Das Dinitril 8a rnit dem Gerust des 2-[(E)-2-(2-Indolyl)vinyl]indols wird auf verschiedenen Wegen synthetisiert; in einfachcr Vcrfahrcnsweise werden Syntheses of Antileukemic (Indolylvinyl)indolesThe dinitrile Die uberlegene antileukiimische Wirksamkeit der biskationischen Vertreter des Typs 1 rnit der Kombination X = NH, Y = 0 gegenuber jenen rnit X,Y = 04', bedingt durch eine spezifischere und stiirkere AuBenbindung der Indolverbindungen an Desoxyribonucleinsauren mit gehauften Adenin-Thymin-Basenpaaren (AT-reiche DNA)5), veranlaDte uns, die neue Kombination X,Y = NH zu synthetisieren.Das Dinitril 8a erhielten wir erstmals'), als der Indolringschlufi rnit Triethylphosphit nach C u d o g~n~~~) auf das Trien 6 und das Dien 7a angewandt wurde, zu deren Darstellung die Wittig-Reaktion diente. Nach den Erfahrungen der Polyensynthesen8' boten sich hierfur die Komponenten 2a9), 2b9), 2d, 2e, 2h und 3a'), 3 b an.Bei der Reaktionsfolge 2d+2f-+2g42h verhinderte nur eine ungewohnlich kurze Hydrierung von 2d rnit NaBH4 zu 2f eine weitergehende Hydrierung der Seitenkette. Eine selektive Hydrierung des Esters 2c, welcher durch Umsetzung van Methoxycarbonylmethylentriphenylphosphoran'") rnit 2a erhalten worden war, gelang nicht. Hochst brauchbar erwies sich Formylmethylentriphenylphosphoran '') sowohl fur die erfreulich glatt verlaufende, doppelte Aldehydverliingerung 2a + 2e, als auch fur die einfachen 2a -+ 2d, 3a -+ 3b und 4a -+ 4b. Reaction of corresponding aldehydes (Ia) or (Id) with phosphonium bromides (Ic) or (If) (prepared from (Ia), (Id) via (Ib), (Ie)) according to literature procedures yields the title compounds (IIa)‐(IIc) from which the derived antileukemic imidazolinyl compounds (IId)‐(IIf) are prepared by known methods.
