Eine modifizierte FRIEDLANDER-Synthese wird beschrieben. Als Ausgangsprodukt dient das Hydrochlorid des leicht darstellbaren und stabilen o-Aminoacetophenons. Die Kondensation mit zahlreichen carbocyclischen Ketonen fuhn zu den entsprechenden 2.3-disubstituierten Lepidinen. Nach P. FRIED LANDER^) (1 882) entstehen aus o-Amino-benzaldehyd und Carbonylverbindungen, die in a-Stellung eine Methylengruppe besitzen, substituierte Chinoline. Die Kondensation erfolgt i m alkalischen Milieu und liefert rneist niedrige Ausbeuten. Die allgemeine Anwendbarkeit dieser Synthese wird aul3erdem durch die schlechte Zuganglichkeit des o-Amino-benzaldehyds sowie seine Instabiliut eingeschrankt3.4). Wir fanden im o-Amino-acetophenon einen Kondensationspartner, der diese Nachteile nicht zeigt. o-Amino-acetophenon kann aus o-Nitro-acetophenon~Xi7) durch Reduktion rnit Sn/HCI 8) bzw. katalytisch erregtem Wasserstoffg. 10) sowie aus 2.3-Dimethyl-1 -acetyl-indol durch Chromsiiure-Oxydation 11) erhalten werden. Es ist als freie Base bzw. als Hydrochlorid praktisch unbegrenzt haltbar. benutzten wir eine neue Methode.
