The Asia Oceania Human Proteome Organisation has embarked on a Membrane Proteomics Initiative with goals of systematic comparison of strategies for analysis of membrane proteomes and discovery of membrane proteins. This multi-laboratory project is based on analysis of a subcellular fraction from mouse liver that contains endoplasmic reticulum and other organelles. Here we present the strategy used for preparation and initial characterisation of the membrane sample, including validation that the carbonate-washing step enriches for integral and lipid-anchored membrane proteins. Analysis of seventeen independent datasets from five types of proteomic workflows is in progress.
Blood and urine were collected for determination of biochemical parameters. Histological changes in the aorta and kidney were examined. At the end of the experiment, blood glucose, HbA1c and microalbumin showed at higher level. All of the high fat diet plus STZ injection rats exhibit remarkable lesions and plaque in their aorta representing damage to large blood vessels. However, they demonstrated features of different glomerulopathy. Rats suffered from hyperglycemia in whole studying time exhibit global glomerulosclerosis, hyaline arteriosclerosis and glomerular nodule which are comparable with characteristics observed in later stage of human nephropathy. The rest diabetic rats demonstrated features of mild nephropathy corresponding with a former stage of kidney disease.
BackgroundWilson disease (OMIM # 277900) is a autosomal recessive disorder characterized by accumulation of copper in liver and brain. The accumulation of copper resulting in oxidative stress and eventually cell death. The disease has an onset in a childhood and result in a significant neurological impairment or require lifelong treatment. Another serious consequence of the disease is the development of liver damage and acute liver failure leading to liver transplant. The disorder is caused by mutations in the ATP7B gene, encoding a P-type copper transporting ATPase.Case presentationWe performed genetic analysis of three unrelated patients from three different Vietnamese families. These patients had clinical features such as numbness of hands and feet, vomiting, insomnia, palsy, liver failure and Kayser–Fleischer (K–F) rings and were diagnosed with Wilson disease in the Human Genetics Department, Vietnam National Children’s Hospital. The entire coding region and adjacent splice sites of ATP7B gene were amplified and sequenced by Sanger method. Sequencing data were analyzed and compared with the ATP7B gene sequence published in Ensembl (ENSG00000123191) by using BioEdit software to detect mutations.ConclusionsIn this study, five mutations in the ATP7B gene were found. Among of these, three mutations were novel: c.750_751insG (p.His251Alafs*19) in exon 2, c.2604delC (p.Pro868Profs*5) in exon 11, and c.3077 T > A (p.Phe1026Tyr) in exon 14. Serum and membrane proteins are two of the most attractive targets for proteomic analysis. Previous membrane protein studies tend to focus on tissue sample, while membrane protein studies in serum are still limited. In this study, an analysis of membrane proteins in normal human serum was carried out. Nano-liquid chromatography-electrospray ionization mass spectrometry (NanoLC-ESI-MS/MS) and bioinformatics tools were used to identify membrane proteins. Two hundred and seventeen membrane proteins were detected in the human serum, of which 129 membrane proteins have at least one transmembrane domain (TMD). Further characterizations of identified membrane proteins including their subcellular distributions, molecular weights, post translational modifications, transmembrane domains and average of hydrophobicity, were also implemented. Our results showed the potential of membrane proteins in serum for diagnosis and treatment of diseases.
Tự kỷ là một hội chứng rối loạn phát triển của hệ thần kinh. Bệnh được biểu hiện bằng những khiếm khuyết về tương tác xã hội, khó khăn về giao tiếp và các hành vi sở thích hạn chế, lặp đi lặp lại. Tỷ lệ mắc bệnh ở trẻ nam nhiều hơn trẻ nữ và có xu hướng ngày càng tăng nhanh trên thế giới. Hiện nay chưa có phương pháp chữa trị dứt điểm cho các triệu chứng của bệnh tự kỷ. Các nghiên cứu trên thế giới cho thấy rằng tự kỷ là một trong bệnh có yếu tố di truyền chiếm từ 40-80%, và do nhiều gen liên quan. Nguy cơ di truyền của bệnh có liên quan đến ảnh hưởng kết hợp của các biến thể khác nhau. Giải trình tự vùng mã hóa - Whole exome sequencing (WES) đã xác định hàng chục nghìn biến thể gen trong mỗi exome ở nhiều bệnh đa gen như: tim mạch, thần kinh Vì thế, WES đang được coi là hướng đi đúng đắn để nghiên cứu di truyền bệnh tự kỷ. Bằng cách ứng dụng các phần mềm tin sinh học chuyên sâu như BWA (Burrows-Wheeler Alignment Tool); Picard; GATK (Genome Analysis Tool Kit), SnpEff, SnpSift, PolyPhen-2, nghiên cứu này đưa ra một quy trình cơ bản nhất để xác định các biến thể di truyền ở người bệnh tự kỷ. Đây là nghiên cứu đầu tiên sử dụng phương pháp WES để phân tích mối liên quan di truyền với bệnh nhân tử kỷ ở Việt Nam. Kết quả của nghiên cứu này làm cơ sở để định hướng cách thức phân tích số liệu WES.
In this work, we propose a design in the proof-of-concept of a 1×3 two-mode selective silicon-photonics router/switch. The proposed device composes of a Y-junction coupler, two multimode interference (MMI) couplers, and two phase-shifters on the silicon-on-insulator (SOI) rib waveguides. The input modes of TE 0 and TE 1 can be arbitrarily and simultaneously routed to the yearning output ports by setting appropriate values (ON/OFF) for two tunable phase shifters (PSs). The structural optimization and efficient characterization processes are carried out by numerical simulation via three-dimensional beam propagation method. The proposed device exhibits the operation ability over the C-band with good optical performances in terms of insertion loss smaller than 1 dB, crosstalk under -19 dB, and relatively large geometry tolerances. Moreover, the proposed device can integrate into a footprint as compact as 5 µm × 475 µm. Such significant advantages are beneficial and promising potentials for very large-scale photonic integrated circuits, high-speed optical interconnects, and short-haul few-mode fiber communication systems.
TÓM TẮT: Hội chứng mạch vành cấp (Acute Coronary Syndrome, ACS) hiện đang là vấn đề sức khỏe rất được quan tâm trên thế giới do bệnh thường uy hiếp tính mạng con người một cách đột ngột, tỷ lệ tử vong và di chứng do bệnh hội chứng mạch vành cấp vẫn chiếm hàng đầu và đang có xu hướng gia tăng nhanh chóng ở nhiều nước phát triển. Vì vậy, nghiên cứu tìm kiếm các biomarker để chuẩn đoán sớm bệnh hội chứng mạch vành cấp là một yêu cầu mang tính cấp thiết. Glycosyl hóa là một biến đổi sau dịch mã phổ biến, đóng vai trò quan trọng trong nhiều quá trình sinh lý như: đáp ứng miễn dịch, điều hòa chu trình tế bào, là nhân tố chỉ thị của môi trường ảnh hưởng lên các quá trình nội bào và liên quan đến nhiều con đường chuyển dạng từ tế bào bình thường thành tế bào ung thư. Những nghiên cứu gần đây đã chỉ ra rằng, những protein bị glycosyl hóa thường liên quan đến các quá trình bệnh lý khác nhau như: tiểu đường, sơ nang, Alzheimer, viêm khớp, các bệnh tự miễn, bệnh tim, bệnh liên quan đến tress, ảnh hưởng đến chức năng thận và đặc biệt là ung thư. Hiện nay, bằng các kỹ thuật proteomics, đã có nhiều glycoprotein được xác định là chỉ thị phân tử đặc trưng cho nhiều bệnh lý trong đó có hội chứng mạch vành cấp. Trong nghiên cứu này, chúng tôi sử dụng kỹ thuật điện di hai chiều kết hợp với sắc ký lỏng nano một chiều kết nối khối phổ liên tiếp (1D nanoLC-ESI MS/MS) để phân tích thành phần glycoprotein trong huyết thanh mẫu người bình thường và các bệnh nhân đau thắt ngực không ổn định, nhồi máu cơ tim cấp. Kết quả cho thấy, 3 glycoprotein có mức độ biểu hiện tăng (ceruloplasmin, haptoglobin và fibrinogen), 1 protein có mức độ biểu hiện giảm (Alpha-2-HS-glycoprotein) ở mẫu bệnh so với mẫu đối chứng.Từ khóa: Đau thắt ngực không ổn định, glycoprotein, hội chứng mạch vành cấp, huyết thanh, khối phổ, nhồi máu cơ tim cấp, proteomics. Thuật ngữ hội chứng mạch vành cấp được sử dụng để chỉ toàn bộ bệnh lý cấp tính của động mạch vành mà theo truyền thống được chia thành cơn đau thắt ngực (angina), cơn đau thắt ngực không ổn định (unstable angina) và nhồi máu cơ tim cấp (myocardial infarction).Nếu người bệnh được phát hiện trong giai đoạn cấp tính, quá trình điều trị bệnh sẽ hết sức khó khăn.
