Ecological and genetic studies of marine turtles generally support the hypothesis of natal homing, but leave open the question of the geographical scale of genetic exchange and the capacity of turtles to shift breeding sites. Here we combine analyses of mitochondrial DNA (mtDNA) variation and recapture data to assess the geographical scale of individual breeding populations and the distribution of such populations through Australasia. We conducted multiscale assessments of mtDNA variation among 714 samples from 27 green turtle rookeries and of adult female dispersal among nesting sites in eastern Australia. Many of these rookeries are on shelves that were flooded by rising sea levels less than 10 000 years (c. 450 generations) ago. Analyses of sequence variation among the mtDNA control region revealed 25 haplotypes, and their frequency distributions indicated 17 genetically distinct breeding stocks (Management Units) consisting either of individual rookeries or groups of rookeries in general that are separated by more than 500 km. The population structure inferred from mtDNA was consistent with the scale of movements observed in long-term mark-recapture studies of east Australian rookeries. Phylogenetic analysis of the haplotypes revealed five clades with significant partitioning of sequence diversity (Phi = 68.4) between Pacific Ocean and Southeast Asian/Indian Ocean rookeries. Isolation by distance was indicated for rookeries separated by up to 2000 km but explained only 12% of the genetic structure. The emerging general picture is one of dynamic population structure influenced by the capacity of females to relocate among proximal breeding sites, although this may be conditional on large population sizes as existed historically across this region.

Strip-transect aerial surveys of Shark Bay, Ningaloo Reef and Exmouth Gulf were conducted during the winters of 1989 and 1994. These surveys were designed primarily to estimate the abundance and distribution of dugongs, although they also allowed sea turtles and dolphins, and, to a lesser extent, whales, manta rays and whale sharks to be surveyed. Shark Bay contains a large population of dugongs that is of international significance. Estimates of approximately 10000 dugongs resulted from both surveys. The density of dugongs is the highest recorded in Australia and the Middle East, where these surveys have been conducted. Exmouth Gulf and Ningaloo Reef are also important dugong habitats, each supporting in the order of 1000 dugongs. The estimated number of turtles in Shark Bay is comparable to the number in Exmouth Gulf plus Ningaloo Reef (7000–9000). The density of turtles in Ningaloo Reef and, to a lesser extent, Exmouth Gulf is exceptionally high compared with most other areas that have been surveyed by the same technique. Shark Bay supports a substantial population of bottlenose dolphins (2000–3000 minimum estimate). Exmouth Gulf and Ningaloo Reef were not significant habitats for dolphins during the winter surveys. Substantial numbers of whales (primarily humpbacks) and manta rays occur in northern and western Shark Bay in winter. Ningaloo Reef is an important area for whale sharks and manta rays in autumn and winter. The Shark Bay Marine Park excludes much of the winter habitats of the large vertebrate fauna of Shark Bay. In 1989 and 1994, more than half of all the dugongs were seen outside the Marine Park (57·4 and 50·7%, respectively). Approximately one-third to one-half of turtles and dolphins were seen outside the Marine Park (in 1989 and 1994 respectively: turtles, 43 and 27%; dolphins, 47 and 32%). Almost all the whales and most of the manta rays were seen outside the Marine Park. Expansion of the Shark Bay Marine Park, to bring it into alignment with the marine section of the Shark Bay World Heritage Area, would facilitate the appropriate management of these populations. This would also simplify the State– Commonwealth collaboration necessary to meet the obligations of World Heritage listing. The coastal waters of Western Australia north of the surveyed area (over 6000 km of coastline) are relatively poorly known and surveys of their marine megafauna are required for wise planning and management.

Hawksbill turtle (Eretmochelys imbricata) populations have experienced global decline because of a history of intense commercial exploitation for shell and stuffed taxidermied whole animals, and harvest for eggs and meat. Improved understanding of genetic diversity and phylogeography is needed to aid conservation. In this study, we analyzed the most geographically comprehensive sample of hawksbill turtles from the Indo-Pacific Ocean, sequencing 766 bp of the mitochondrial control region from 13 locations (plus Aldabra, n = 4) spanning over 13500 km. Our analysis of 492 samples revealed 52 haplotypes distributed in 5 divergent clades. Diversification times differed between the Indo-Pacific and Atlantic lineages and appear to be related to the sea-level changes that occurred during the Last Glacial Maximum. We found signals of demographic expansion only for turtles from the Persian Gulf region, which can be tied to a more recent colonization event. Our analyses revealed evidence of transoceanic migration, including connections between feeding grounds from the Atlantic Ocean and Indo-Pacific rookeries. Hawksbill turtles appear to have a complex pattern of phylogeography, showing a weak isolation by distance and evidence of multiple colonization events. Our novel dataset will allow mixed-stock analyses of hawksbill turtle feeding grounds in the Indo-Pacific by providing baseline data needed for conservation efforts in the region. Eight management units are proposed in our study for the Indo-Pacific region that can be incorporated in conservation plans of this critically endangered species.

In Australia, the olive ridley sea turtle Lepidochelys olivacea has received little research attention and monitoring. The Australian populations are relatively small and their distribution is limited to remote areas in the northern part of the country. Previous global genetic studies of olive ridley populations showed that the Australian breeding population at the McCluer Group of islands, Northern Territory, is genetically distinct from other olive ridley populations breeding in the Indo-Pacific. However, nothing is known about the genetic stock structure among Australian olive ridley rookeries found across northern Australia. High predation of eggs by feral pigs, dogs and monitor lizards Varanus spp. is believed to have severely impacted the number of nesting females at some rookeries. Of particular concern is the small nesting population on the western Cape York Peninsula, and without immediate conservation action this population could face extinction. The results presented here establish that there are at least 2 independent management units (stocks) of olive ridley turtles nesting in Australia and emphasise the importance of conserving the genetically distinct small breeding population nesting along the western Cape York Peninsula. In addition, results from 44 turtles caught in ghost nets across the Gulf of Carpentaria revealed that 45% of the haplotypes (32% of all ghost net samples) had not been observed at any rookery in Australia or SE Asia. This research highlights the need for better information on olive ridley population structure in the region and for urgent conservation action for the western Cape York population.

The hawksbill turtle, Eretmochelys imbricata, is endangered and currently declining in many parts of its global distribution. Efforts to manage the species are hampered by the lack of knowledge of the appropriate geographic units of management and the relationship among breeding populations and feeding assemblages. The Australian populations are among the few remaining large assemblages in the world, but may be under threat from harvesting in neighbouring countries. We use patterns of mitochondrial DNA variation to determine the geographic scale of breeding populations and to compare the genetic composition of turtles in feeding populations to those nesting at nearby major rookeries. Four of the major Australian rookeries were sampled, two on the western coast and two in northeastern Australia, as were turtles at two foraging sites. Significant differences in the frequency of two divergent mitochondrial DNA types separated the turtles nesting at major rookeries sampled on the northeastern versus western coasts of Australia, demonstrating that these populations are not connected by significant amounts of gene flow and should be considered as separate entities for management. There was no significant difference between the turtles nesting at two western rookeries 100 km apart, nor between those using the two northeastern rookeries separated by 750 km. This indicates that the size of the interbreeding unit for the hawksbill turtle is likely to be a region consisting of a group of islands rather than an individual island. Feeding populations were screened with a gene amplification test that discriminates between the two major DNA types. In each case, there was a significant difference in allele frequency between feeding populations and the nearest major rookery. This, together with previous reports of long distance migrations from tag returns, suggests that individual foraging areas support hawksbill turtles from distant breeding populations.

Infections caused by salmonellae, E coli and other Gram-negative bacteria should be considered as causes of systemic illness and death in wild green sea turtles infected with spirorchid cardiovascular flukes and other internal parasites.

Large vagile mammals typically exhibit little genetic structuring across their range, particularly when their habitat is essentially continuous. We investigated the population genetic structure of a large vagile Australian macropodid, Macropus fuliginosus, which is continuously distributed across most of southern Australia, using nine highly polymorphic nuclear microsatellite loci. Five distinct genetic units were identified across the range, four on the mainland and one on Kangaroo Island. In addition to the predicted historic Nullarbor Plain Barrier, two unexpected mainland barriers to gene flow were identified. Both were associated with landscape discontinuities (Swan River, Flinders Ranges), which appear within the dispersal capabilities of M. fuliginosus. Typical of large vagile mammals, M. fuliginosus displays high genetic diversity (with the exception of an insular population) and weak genetic structuring (within genetic units). However, the expansion of M. fuliginosus from southwestern Australia during the Pleistocene has resulted in significantly reduced genetic diversity in eastern populations. No significant sex-biased dispersal was detected, although differences in habitat, densities and climatic conditions between the eastern and western regions of the range appear to influence dispersal with the effects of isolation by distance only evident in the west. These results suggest that the biogeography of southern Australia is more complex than previously thought and reveal that seemingly minor landscape features can significantly impact genetic structuring in large vagile mammals.
