β-Coronaviruses are a family of positive-strand enveloped RNA viruses that include the severe acute respiratory syndrome-CoV2 (SARS-CoV2). Much is known regarding their cellular entry and replication pathways, but their mode of egress remains uncertain. Using imaging methodologies and virus-specific reporters, we demonstrate that β-Coronaviruses utilize lysosomal trafficking for egress, rather than the biosynthetic secretory pathway more commonly used by other enveloped viruses. This unconventional egress is regulated by the Arf-like small GTPase Arl8b and can be blocked by the Rab7 GTPase competitive inhibitor CID1067700. Such non-lytic release of β-Coronavirus results in lysosome deacidification, inactivation of lysosomal degradation enzymes and disruption of antigen presentation pathways. The β−coronavirus-induced exploitation of lysosomal organelles for egress provides insights into the cellular and immunological abnormalities observed in patients and suggests new therapeutic modalities.
X-ray radiation is widely used in medical and industrial applications. The basic design of the x-ray tube has not changed significantly in the last century. In this paper, we demonstrate that medical diagnostic x-ray radiation can be generated using a carbon nanotube (CNT) based field-emission cathode. The device can readily produce both continuous and pulsed x-ray with a programmable waveform and repetition rate. A total emission current of 28 mA was obtained from a 0.2 cm 2 area CNT cathode. The x-ray intensity is sufficient to image human extremity at 14 kVp and 180 mAs. Pulsed x-ray with a repetition rate greater than 100 kHz was readily achieved by programming the gate voltage. The CNT-based cold-cathode x-ray technology can potentially lead to portable and miniature x-ray sources for industrial and medical applications.
We report a field emission x-ray source that can generate a scanning x-ray beam to image an object from multiple projection angles without mechanical motion. The key component of the device is a gated carbon nanotube field emission cathode with an array of electron emitting pixels that are individually addressable via a metal-oxide-semiconductor field effect transistor-based electronic circuit. The characteristics of this x-ray source are measured and its imaging capability is demonstrated. The device can potentially lead to a fast data acquisition rate for laminography and tomosynthesis with a simplified experimental setup.
Carbon nanotube tips (see Figure) with controllable length, diameter, and orientation have been fabricated by the dielectrophoresis process. They are capable of delivering high and stable electron‐field‐emission currents of 3–5 μA (105 A cm–2). The emitters can potentially be used as the field‐emission electron sources for precision vacuum electronic instruments such as electron microscopes.
Objective
To assess the case fatality risk (CFR) of COVID-19 in mainland China, stratified by region and clinical category, and estimate key time-to-event intervals.
Methods
We collected individual information and aggregated data on COVID-19 cases from publicly available official sources from December 29, 2019 to April 17, 2020. We accounted for right-censoring to estimate the CFR and explored the risk factors for mortality. We fitted Weibull, gamma, and lognormal distributions to time-to-event data using maximum-likelihood estimation.
Results
We analyzed 82,719 laboratory-confirmed cases reported in mainland China, including 4,632 deaths, and 77,029 discharges. The estimated CFR was 5.65% (95%CI: 5.50%-5.81%) nationally, with highest estimate in Wuhan (7.71%), and lowest in provinces outside Hubei (0.86%). The fatality risk among critical patients was 3.6 times that of all patients, and 0.8-10.3 fold higher than that of mild-to-severe patients. Older age (OR 1.14 per year; 95%CI: 1.11-1.16), and being male (OR 1.83; 95%CI: 1.10-3.04) were risk factors for mortality. The time from symptom onset to first healthcare consultation, time from symptom onset to laboratory confirmation, and time from symptom onset to hospitalization were consistently longer for deceased patients than for those who recovered.
Conclusions
Our CFR estimates based on laboratory-confirmed cases ascertained in mainland China suggest that COVID-19 is more severe than the 2009 H1N1 influenza pandemic in hospitalized patients, particularly in Wuhan. Our study provides a comprehensive picture of the severity of the first wave of the pandemic in China. Our estimates can help inform models and the global response to COVID-19.
Uniform carbon nanotube films (see Figure) can be readily formed by electrophoretic deposition, as is presented in this communication. By varying deposition current and time, films with thicknesses in the range between several tens of nanometers and a few micrometers can be fabricated. The macroscopic nanotube films also show excellent electron field emission characteristics.
