Transformation of the above N-CBO-tripeptide ester to the N-acetyltripeptide ester and condensation by the hydrazide-azide procedure with the above dipeptide gives N-acetyl-L-seryl-L-tyrosyl-L-seryl-L-met~onyl-~-benzyl-L-glutamate. N‐Acetyl‐L‐seryl‐L‐tyrosyl‐L‐seryl‐L‐methionyl‐γ‐O‐benzyl‐L‐glutamyl‐L‐histidyl‐L‐phenylalanyl‐L‐arginyl‐L‐tryptophanyl‐glycyl‐ε‐N‐CBO‐L‐lysyl‐L‐prolyl‐L‐Valyl‐amide has been obtained by condensing N‐acetyl‐L‐seryl‐L‐tyrosyl‐L‐seryl‐L‐methionyl‐γ‐benzyl‐L‐glutamate with L‐histidyl‐L‐phenylalanyl‐L‐arginyl‐L‐tryptophanyl‐glycyl‐ε‐N‐CBO‐L‐lysyl‐L‐prolyl‐L‐valylamide. After splitting the N‐CBO and the O‐benzyl protecting groups by hydrogen bromide in trifluoroacetic acid, a free acetyl‐tridecapeptide‐amide has been obtained, which, after purification by counter‐ current and paper electrophoresis, has been found by several methods to be homogeneous and to possess the physical, chemical and biological properties of the natural hog α‐MSH.
