O teor de água das sementes é fator de extrema importância na padronização do método do teste de condutividade elétrica, visto que influencia significativamente os resultados. O objetivo deste trabalho foi estudar o efeito do teor de água inicial sobre os resultados da condutividade elétrica das sementes de soja, e determinar uma equação capaz de corrigir a sua influência. Foram utilizadas seis cultivares de soja, em dois anos agrícolas, cada uma representada por lotes com três níveis de vigor. Foi ajustado o teor de água das sementes de cada lote para 7, 9, 11, 13, 15 e 17%, antes da condução do teste de condutividade elétrica; a equação de correção para ajuste dos dados foi determinada com base em regressões, com os resultados dos dois anos agrícolas, cultivares e níveis de vigor. O teor de água inicial das sementes de soja pode influenciar a interpretação dos resultados da condutividade elétrica e a equação de correção é eficiente para eliminar o efeito do teor de água sobre os resultados do teste.

Although drought and defoliation stress have been shown to reduce soybean [Glycine max (L.). Merr.] yield, little information has been published regarding their effects on soybean seed quality. Field experiments were conducted in 1986, 1987, and 1989 to evaluate the effect of drought and defoliation (1989 only) stress during soybean seed development on seed germination and vigor. Essex (MG [maturity group] V) and Union (MG III) were grown in 1986 and 1987, and Harper (MG III) and McCall (MG 00) in 1989. Moisture treatments were either well watered or drought stressed during seed development (R5 to R7). In 1989, a total defoliation treatment was also imposed at R6 as an additional stress factor. There were significant reductions in yield and yield components following drought stress in all 3 yr and following defoliation in 1989. Leaf conductance and transpiration also decreased in the drought stress treatments. There was no effect of drought stress on seed germination or seed vigor as measured by accelerated aging germination and the cold test across the four cultivars (determinate and indeterminate) and 3 yr. In 1989 slight changes in 3‐d germination and conductivity occurred for some drought stress treatments. Most of this response, however, was related to increased occurrence of hard seed, which does not represent an indication of a change in vigor. Seed germination and vigor were significantly reduced for small, flat, shriveled, and underdeveloped seeds that only occurred following defoliation. These seeds represented a small portion of the seed lot that would normally be removed during conditioning. The data suggest that drought stress would have no effect on seed germination or vigor, unless the stress was severe enough to produce shriveled, flat, underdeveloped seeds.

Considering the zoonotic potential of tick-borne disease (TBD) agents and the fact that dogs may act as sentinels for human infection, the aim of the present study was to determine the seroprevalence of TBD agents and risk factors for exposure in two different canine populations from Parana State, Southern Brazil. A total of 138 dog serum samples from urban (UA) (n=68) and rural (RA) (n=70) areas were tested with commercial ELISA rapid test for Anaplasma phagocytophilum, Ehrlichia canis and Borrelia burgdorferi antibodies and indirect immunofluorescence assay (IFAT) for Babesia vogeli. An overall of 92/138 (66.7%) dogs, being 62/68 (91.2%) from UA and 30/70 (42.9%) from RA, were seropositive for at least one TBD agent. From the total number of dogs, sixty-two were positive for E. canis (44.9%), 19 (13.8%) for A. phagocytophilum, and 64 (46.4%) for B. vogeli. Anti-B. burgdorferi antibodies were not detected. Dogs from UA showed a higher percentage of tick infestation (p = 0.0135) and were highly associated with seropositivity to E. canis (p = 0.000005), A. phagocytophilum (p = 0.0001), and B. vogeli (p = 0.0012). In summary, the findings indicate that dogs from urban areas present higher potential risk exposure to TBD pathogens than those from rural areas.Keywords: Ehrlichia canis, Babesia vogeli, Anaplasma phagocytophilum, Borrelia burgdorferi, serology, Parana. ResumoConsiderando o potencial zoonótico das doenças transmitidas por carrapatos (DTCs) e que os cães podem atuar como sentinelas para infecções em humanos, os objetivos deste estudo foram determinar a soroprevalência de agentes das DTCs e fatores de risco para a exposição em duas diferentes populações caninas do Estado do Paraná, região Sul do Brasil. Um total de 138 amostras de soro de cães de área urbana (AU) (n = 68) e rural (AR) (n = 70) foram testadas utilizando um teste de ELISA comercial rápido para detecção de anticorpos contra Anaplasma phagocytophilum, Ehrlichia canis e Borrelia burgdorferi e imunofluorescência indireta (IFI) para Babesia vogeli. Um total de 92/138 (66,7%) cães, sendo 62/68 (91,2%) da AU e 30/70 (42,9%) da AR, foram soropositivos para pelo menos um agente. Do número total de amostras, sessenta e duas (44,9%) foram positivas para E. canis, 19 (13,8%) para A. phagocytophilum e 64 (46,4%) para B. canis vogeli. Anticorpos anti-B. burgdorferi não foram detectados. Os cães da AU apresentaram o maior percentual de infestação por carrapatos (p = 0,0135) e foram altamente associados com a positividade para E. canis (p = 0,000005), A. phagocytophilum (p = 0,0001) e B. vogeli (p = 0,0012). Em resumo, nossos achados indicam que cães de áreas urbanas têm um maior risco potencial de exposição a agentes patogênicos das DTCs comparados aos das áreas rurais.

Research on soybean seed has revealed that conductivity test results may be influenced by storage temperature and that an apparent direct relationship between deterioration at low temperatures and loss of cellular membrane integrity is not evident. The objective of this study was to verify the influence of low storage temperatures on electrical conductivity results in other species, such as seeds of two pea cultivars studied temperatures were: 10, 20, 25, 20/10 and 25/10ºC. The physiological potential of the seeds was evaluated every three months during an 18 month storage period using germination, accelerated aging and electrical conductivity tests. The electrical conductivity test was not a good indicator for the degree of seed deterioration stored at low temperatures. Seed deterioration at 10ºC did not seem to be directly related to the loss of membrane integrity, possibly due to membrane repair or reorganization during storage at this temperature. O presente estudo foi conduzido com o objetivo de verificar se a influência de baixas temperaturas de armazenamento nos resultados da condutividade elétrica ocorre também para outra espécie, sendo utilizadas para tanto, sementes de dois cultivares de ervilha. O efeito das seguintes temperaturas foi estudado: 10, 20, 25, 20/10 e 25/10ºC. A cada três meses, num total de 18 meses de armazenamento, foi avaliado o potencial fisiológico das sementes usando-se os testes de germinação, envelhecimento acelerado e condutividade elétrica. O teste de condutividade elétrica não se mostrou um bom indicador da intensidade do processo de deterioração de sementes armazenadas a baixas temperaturas. A deterioração das sementes a 10ºC parece não estar relacionada diretamente com a perda da integridade das membranas, possivelmente devido ao reparo ou à reorganização das mesmas durante o período de armazenamento nesta temperatura. -The objective of this work was to evaluate the effects of temperature (10, 20, 30, 20/10 and 30/10ºC) and period of storage on electrical conductivity (EC) in four seed lots of corn (Zea mays L.), as well as the mineral composition of the soaking solution. EC test determines indirectly the integrity of seed membrane systems, and is used for the assessment of seed vigor, because this test detects the seed deterioration process since its early phase. The research comprised determinations of water content, germination, accelerated aging (AA), cold (CT) and EC vigor tests, and determinations of Ca 2+ , Mg 2+ and K + release to the solution, after seed soaking of four corn seed lots. The evaluations were performed each four months during a period of 16 months. For statistical analysis, a completely randomized split plot design was used with eight replications. Except for seed lots stored at 10ºC, all vigor evaluations revealed a decline in vigor, but AA and CT showed more sensitiveness to declines of seed physiological quality than EC. Potassium was the main leached ion regardless of the storage temperature.

Vigor of soybean [Glycine max (L.) Merrill] seeds can be evaluated by measuring the electrical conductivity (EC) of the seed soaking solution, which has shown a satisfactory relationship with field seedling emergence, but has not had aproper definition of range yet. This work studies the relationship between EC and soybean seedling emergence both in the field and laboratory conditions, using twenty two seed lots. Seed water content, standard germination and vigor (EC, accelerated aging and cold tests) were evaluated under laboratory conditions using -0.03; -0.20; -0.40 and -0.60 MPa matric potentials, and field seedling emergence was also observed. There was direct relationship between EC and field seedling emergence (FE). Under laboratory conditions, a decreasing relationship was found between EC and FE as water content in the substrate decreased. Relationships between these two parameters were also found when -0.03; -0.20 and -0.40 MPa matric potentials were used. EC tests can be used successfully to evaluate soybean seed vigor and identify lots with higher or lower field emergence potential.

SUMMARYThe aims of this study were to determine the seroprevalence of Ehrlichia spp. and risk factors for exposure in a restricted population of dogs, horses, and humans highly exposed to tick bites in a Brazilian rural settlement using a commercial ELISA rapid test and two indirect immunofluorescent assays (IFA) with E. canis and E. chaffeensis crude antigens. Serum samples from 132 dogs, 16 horses and 100 humans were used. Fifty-six out of 132 (42.4%) dogs were seropositive for E. canis. Dogs > one year were more likely to be seropositive for E. canis than dogs ≤ one year (p = 0.0051). Ten/16 (62.5%) and 8/16 (50%) horses were seropositive by the commercial ELISA and IFA, respectively. Five out of 100 (5%) humans were seropositive for E. canis and E. chaffeensis. Rhipicephalus sanguineus (n = 291, 97.98%) on dogs and Amblyomma cajennense (n = 25, 96.15%) on horses were the most common ticks found. In conclusion, anti-Ehrlichia spp. antibodies were found in horses; however, the lack of a molecular characterization precludes any conclusion regarding the agent involved. Additionally, the higher seroprevalence of E. canis in dogs and the evidence of anti-Ehrlichia spp. antibodies in humans suggest that human cases of ehrlichiosis in Brazil might be caused by E. canis, or other closely related species.

ELECTRICAL CONDUCTIVITY TEST OF SOYBEAN SEEDS STORED UNDER DIFFERENT TEMPERATURESThere are some results showing that the electrical conductivity test can be influenced by low temperature of seed storage, such as 10 o C. This work was carried out in order to study the effect of temperature and period of storage on electrical conductivity and chemical composition of the imbibing solution of soybean seeds. For that, seed water content, germination, vigor (accelerated aging, cold test and electrical conductivity), and chemical composition (K + , Ca 2+ and Mg 2+ ) of the imbibing solution were determined. Two soybean seed lots of cultivar FT-20 with high and low vigor were used. The evaluations were performed at every three months intervals, during 15 months. Both temperature and storage period influenced seed vigor, as well as the ions leakage. It can be concluded that the electrical conductivity test is not suitable to evaluate vigor of soybean seeds stored at low temperature, and potassium is the major ion leakage regardless of the storage temperature.
