Every year about 3 million tourists from around the world visit Brazil, Argentina and Paraguay´s triple border region where the Iguaçu Falls are located. Unfortunately, in recent years an increasing number of autochthonous canine and human visceral leishmaniasis (VL) cases have been reported. The parasite is Leishmania (Leishmania) infantum and it is transmitted by sand flies (Phlebotominae). To assess the risk factors favorable for the establishment and spread of potential vectors the Centers for Disease Control and Prevention light trap (CDC-light trap) collections were made in the Foz do Iguaçu (FI) and Santa Terezinha de Itaipu (STI) townships and along two transects between them. Our study determined the Phlebotominae fauna, the factors that affect the presence and abundance of Lutzomyia longipalpis and Nyssomyia whitmani, the presence of L. infantum in different sand fly species and which Leishmania species are present in this region. Lutzomyia longipalpis was the prevalent species and its distribution was related to the abundance of dogs. Leishmania infantum was found in Lu. longipalpis, Ny. whitmani, Ny. neivai and a Lutzomyia sp. All the results are discussed within the Stockholm Paradigm and focus on their importance in the elaboration of public health policies in international border areas. This region has all the properties of stable VL endemic foci that can serve as a source of the disease for neighboring municipalities, states and countries. Most of the urban areas of tropical America are propitious for Lu. longipalpis establishment and have large dog populations. Pan American Health Organization´s initiative in supporting the public health policies in the border areas of this study is crucial and laudable. However, if stakeholders do not act quickly in controlling VL in this region, the scenario will inevitable become worse. Moreover, L. (Viannia) braziliensis found in this study supports the need to develop public health policies to avoid the spread of cutaneous leishmaniasis. The consequences of socioeconomic attributes, boundaries and frontiers on the spread of diseases cannot be neglected. For an efficient control, it is essential that urban planning is articulated with the neighboring cities.

Aim Sea‐level changes and coastal geomorphology (extension of continental shelves) have affected the pattern of diversification of brackish/freshwater fish populations due to the confluence of basins/estuaries during low sea‐level periods (opportunity) and isolation during high sea‐level periods. It is hypothesized that populations from estuaries in extensive continental shelves, which have greater potential for confluence, should have more opportunities for gene exchange than those in narrow continental shelves. To test this, we assessed the distribution of the genetic diversity and the patterns of diversification of the estuarine silverside, Atherinella brasiliensis, from the Brazilian coast. Location Eight estuaries along the southern and north‐eastern portions of the Brazilian coast. Materials and methods Sequences of the mitochondrial control region (D‐loop) and the cytochrome B were used to assess the distribution of genetic diversity, and these mitochondrial fragments and two nuclear introns were used to assess the patterns of diversification of A. brasiliensis. The palaeochannels that were exposed along the Brazilian coast during the Last Glacial Maximum were reconstructed using bathymetry data and the GIS method. Results The populations of A. brasiliensis from the estuaries of the north‐eastern Brazilian coast (narrower and shallower continental shelf) showed higher levels of genetic differentiation and larger (more haplotypes) endemic lineages than the populations in the southern coast (broader and deeper continental shelf). Furthermore, south coast lineages showed more evidence of genetic exchange among populations, which have resulted in a reticulated relationship between lineages and estuaries. The reconstruction of palaeochannels revealed more connections among southern than north‐eastern basins. Main conclusions Our results support the hypothesis that continental shelf extension and sea‐level changes have influenced the diversification of A. brasiliensis. By extension, they also support the hypothesis of diversification by taxon pulse, which makes predictions based on the opportunities for expansion during periods of low sea level and the width of the continental shelf. We suggest this as a model for the genetic diversification and distribution of estuarine species along the Brazilian coast and other regions of the world.

Leishmania (Leishmania) infantum is the zoonotic agent of visceral leishmaniasis (VL), a disease with a global distribution. The transmission scenario of VL has been undergoing changes worldwide, with the biologic cycle invading urbanized areas and dispersing the parasites into other previously free areas. The epidemiological cycle in Brazil has dispersed from the Northeast to other regions of the country. In this study, an integrative approach, including genotyping Brazilian strains of L . (L . ) infantum for 14 microsatellite markers and reviewing historical records of the disease, was used to assess dispersion routes throughout central-southern Brazil. Our results support three L . (L . ) infantum dispersion routes: A) dispersion from Bolivia to the states of Mato Grosso, Mato Grosso do Sul and São Paulo via the Bolivia-Brazil gas pipeline from 1998 to 2005; B) VL dispersion from Paraguay to the Brazilian side of the triple border (Foz do Iguaçu and Santa Terezinha de Itaipu) during after 2012; and C) emergence of a new L . (L . ) infantum cluster in western Santa Catarina State and its dispersion to southern Paraná State (municipality of Pato Branco), after 2013. Hypotheses regarding possible entries of Leishmania (L . ) infantum into the area of the triple border are presented and discussed. Understanding how VL has dispersed is vital to the development of control measures for this disease and to avoid future dispersion events.

Variation in a fragment of the mitochondrial control region was investigated to assess the population history of the mangrove land crab, Ucides cordatus L., based on 100 individuals from five locations throughout the Brazilian coast. There was considerable variability in the studied fragment, with every individual showing a distinct haplotype. When regions of ambiguous alignment were removed, 90 haplotypes were still recognized, of which only 1 was shared among locations. There was no statistically significant evidence for geographical structure in the pattern of genetic variability based on a nested clade analysis, even though the geographical distances among the farthest collection sites exceeded 3000 km. Moreover, the mismatch distribution of pairwise differences showed a smooth unimodal distribution that is consistent with a recent population expansion of U. cordatus, either when each location was analysed in isolation or when all samples were combined. Interestingly, the estimates for the duration of such expansion vary latitudinally, suggesting that the expansion of the population of U. cordatus proceeded in a southward direction. These results indicate a possible relationship between the growth in the population of the mangrove land crab and the dynamics of mangrove ecosystems during the Quaternary.

This study surveyed the mitochondrial haplotype diversity of nine freshwater fish species and two estuarine-marine species from the coastal basins and drainages of the highland plateaus of Paraná, Brazil. Portions of the cytochrome b gene or the control region were sequenced. The demographic history of each species was inferred using the Bayesian skyline method, mismatch distribution analysis and statistical neutrality tests. Demographic reconstruction analyses revealed a single pattern of variation in the effective population size (N ) among species. No dramatic changes in N were detected in upland species. By contrast, evidence of population expansion over the past 200 000 years was detected in all coastal plain and estuarine species. These findings correspond to periods of low sea-level (regressions) followed by a rapid increase in the sea-level by >100 m. The resulting reconnections and subsequent fragmentation and isolation between the estuarine and freshwater bodies were putatively relevant to the historical demography of the fish species in these areas.

A cross-sectional epidemiological study in the extreme-west of the state of Paraná was carried out to access the prevalence, distribution, and risk variables of canine Visceral Leishmaniases (cVL). This study was conducted in three areas, two cities of far west of Parana state: Foz do Iguaçu (FI) and Santa Terezinha de Itaipu (STI), and along two transects between these two municipalities. To sample the entire urban area, the cities (FI and STI) were divided into a grid of squares of 400 m2 (patch). Among the 526 patches, 123 in FI, 40 in the transects and 33 in STI were selected according to the ‘worst scenario’ criterion. In the transect areas, in each 0.86 km five dogs from houses were surveyed to leishmaniasis. In each patch, blood of five dogs from houses (and from neighborhood when necessary) in the areas that seemed to be the most appropriate for the proliferation of vector were surveyed. The infection of the dogs by cVL were assessed using two serological tests were used (cELISA and TR-DPP®), and, for those seropositive for both methods, the PCR method were used. Moreover, dogs presenting clinical signs or cutaneous lesions were sampled to PCR. The identification of Leishmania species was confirmed using PCR-RFLP followed by DNA sequencing. Micro, meso and macro scale environmental variables were also surveyed and statistically analyzed. The prevalence rate Leishmania infantum was 23.8% in FI, 4.7% in STI and 9.1% in the transects areas. Among the extrinsic variables analysed, the number of vectors and the presence of infected dogs in neighbouring were positively correlated with the occurrence of infected dogs. Dog size was positively correlated with cVL infection, while the quality of the dog’s nutrition affected cVL negatively. As for cutaneous leishmaniasis (CL), the first registry of dogs infected with L. braziliensis in the region shows that there is potential for transmission in peri-urban areas, since environmental conditions allow the proliferation of vectors capable of transmitting this species of parasite. cVL is widely spread in FI, with high prevalence. This supports the hypothesis that the parasite has been present in the region for longer than previously believed, despite the fact that the presence of leishmaniais in the region has only been recognized recently. It is important to control the population of dogs infected with L. infantum (parasite and non-antibodies) to prevent the spread of the disease to other dogs and also to people in the region.

The Iguaçu National Park (INP) is the largest remnant of Atlantic Forest in southern Brazil, representing an ecological continuum with Argentina. The INP harbours a diverse fauna, with ring-tailed coatis (Nasua nasua Linnaeus, 1976, Carnivora: Procyonidae) in close contact with tourists either begging and/or snatching food from visitors. A potentially novel haemotropic Mycoplasma sp. has been previously detected in the ring-tailed coatis from central-western and southern Brazil. Therefore, the aims of this study were to investigate the occurrence of haemotropic Mycoplasma sp. and tickborne pathogens in wild ring-tailed coatis from the INP, Foz do Iguaçu municipality, Paraná State, southern Brazil. Blood samples were collected from 18 wild ring-tailed coatis and evaluated by conventional PCR (cPCR) assays for haemotropic Mycoplasma spp. (16S and 23S rRNA), Theileria/Babesia spp. (18S rRNA) and Ehrlichia/Anaplasma spp. (16S rRNA, sodB, dsb and groEL). Eight out of 18 (44.44%; 95% CI: 24.56%-66.28%) animals were positive for haemotropic Mycoplasma spp. All ring-tailed coatis tested negative for Theileria/Babesia spp. and only one out of 18 (5.56%; 95% CI: 0.99%-25.76%) animals tested positive for Ehrlichia/Anaplasma spp. by the 16S rRNA cPCR. Unfortunately, multiple attempts to sequence the 16S rRNA gene of the Ehrlichia/Anaplasma-positive sample have failed. Phylogenetic and network analysis of the hemoplasma 16S and 23S rRNA gene fragments confirmed that animals were infected by a potentially novel haemotropic Mycoplasma sp. previously reported in ringtailed coatis from Brazil. The name 'Candidatus Mycoplasma haematonasua' is proposed for this novel organism.
