (1-50 microM) of Cd on root growth, cell division and nucleoli in root tip cells, protective enzyme activities and lipid peroxidation in Vicia faba were investigated in order to better understand the processes of Cd-induced senescence. The results indicated that lower concentration of Cd (1 microM) had no obviously influence on the root growth during 24-48 h treatment, but higher concentrations (5-50 microM) inhibited significantly after 48 and 72 h. The mitotic index decreased with increasing of Cd concentration and duration of treatment except for the group exposed to 1 microM Cd. Cd induced c-mitosis, chromosome bridges, chromosome stickiness and lagging chromosomes. The rate of aberrant dividing cells increased with prolonging duration of treatment and increasing of Cd concentration. On nucleolus, some particulates containing the argyrophilic proteins were extruded from the nucleus into the cytoplasm in the cells stressed by Cd and some were scattered in the nucleus. After the treatment with Cd (10 microM Cd, 48 h), the nucleolus did not disaggregate normally and still remain its characteristic structure during metaphase and the particles of similar silver-stained materials were localized on chromosomes. In leaves, Catalase (CAT) activity declined but Peroxidase (POD) activity increased with increasing of the duration of treatment. In roots, CAT activity increased with increasing of the duration of treatment, POD activity increased during early days and then declined. Superoxide dismutase (SOD) activity showed an upward trend with increasing of the duration of treatment after 3 and 6 days, then declined both in leaves and roots (9 days). SOD and POD had highest activities at 50 microM Cd in leaves. CAT activity was lowest at 50 microM Cd. Malondialdehyde (MDA) content increased with the increasing of Cd concentrations and duration of treatment in leaves. In roots, MDA content showed an upward trend with increasing of the duration of treatment at early time and then declined.
MVD may be one of the important prognostic factors for gastric cancer patients. COX-2 and VEGF may play an important role in tumor progression by stimulating angiogenesis. VEGF might play a main role in the COX-2 angiogenic pathway. The inhibition of angiogenesis or COX-2, VEGF activity may have an important therapeutic benefit in the control of gastric cancer.
Purpose As the premalignant lesion of human esophageal adenocarcinoma (EAC), Barrett’s esophagus (BE) is characterized by intestinal metaplasia in the normal esophagus (NE). Gene expression profiling may help us understand the potential molecular mechanism of human BE. Methods We analyzed three microarray datasets (two cDNA arrays and one oligonucleotide array) and one SAGE dataset with SAM and SAGE(Poisson) to identify individual genes differentially expressed in BE. GSEA was used to identify a priori defined sets of genes that were differentially expressed. These gene sets were either grouped according to certain signaling pathways (GSEA curated), or the presence of consensus binding sequences of known transcription factors (GSEA motif). Immunohistochemical staining (IHC) was used to validate differential gene expression. Results Both SAM and SAGE(Poisson) identified 68 differentially expressed genes (55 BE genes and 13 NE genes) with an arbitrary cutoff ratio (≥4 fold). With IHC on matched pairs of NE and BE tissues from 6 patients, these genes were grouped into 6 categories: Category I (25 genes only expressed in BE), Category II (5 genes only expressed in NE), Category III (8 genes expressed more in BE than in NE), and Category IV (2 genes expressed more in NE than in BE). Differential expression of the remaining genes was not confirmed by IHC either due to false discovery (Category V), or lack of proper antibodies (Category VI). Besides individual genes, the TGFβ pathway and several transcription factors (CDX2, HNF1, and HNF4) were identified by GSEA as enriched pathways and motifs in BE. Apart from 9 target genes known to be up-regulated in BE, IHC staining confirmed up-regulation of 19 additional CDX1 and CDX2 target genes in BE. Conclusion Our data suggested an important role of CDX1 and CDX2 in the development of BE. The IHC-confirmed gene list may lead to future studies on the molecular mechanism of BE.
